Hánlová, P: Effect of chromium on semen quality and carbohydrate metabolism in bre-eding boars, Diploma Thesis MENDELU, Brno, 2015, s. 59 The aim of this thesis was to develop a literature search and experimentally de-termine the influence of mineral nutrition on boar semen quality. The thesis is divided into two parts. The first assesses the impact of the addition of chromium on semen qua-lity of breeding boars. The second part examines the influence of the addition of chro-mium on glucose and insulin levels in the blood of breeding boars. The experiment was conducted in Velké Meziříčí at the insemination center boars. The experiment consisted of 28 Duroc boars breed for 14 weeks. These boars were divided into 4 groups according to age balanced. In the control (n = 7) group (K), was chromium receiving 205 ug/kg/day. In the 1st group (n = 7) animals (CR1) was dose chromium even increased by 60.6 ug/kg compound feed for the second group (n = 7) boars (Cr2) of 121.2 ug/kg and 3. group (n = 7) boars (Cr3) was added chromi-um in an amount of 181.8 ug/kg compound feed. The addition of chromium to the semen quality of breeding boars too had a signifi-cant impact. During the first months of feeding chromium occurred in the control group to a decrease in ejaculate volume of 29 ml. In the group S3 during the experiment is bound to increase sperm concentration, but in the fourth taking under review decreased significantly compared to previous months. The difference between the third and fourth taking was 131.5 thousand/mm3. This difference is statistically insignificant, and could be caused by external influences or errors in nutrition. Sperm motility during certain months was below 70%. This ejaculate should not be used for insemination. In the second part of the experiment, the addition of the chromium significantly reduced glukose levels and insulin activity. The obtained results can not recommend feeding chromium breeding boars.

If gravid women suffer from diabetes,their unborn children have 10x higher risk of development of malformation,prenatal and postnatal death and post partum complications than children of women belonging to healthy population.The main and very controversial potential teratogenic factors are glucose and insulin.However,there are very ambivalent opinions on which one of these two substances causes damage to embryo. Therefore,the aim of this diploma thesis was to contribute to solving of this problem and test direct embryotoxicity of insulin and glucose. For solving of this issue, the so-called CHEST.The principle of this method is creation of a window in eggshell and consequent subgerminal or intraamnial application of the substance being tested.Embryos were tested from the second until the sixth incubation day.Firstly, two types of insulin were injected.Injected doses varied between 3µg/3µl to 0,003µg/3µl. Then glucose was tested,with dosage of 300µg/3µl and 30µg/3µl. Embryotoxic effect was detected for both types of insulin. The beginning of embryotoxicity line of insulin's lies between the dosage of 0,03µg/3µl and 0,003µg/3µl. Abstract The diploma thesis is focused on the application of insulin in nursing practice. In 2013, there were 678,935 patients with diabetes mellitus in the Czech Republic. Of this, 17.5 % of patients were treated with insulin. When applying insulin, we must observe certain principles and procedures to avoid complications. The diploma thesis consists of the theoretical and research part. The theoretical part describes the characteristics of diabetes mellitus, describes insulin and its history, regimens, types and storage of insulin. It is also focused on the insulin application process and the education of patients. In relation to the topic, two objectives were set. The first objective was to find out how to apply insulin in nursing practice based on evidence. The second objective was to find out how the nursing process is provided by a nurse when applying insulin in nursing practice. Following these objectives, two research questions have been identified. First research question: How is insulin applied in nursing practice? The second research question: How is the nursing process filled with the insulin application? The practical part was elaborated using a qualitative research survey. The research was carried out using two variants of a semi-structured interview. The first variation of the interview was for insulin-like patients. The second variant was intended for general nurses from standard departments of selected hospitals and for nurses working in diabetological outpatient clinics. The first set consisted of ten respondents, of which 5 were women and 5 men. The second group consisted of 9 respondents, of whom all were women. The conversations were recorded and then literally overwritten. Interviews were overwritten using the pencil-paper method and further analyzed by Open Encoding. Based on data analysis, 5 categories were created with several subcategories for patients and 6 categories with several subcategories for nurses. Based on the analysis of the data, it can be said that patients or nurses do not apply insulin completely correctly. I perceive the basic problem in the distribution of insulin by species and its correct application, which varies for each species. I see another problem in incorrect application according to the length of the needle, such as the angle of the needle and the formation of the skin kelp. The output of the diploma thesis will be the standard of nursing care based on EBP, in order to highlight the shortcomings we found in the research investigation and to prevent their prevention. This standard aims to refine and improve the care provided.

Obesity leads to many severe metabolic disorders, e.g. dyslipidemia, insulin resistance, ectopic fat accumulation in the liver and skeletal muscles, non-alcoholic fatty liver disease and finally diabetes mellitus type 2. Metformin (1,1-dimethylbiguanide) is the most favored medicament for the treatment and prevention of these disorders. It stimulates cellular glucose uptake and normalizes blood levels of lipid metabolites without triggering insulin secretion. Research on insulin resistance and diabetes is often realized through developing diet- induced obesity in laboratory animals. The aim of this project is to compare metabolic effects of two different high-fat diets named HFD and HSD. The HFD diet consists chiefly of n-6 polyunsaturated fatty acids (corn oil) and starch (100% glucose). The HSD diet contains mainly saturated fatty acids (lard) and sucrose (50% glucose and 50% fructose). I also studied metabolic effects of metformin by adding it continuously to the drinking water given to obese mice fed with the HFD or the HSD diet. The thesis deals with the measurement of glucose levels at regular intervals in a selected group of volunteers after the consumption of breakfast cereals in combination with milk products. The theoretical part is devoted to the basic components of the diet and their digestion and metabolism. It also deals with the regulation of blood glucose level, methods for measuring glycemia, disorders related to metabolism of carbohydrates and the concepts of the glycemic index, glycemic load. The aim of the practical part was to find out how your chosen breakfast cereal in combination with dairy products have an effect on blood glucose levels in a selected group of volunteers. 16 students of the Mendel university in Brno were attended the experimental measurements. The students measured on the InBody, in order to determine composition of the human body. The level of blood glucose was always measured in time 0 (fasting), 60, 90 and 120 minutes from the beginning of the consumption of the food. Statistically significant difference was shown from measured values of blood glucose in 60. and 90. minute between the glucose solution, and breakfast cereals (P < 0.05).

Ph.D. Thesis abstract: Diabetes mellitus is a chronic disease characterized by a metabolic disorder in which there is a low level or complete lack of the insulin. Diabetes mellitus type 1 (DM1) is caused by an autoimmune reaction leading to the destruction of the insulin producing beta cells in the pancreas. In consequence, low or non-existent insulin production leads to a complete dependence on exogenous insulin supplementation. DM1 causes serious long-term complications. Although strict control of blood sugar could prevent the onset and development of diabetic complications only 5% of diabetic patients are able to achieve such control. Hence it is evident that the current methods of treatment are neither sufficient to treat this disease, nor prevent late complications in most patients. The most promising therapeutic approach in the treatment of diabetes is the restoring of insulin production. One such method is the transplantation of insulin-producing tissue. However, a lack of available insulin- producing tissue limits such therapeutic approach. Therefore an alternative source of insulin producing cells have to be found to obtain a sufficient amount of safe and efficient insulin producing tissue.
