Vliv potravy na výši fertility u zákeřnic druhu \kur{Platymeris biguttatus} / Effect of food on fertility in assassin bug \kur{Platymeris biguttatus}

This thesis was developed in order to determine whether the selected species most commonly kept feeding insects affect fertility in captive bred assassin bugs \kur{Platymeris biguttatus}. Used feeding insects were \kur{Blaptica dubia} roaches, \kur{Acheta domestica} crickets and larvae of \kur{Tenebrio molitor} bugs. The reason of using assassin bugs \kur{Platymeris biguttatus} was their popularity among breeders, their easy care needs and their superior fertility. Assassin bugs were kept in an air conditioned room with a constant temperature conditions and light regime. It has been shown that the given diet has no effect on fertility in this kind of bugs, also that breeding in groups has negative impact on fertility of this assassin bugs species. The results of this study could demonstrate the importance of proper feeding in keeping these bugs and help breeders to have better results.

This work contains a review of botanical characteristics of the species used, the description of methods, the design, results and discussion of two preliminary experiments.

Eimeria is an apicomplexan parasite causing disease coccidiosis that is most prominent in poultry farming industry. This thesis is aimed to develop new molecular tools and resolve issues that would be a valuable contribution in the field from both research and industry perspective. Because immunity to Eimeria is strictly species- specific, it is important to know and recognize correctly all species that parasitize the host. Traditional diagnostic approaches rely on classical methods such as oocyst morphology determination under the microscope, measurement of prepatent period or in-vivo assessment of lesions caused by this parasite. However, diagnostics of individual species using these methods is very time-consuming and it is often unreliable, especially when mixture of multiple species whose parameters overlap is analyzed. Methods utilizing conventional PCR to distinguish species already exist, however, they lack advantages offered by quantitative real-time PCR (qPCR). The first aim of this thesis was to develop qPCR assays for detection and quantification of seven Eimeria species which infect chicken utilizing single-copy non-polymorphic targets in order to ensure maximal specifity and coverage of all strains of each species. The resultant racemic &amp / #945 / &#039 / -acetoxylated substrates were resolved into corresponding enantiomerically enriched &amp / #945 / &#039 / -hydroxylated and &amp / #945 / &#039 / -acetoxylated compounds via PLE hydrolysis. &amp / #945 / &#039 / -Hydroxylated compounds are racemized quickly, so they were acetylated with acetyl chloride and pyridine in situ to give the corresponding &amp / #945 / &#039 / -acetoxylated compounds. Resultant &amp / #945 / &#039 / -acetoxy &amp / #945 / ,&amp / #946 / -unsaturated cyclic ketones reacted with excess amount of diazomethane under the catalsts of Pd(OAc)2 to give the resulting bicyclic diastereomeric products. At the end of the experiment, Enantiomeically enriched 2-oxobicyclo[3.1.0]hexan-3-yl acetate and 2-oxobicyclo[4.1.0]heptan-3-yl acetate were chemoenzymatically synthesized.

International trade in exotic pets is an important and increasing driver of biodiversity loss. The African grey parrot has been noted as one of man's friendliest pets with popularity attributed to its intelligence, cognitive and communicative abilities. Worldwide, the African Grey Parrot is the third most commonly traded wild bird species. The subject of this thesis is an evaluation of international trade in the gray parrot (\kur{Psittacus erithacus}), covered by CITES convention to Appendix II, between European Union and third countries, according CITES Trade Database and EC Annual Report 2004 in 1994-2013. The trade has been divided into export and import, by the two existing subspecies, and within these categories were evaluated main countries of export and import, source and purpose of the trade. It was found, that the trend of import of live specimens to EU during the period 1994-2005 was increasing, although the trend between 2006-2013 was declining. The trend of export of live specimens was slightly increasing for the period 1994-2001 and 2006-2013.

Entomopathogenic fungus \kur{Metarhizium anisopliae} is one of the most common species used in biological control against pests. The thesis is analyzing effectiveness of original strains and continuously passaged strains of \kur{M. anisopliae} through nutrient substrates and different developmental stages mealworm \kur{(Tenebrio molitor)}. For original and continuously passaged strains were also evaluated the growth and spore production at different temperatures cultivation. In this thesis was investigated the efficacy of the original strains of \kur{M. anisopliae} on selected economically important pests. Strains were tested on populations of adults Pollen beetles \kur{(Meligethes aeneus)} and Cabbage seedpod weevil \kur{(Ceutorhynchus obstrictus)} and the eggs of Colorado potato beetle \kur{(Leptinotarsa decemlineata)} in laboratory conditions. Other entomopathogenic fungi were tested on the eggs of Colorado potato beetle eggs.
