Separation and determination of bioactive compounds in food

Herbs are widely known for their numerous applications in the food industry; however, there is increasing interest in their pharmaceutical properties. In recent years, major research has been focused on the biologically active derivatives of medicinal plants, mainly because of their potential anti-tumor activity and protective effects with respect to the cardiovascular system. This thesis summarizes the biosynthesis of phenolic compounds mainly responsible for such positive health impacts, the methods of antioxidant capacity determination, and the protective health effects of medicinal herbs. The experimental part is dedicated to extraction methods and the determination of phenolic compounds from herbs, as well as the possibility of applying herbal infusions into raw goat whey in order to extend its shelf-life.

Diploma thesis "Methods of honey quality assessment" was made at the Department of Post-Harvest Technology of Horticultural Products of Horticultural Faculty at Mendel University in Brno in the years of 2013 -- 2015. Theoretical part of the thesis includes todays legal requirement for honey quality and description of the analytical possibilities of honey qualitative parameters assessment. Experimental part describes 35 honey samples in which the determination of volumetric acidity at 23°C was analyzed. Furthermore, the assessment of antioxidant capacity was made by two methods - DPPH and FRAP. The samples were measured at 23°C and then at 95°C for 5 minutes in both methods. Comparison of antioxidant capacity changes owing to warming was made in both methods DPPH and FRAP. It was found, that the antioxidant capacity after warming increased in DPPH method. In FRAP method, the number of samples, in which decrease or increase occured was comparable.

Antioxidační kapacita a fenolické látky v révovém víně

This thesis is focused on monitoring the health beneficial substances with phenolic character and antioxidant activity sampled from leaves, lateral shoots, stems, berries, juice and wine. There were 4 interspecific varieties selected for the monitoring. These varieties are characterized by an increased resistance to fungal diseases. The monitoring was carried out for 3 consecutive years. Samples were taken from healthy and infected plants in three sampling periods, so it was possible to assess the effect of maturation on the antioxidant activity and the content of phenolic compounds. Samples were analyzed on EPR (antioxidant activity determination), spectrophotometer (antioxidant activity, total phenolics) and assayed by HPLC (determination of selected phenolic compounds, especially trans-resveratrol). The results show that all samples exhibit antioxidant activity and contain a wide range of phenolic compounds. All the samples were analyzed for trans-resveratrol, which is known for its beneficial health effects. The content of resveratrol ranged from 9.07 to 566.14 mg.kg-1 of lyophilized material. The highest content was found in the stems. Antioxidant activity determined by spectrophotometry ranged from 308.0 to 1357.7 mg GA.kg-1 in berries, and from 44.0 to 839.5 mg GA.l-1 in wines. Decreases of free radicals representing the antioxidant activity, which were measured by EPR, ranged from 1.21 to 57.17 % in the leaves. Blue varieties showed higher antioxidant activity. There was a positive correlation shown between the content of trans-resveratrol in the stems in interspecific and European varieties. It also demonstrated a dependence of the antioxidant activity on the content of phenolic compounds in the samples.

The thesis deals with production of biomass of spring barley (Hordeum vulgare, L.) depending on the parameters of a root system. The theoretical part is focused on the methods of assessment of root system of plants, the influence of environment and genotype on the qualities of a root system of plants and on the relation between the parameters of a root system and the yield of field crops. Another part of the thesis is a statistical assessment of the results of a field experiment with spring barley, which was realized in the locality Hořejší Kunčice in 2014 and in the localities Hrubčice and Želešice in 2012 and 2013. By means of the method of measuring electrical capacity, the size of the root system of four varieties of spring barley and their hybrids was assessed in three development stages: BBCH 36, BBCH 55 and BBCH 71. The yield of grain and the yield of biomass were assessed in the stage BBCH 89. An important influence of the size of the root system on the yield of grain in stage BBCH 55 (r = 0,643*) was discovered in 2013 in Hrubčice. In Želešice, 2013, an important influence of the size of the root system on the yield of grain was found in all the examined development stages, BBCH 36 (r = 0,650*), BBCH 55 (0,824*) and in the stage BBCH 71 the influence was highly important (r = 0,839**).

The Diploma thesis deals with the topic enrichment of beverages by additional antioxidants and has been processed at the Faculty of Horticulture, Mendel University in the Institute of Post-Harvest Technology of Horticultural Products. The thesis is divided into theoretical and practical part. In the theoretical part there are described antioxidants, free radicals, antioxidant capacity, means of antioxidant measurement in food, beverages containing antioxidants like fruit juice, tea, wine, and beer. There is also described chemical structure of food. Practical part deals with measurement of antioxidant capacity by DPPH and FRAP method and volume of polyphenolic substance in distillery samples with extracted oak wood dried by several temperatures and comparison of these samples with the commercial sample, subsequently sensory analysis of these mentioned samples. From results it´s obvious that distilleries can be enriched by additional wood especially by wood dried by 175 °C. Sensory analysis has proved that enrichment of the distillery by oak chips can significantly improve it´s quality.
