The author in his doctoral thesis, proposed primers and conditions optimized design Quantitative PCR for determining the gene expression of MMP-7, TIMP-1, MMP-2 and TIMP- second Revealed the presence of gene expression of GAPDH TIMP-1 and TIMP-2 in cancer line HT-29 SW480 and SW620. Noted the high level expression of MMP-7 in line HT-29th Expression of the 2-3 orders of magnitude higher than the lines SW480 and SW620. MRNA expression of MMP-2 in line HT-29 noticed. MRNA expression of MMP-2 by the detected lines SW480 and SW620. He found that determining absolute and relative expression in tumor lines is ekvivalentní. significantly higher mRNA expression of MMP-7, TIMP-1, MMP-2 and TIMP-2 in tumor tissue compared to normal tissue. It can be used in therapy. Noticed the presence of correlation of gene expression MMP-7, TIMP-1, MMP-2 and TIMP-2 with survival and DFI. The level of MMP-7, TIMP-1 and MMP-2 mRNA expression was significantly higher in the tumour tissue of colorectal carcinoma than in normal colorectal tissue. It could be possible to inhibit matrix metalloproteinases activity using appropriate antibodies, which could have therapeutic effect on tumour tissue and its vicinity. Some of the preparations are being tested (Bay 12-9566 /Bayer/, BB94 /British Biotechnology/). We have not proved correlation between expression of these genes and disease stage and diagnosis. We have succeeded to prove that if the surgical principles for colon resection performed due to colorectal carcinoma are observed, the resection line does not show any signs of the presence of tumour cells - mRNA for CEA is not present. The level of mRNA for TIMP-1 is present in the resection line at lower levels than in tumour tissue, and this is due to the role that TIMP- 1 plays in the colon. Its level increases not only in all tumour diseases, but also in inflammatory diseases of the colon. The question whether the expression of mRNA for TIMP-1 is also increased outside the resection line and therefore it is a reaction of the colon as an organ will be subject to further research, as will be a potential comparison with samples of colon unaffected by tumour or inflammation. Breast tumors are not only main incidence of tumor based load of women population, but they are still the most common cause of death in the area of tumor diseases. 1950 women dies on breast tumors every year, which is 37 out of 100 thousand women. Overall are breast tumors cause of deaths in 3.5% women in population. Despite still growing incidence of breast tumors, death rate caused by this type of tumor stagnate on long tearm basis, which clearly shows improvement in treatment success mostly thanks to higher finding in earlier stages. In our country is in 28 years evident shift of incidence into younger age groups, which starts to be obvious in age group of 40-44 years already. Today's prevention possibilities are on high level. It depend's only on patients themselves, if they will obey these rules. At early finding illness of breast carcinoma is prognosis of recovery high. Because of this disease topicality I've decided to process this topic and expand my knowledge.

Prostate carcinoma is one of the most frequent cancer diseases of maturated men. Diagnose of this cancer disease is based on level of prostatic specific antigen and digital exam per rectum. However, presence of tumor, grade and stage must be examined by biopsy. This method may not to be accurate. This is the reason why are new diagnostic methods investigated, to increase the accuracy of diagnostic prostate cancer. Amino acid sarcosine is currently on of the most widely discussed biomarker, that can serve as a diagnostic biomarker for early stage detection in prostate cancer. The present work deals with the study of sarcosine metabolism in prostate cancer cell lines, mainly, but also by amino acid metabolism in cells in general. Understanding to importance of single amino acids in malignant and non-malignant cell pathways can significantly contribute to effective targeted therapy without side effect on healthy tissue. Prostate cancer cells in presence of sarcosine increase their migration potential, malignancy by increased doubling time, and concentration of sarcosine N-demethylation enzymes, sarcosine dehydrogenase and sarcosine oxidase. Through this biochemical pathway is increased synthesis of main methylation donor S-adenosilmethyonine (SAM) ad cellular methylation potential.

Thesis on the topic "Search of New biomakers suitable for screening and early diagnostics of cancer" is dedicated to the issue of cancer, a cancer of the breast, prostate, ovarian and after the new findings out as melanom determination of tumor markers and their implications for clinical practice. Their aim is skouted up biomaker suitable for screening and early diagnostics of carcinoma( cancer ) of the breast, ovarian, prostate and malignant melanoma with utilization of various kinds of immunoassay. The theoretical part gives a brief and comprehensive overview of the epidemiology of selected tumors and tumor markers used and their qualities. In the methodological part are given principles applied iminoanalytics methods a list of the statistical methods and a detailed description of the groups of patients. Results section contains tables with results of biomarkers diveded by sort of avalueted tumors. In the discussion are then analyzed facts that follow from the results. It discussed the process and actualy status of searching convenients biomekers and their combinations that can distinguish a population of benign tumors of the population with malignant tumors and markers which are able to distinguish healthy population from the population with malignant tumors of the breast, ovary, prostate and melanoma. This work may serve as a source of knowledge and results to continue in search of such marker that could be used for screening and early diagnosis of cancer and so to improve the prevention of these illneses in the population.
