In my diploma thesis I deal with breathing when playing the brass instruments. In the first chapter called Process of Breathing I describe the respiratory system, its breathing mechanism and a separate breathing process with the main emphasis on the types of breathing, control of breathing and a lung capacity. Subsequently I occupy with the breathing process when playing, it means with breathing in cooperation with creation of tones, embouchure, activity of tongue and with the finger technique. In the second chapter I mention some important brass players and teachers of the breathing in brass instrumental music; the view of the East Cultures on ordinary breathing is also supplemented to this overview. In the third chapter I quote some methods of breathing development. Firstly I focus on the evolution and usage of various types of breathing, secondly on the methods of breathing development. This chapter includes many breathing exercises done with or without an instrument, which should lead to liberty and speed of inhalation and smoothness and holding of exhalation. The exercises help to the complete fastening of the muscles of respiration.

This thesis focuses on the Violin Concerto composed by L. v Beethoven, its interpretation and the changes in attitude towards the composition within the two centuries of its existence. The paper is divided in four chapters. First of all it provides information based on primary sources about the development and changes in the violin technique and interpretation. After that an insight into the fate of the concerto after its premiere occurs, followed by an account of its performers. The following chapters compare first the different editions of the concerto and later on its numerous recordings. A live recording of the concerto performed by the author accompanied by Musica Florea with the conducter Marek Štryncl makes part the thesis.

My diploma thesis is called Main repertoire for the trio clarinet, cello and piano. The work is devided to three chapters. The first one is called History and present of the trio clarinet, cello and piano. In this chapter are three independent subchapters. The first is about historical development of these instruments. In the seconde is written something about repertoire options for this group and the third one is about tree well-known groups.
The seconde chapter is about the main tema and there is written about the most played and well-known compositions for this ensemble. There is usually written something short about life of the composer and then formal analysis of every piece. The main compositions are written in this order: Archduke of Austria and The Trio for clarinet, cello and piano, Johannes Brahms and The trio a minor op. 114, Max Bruch and Eight pieces op. 83, Vincent d´Indy and The trio B flat major op. 29, Anton Eberl and Grand trio op. 36, Michail Ivanovič Glinka and The trio Pathétique, Emil Hartmann and Serenada op. 24, Adalbert Gyrowetz and The Trio op. 38, Robert Kahn and Serenada op. 73 a Alexandr Zemlinsky and The trio d minor op. 3.
The last chapter is only about The trio B flat major of Ludwig van Beethoven. In three short subchapters is written something about his life, formal analysis and creation of the composition and analysis of interpretation of two records.

Praktické aspekty analýzy jednotlivých buněk pomocí RT-qPCR / Practical aspects of single-cell RT-qPCR analysisŽucha, Daniel January 2020 (has links)
Recent breakthroughs in the RNA quantification of single cells are rapidly transforming the view on biology and medicine. Flexibility and sensitivity of reverse transcription quantitative PCR (RT-qPCR) make it an ideal method for quantification of single-cell material, but its limits had not been yet fully explored. In this thesis, various factors influencing RT-qPCR performance in single-cell application have been assessed, including conditions of sample collection and processing, importance of quality control, performance of reverse transcription, preamplification and role of qPCR assays. We showed that prolonged time for single cell collection as well as repeated freeze-thaw cycles had negligible effect on RT-qPCR data quality. Direct lysis routinely applied for RNA extraction from single cells may be scaled up to 256 cells. The comprehensive comparison of 11 reverse transcriptases in low RNA input conditions identified 2 best-performing enzymes. Decrease in preamplification volume as well as poor primer design resulted in the loss of sensitivity. Finally, the established workflow has been applied to profile gene expression of astrocytes in mouse model of amyotrophic lateral sclerosis (ALS) identifying important components of ALS-induced changes to astrocyte transcriptome.
