Características qualitativas de oócitos obtidos por Laparoscopic Ovum Pick-up (LOPU) e da maturação nuclear em ovinos /

Orientador: Sony Dimas Bicudo / Banca: Eunice Oba / Resumo: O presente estudo visou avaliar a resposta ovariana a superestimulação utilizando 80UI de FSH e 300UI de eCG após ablação por laparoscopic ovum pic-up (LOPU1) de todos os folículos visíveis presentes em ambos os ovários. Determinar o melhor momento para realização de uma nova LOPU (LOPU2), mediante a avaliação ultrassonográfica dos ovários a cada 12 horas após LOPU1. E comparar a respostas ovariana do tratamento Duplo LOPU (DLOPU) com o tratamento One Shot adaptado de Baldassarre (1996), quanto a quantidades dos Complexos Cumulos-Oócito (COCs), e qualidade das estruturas obtidas e após processo de maturação in vitro (MIV) por 24 horas. Foi determinado o momento para realização da LOPU2 em 36 horas após a superestimulação. A média de folículos presentes na avaliação ultrassonográfica imediatamente antes a LOPU em ambos os tratamentos foi cerca de 9 estruturas, não diferindo da media de folículos aspirados, a taxa de recuperação comum aos dois tratamentos foi de 62,5%. A avaliação imediata das estruturas obtidas bem como a após 24 de MIV demonstrou que não houve diferença qualitativa COCs entre os tratamentos / Abstract: The present study aimed to evaluate the ovarian response to superstimulation using 80UI FSH and eCG 300UI after ablation LOPU (LOPU1) of all follicles present in both ovaries. Determine mlehor time to make a new LOPU (LOPU2) by ultrasound evaluation of the ovaries every 12 hours LOPU1. Qualidade fisiológica de sementes de quiabeiro em função da idade e do repouso pós-colheita dos frutos /

Orientador: Antonio Ismael Inácio Cardoso / Banca: Antonio Celso Wagner Zanin / Banca: Cibele Chalita Martins / Banca: Keigo Minami / Banca: Francisco Antonio Passos / Resumo: Este trabalho foi conduzido no Departamento de Produção Vegetal, setor Horticultura, da Faculdade de Ciências Agronômicas, da Universidade Estadual Paulista (UNESP), Botucatu-SP, com o objetivo de estudar o efeito da idade do fruto e do repouso pós-colheita do mesmo sobre a qualidade de sementes de quiabeiro cultivar Santa Cruz-47. No dia da antese as flores foram etiquetadas, sendo os frutos colhidos com 34, 41, 48 e 55 dias após a mesma (DAA). Metade dos frutos de cada idade permaneceram em repouso pós-colheita por quatorze dias antes de se extrair as sementes, enquanto que da outra metade extraiu-se as sementes no dia da colheita. O delineamento experimental foi em blocos ao acaso, com 5 repetições e 6 plantas por parcela. Para caracterização das sementes foi obtido o teor de água no momento da colheita e para verificar a sua qualidade os seguintes testes foram realizados: massa de 100 sementes, germinação, primeira contagem de germinação, índice de velocidade de germinação, emergência em bandeja e condutividade elétrica. Também foi feita a caracterização dos frutos e sementes. Foi realizada análise de variância em parcela sub-dividida e para as características em que houve diferença entre as idades, realizou-se análise de regressão. Pode-se concluir, que o repouso pós-colheita dos frutos melhorou a qualidade das sementes, principalmente das mais novas (34 e 41 DAA). Quanto mais novas as sementes sem repouso pós-colheita menor a qualidade. / Abstract: This work was carried out at FCA-UNESP, in Botucatu (SP), with the objective of studing the effect of fruit age and post harvest storage in the quality of okra seeds cv. Santa Cruz-47. The flowers were labeled at anthesis and the fruits were picked with 34, 41, 48 and 55 days after that event (DAA). For each age, in half of the fruits, the seeds were immediately extracted and in the other half they were stored fourteen days before the extraction of the seeds. The following tests were accomplished to verify the quality of the seeds: test germination pattern (TGP), first counting and index of germination speed (IGS), emergency, electrical conductivity and characterization of the fruits and seeds. Analysis of variance followed the split plot model. Ages were compared by regression analysis. Post harvest storage performances were compared by Tukey test. It can be concluded that the post harvest storage improved the quality of the seeds, mainly the new ones (34 and 41 DAA) and the newer the seeds without post harvest storage the lesser the quality. / Doutor

Qualidade de compostos orgânicos produzidos com resíduos do processamento de plantas medicinais /

Todavia, ainda são necessárias pesquisas mais aprofundadas para a melhoria da eficiência deste processo, o que levaria à produção de compostos de maior qualidade, assim como o fornecimento de nutrientes como condicionadores de solo. Sendo assim, este trabalho teve como objetivos avaliar a qualidade de compostos produzidos a partir de combinações de resíduos do processamento de plantas medicinais com esterco bovino e verificar o potencial agronômico dos materiais resultantes na produção da alface. Em uma primeira etapa foram utilizados quatro resíduos do processamento de plantas medicinais: 1) Unha-de-gato (Uncaria tomentosa); 2) Cascara Sagrada (Rhamnus Purshianus); 3) Ipê roxo (Tabebuia impetiginosa) e 4) Boldo (Phaemus boldus), a partir dos quais produziram-se quatro compostos de composição distinta. A amostragem para determinação de parâmetros de maturação (pH, condutividade elétrica, teores de amônio e nitrato, fracionamento da matéria orgânica, macronutrientes, carbono total. relação C/N e testes de germinação) foi feita aos 0, 30, 60, 90 e 120 dias de compostagem. A temperatura, entretanto, foi medida diariamente apenas até aos 90 dias. Na segunda etapa, buscando avaliar o potencial agronômico dos compostos sobre a produção da alface (Lactuca sativa), cultivar crespa Verônica, bem como o efeito residual dos materiais compostados instalaram-se experimentos em vasos com dois cultivos consecutivos. Nestes experimentos, constaram os seguintes tratamentos: composto 1 (C1) (Unha-de-gato), composto 2 (C2) (Cascara Sagrada), composto 3 (C3) (Ipê roxo), composto (4) (C4) mistura dos materiais nas doses 30, 60, 90 e 120t/ha. However, more accurate studies are required to improve the efficiency of the composting process, producing composts with better quality, supplying nutrients to soil and acting as a soil conditioning. This work aimed to evaluate composts quality produced by combination of residues obtained from medicinal plants industrial processing and bovine manure, verifying the agronomic potential of these obtained composts in lettuce production. In a first step, it was used four residues from medicinal plants industrial processing: 1) Unha-de-gato (Uncaria tomentosa); 2) Cascara Sagrada (Rhamnus Purshianus); 3) Ipê roxo (Tabebuia impetiginosa) e 4) Boldo (Phaemus boldus), which produced four organic composts with distinct composition. The sampling to determine maturation parameters (pH, electric conductivity, ammonium and nitrate, organic matter fractionation, macronutrients, total carbon, C/N relation and germination tests) was made at zero, 30, 60, 90 and 120 days after composting beggining. Temperature was daily verified until 90 days. In a second step, aiming to evaluate the agronomic potential of the composts in lettuce (Lactuca sativa) production,as well the residual effect of these composts in soil. Estudo da maturação epididimária em cães / Epididymal maturation in dogs

A hipótese desta pesquisa foi que a maturação epididimária dos espermatozoides caninos envolve modificações nas proteínas e ácidos graxos do fluido sintetizado pelos distintos segmentos epididimários, além de alterações no perfil de ácidos graxos da membrana plasmática, organização celular e morfológica dos espermatozoides. Para tanto, este projeto foi realizado com 20 cães em idade reprodutiva e negativos para Brucella canis. Após a orquiectomia bilateral, os testículos e epidídimos foram acondicionados a 5°C por no máximo 24 horas, em seguida, as amostras foram colhidas por incisões na cauda, corpo e cabeça do epidídimo. As amostras foram imediatamente processadas por avaliação subjetiva e automática da motilidade e vigor espermático, concentração, morfologia espermática, integridade da membrana plasmática, acrossomal e atividade mitocondrial. Foi possível observar determinar maior número de espermatozoides dentro da normalidade na cauda do epidídimo, especialmente, referindo-se à motilidade, integridade de membrana e atividade mitocondrial. A avaliação das modificações ultraestruturais dos espermatozoides permitiu observar a migração da gota citoplasmática e alterações acrossomais. No perfil de ácidos graxos observaram-se variações na quantidade e presença dos ácidos durante o traje epididimário, destacando o acréscimo do DHA na região da cauda epididimária. Ainda, no perfil protéico do plasma epididimário canino, foi possível identificar um padrão regional de secreção de proteínas, maior nas regiões da cabeça e corpo em relação à cauda do epidídimo. Apesar das importantes informações geradas a partir do presente trabalho, mais estudos são necessários para a compreensão da fisiologia reprodutiva, especialmente da maturação espermática epididimária na espécie canina. / The hypothesis of this research is that the canine maturation of epididymal spermatozoa involves progressive changes in the protein and fatty acid content of the fluid synthesized by the different epididymal segments, as well as changes in the fatty acid profile of the sperm plasma membrane, cell organization and morphology of the canine sperm. Therefore, this experiment was conducted with 20 dogs in reproductive age and free of Brucella canis. After bilateral orchiectomy, testes and epididymis were stored at 5°C for up to 24 hours and then the samples were harvested through incisions of the tail, corpus and caput of the epididymis. The samples were immediately processed for subjective and automatic motility and sperm vigor, sperm count, morphology, plasma membrane integrity, acrosomal and mitochondrial activity. It was possible to observe a greater number of mature sperm in the epididymal tail compared to the corpus and caput, especially referring to motility, membrane integrity and mitochondrial activity. The evaluation of ultrastructural changes of the spermatozoa allowed to observed the migration of the cytoplasmic droplets and acrosomal changes. In the fatty acid profile was observed variations in the amount and presence of acids during epididymal maturation, highlighting the addition of DHA in the epididymal tail region. Moreover, in the protein profile of the canine epididymal plasma, it was possible to identify a regional pattern of protein secretion, higher in the caput and corpus in relation to the tail epididymis. In spite of the important data generated from this study, further studies are needed for understand the reproductive physiology, especially the epididymal sperm maturation in dogs.

A resposta da produção in vitro de embriões (PIVE) é reduzida quando comparada à in vivo. O aprimoramento do conhecimento dos mecanismos de maturação de oócitos bovinos permite fornecer embasamento para incrementar os sistemas in vitro, aproximando-os do ideal fisiológico. O presente estudo visou investigar os efeitos da suplementação dos meios de maturação com FGF16 (10 ng/ml), BMP15 (100 ng/ml) e a interação de ambos sobre parâmetros relevantes ao desenvolvimento do complexo cumulus oócito (COC), tais como: expansão as células do cumulus (CC), fragmentação de DNA em CC e oócito, maturação nuclear oocitária, metabolismo energético e produção de progesterona. Os COC foram maturados em meios de tratamento (controle, FGF16, BMP15 e FGF16+BMP15) e avaliados em diferentes momentos da MIV (0 e 22 horas). A análise da expansão das CC demonstrou efeito positivo (p=0,0071) da BMP15 (11,34±1,09 unidade arbitrária/UA) e da combinação FGF16+BMP15 (11,34±0,61 UA) em relação ao grupo controle (8,73±0,44 UA) e ao suplementado com FGF16 (9,42±0,65 UA). A presença de fragmentação de DNA em CC (p=0,0015) e oócitos (p=0,036) foi significativamente menor em COC tratados com BMP15 (11,73±1,24 % e 3,81±2,76 %, respectivamente) em comparação ao grupo FGF16 (22,54±2,80 % e 31,13±7,81 %, respectivamente). Ainda, o FGF16 causou aumento na incidência de fragmentação de DNA em CC, quando relacionado ao controle (16,04±1,45 %). A taxa de maturação nuclear oocitária foi superior (p=0,014) no grupo suplementado com BMP15 (93,60±4,03 %) em comparação aos grupos controle (80,80±2,49 %) e FGF16 (76,75±2,28 %), aproximando-se da totalidade. De forma inédita, descrevemos ação da BMP15 (10,79±0,72 ng/ml) no incremento da produção de progesterona, sendo maior (p=0,0113) do que a produzida nos grupos controle (8,38±0,39 ng/ml) e FGF16 (8,84±0,45 ng/ml). Não foi evidenciado efeito dos tratamentos sobre o consumo de glicose e a produção de lactato. O presente estudo reforça o envolvimento da BMP15 na foliculogênese e na diferenciação do COC. Deste modo, a adição da BMP15 (100ng/ml) aos convencionais protocolos de PIVE pode ser de grande valia para elevar a efetividade desta biotecnologia. A suplementação de FGF16 (10ng/ml) se mostrou indiferente ao processo de maturação, permitindo inferir que o FGF16 não tenha envolvimento nas etapas da maturação compreendidas pelo presente estudo in vitro. Não foi observada ação sinérgica entre o FGF16 e a BMP15. / In vitro embryo production (IVEP) efficiency is reduced when compared to in vivo. Gaining knowledge of bovine oocyte maturation mechanisms will provide bases to improve in vitro systems. The present study assessed the in vitro effects of fibroblast growth factor 16 (FGF16), bone morphogenetic protein 15 (BMP15) and their interaction on relevant parameters to cumulus oocyte complex (COC) development, such as: cumulus cells (CC) expansion, oocyte and CC DNA fragmentation, nuclear maturation, energetic metabolism and progesterone production. COCs were matured in control or supplemented media containing, FGF16 (10ng/ml), BMP15 (100ng/ml), FGF16±BMP15 and analyzed at different times of IVM (0 and 22 hours). CC expansion evaluation demonstrated a positive effect (p=0.0071) of BMP15 (11.34±1.09 arbitrary unit/AU) and FGF16+BMP15 (11.34±0,61 AU) when compared to control (8.73±0.44 AU) and FGF16 groups (9.42±0.65 UA). The presence of DNA fragmentation in CC (p=0.0015) and oocytes (p=0.036) were lower in COCs treated in media supplemented with BMP15 (11.73±1.24 % and 3.81&plusmn2.76 %, respectively) in comparison to FGF16 group (22.54±2.80 % and 31.13±7.81 %, respectively). Moreover, FGF16 caused an increase in CC DNA fragmentation, when related to control (16.04±1.45 %). Oocyte nuclear maturation rate was higher (p=0.014) in groups supplemented with BMP15 (93.60±4.03 %) compared to control (80.80±2.49 %) and FGF16 treatments (76.75±2.28 %), almost reaching the totality of COCs. In an unprecedented way, we described the BMP15 increasing action on progesterone production (10.79±0,72 ng/ml; p=0.0113) when compared to control (8.38±0.39 ng/ml) and FGF16 groups (8.84±0.45 ng/ml). There were no differences in glucose consumption and lactate production. The present study reinforces BMP15 involvement in folliculogenesis and COC differentiation. FGF16 (10 ng/ml) media supplementation did not improve any of the outcomes measured, suggesting that FGF16 is not involved in the maturation steps analyzed in the present in vitro study. Thus, the inclusion of BMP15 (100 ng/ml) to conventional IVEP protocols can be valuable to increase the effectiveness of this biotechnology. Synergistic action between FGF16 and BMP15 was not observed.

Its trees are widely spread over Brazilian territory occurring more specifically on rain forest formations with seasonal semidecidious characteristics. Garapa seeds are an important source of medicinal substances as well as used for ornamental and environmental recovery purposes. It has been used on both tanning and timber industry and notably its natural populations have deeply decreased its growth due to massive exploitation. In this context, this work aimed to evaluate the development of A. leiocarpa seeds during the maturation phase. Researches were conducted for two successive crop years (2002 and 2003) and also analyzed seeds performance under specific storage conditions and the physiological quality of seeds. This scientific study results might be used as database information to guide the elaboration of specie s preservation proposals. Research samples were collected at Tingu? Biological Reserve, state of Rio de Janeiro, Brazil and biological analysis and tests were performed at Seeds Analysis Laboratory of Universidade Federal Rural do Rio de Janeiro at Serop?dica city, Brazil. This work results concludes that A. leiocarpa seeds physiological maturity was achieved within 98 days after anthesis (98DAA) in 2002. In 2003 it was not able to objectively determine the seeds maturity cycle. Seeds harvests are recommended as soon as fruits aspect changes from green-yellow to brown color combined with a higher proportion of brown seeds availability. The most efficient technique to overcome seed dormancy was the use of immersion treatment into concentrated sulphuric acid during 20 minutes. It was not observed any influence of either testa color or its size on A. leiocarpa seed germination process. The use of half-permeably packaging (polyethylene bags) under a nominal temperature of 18?C and 50% relative humidity was considered the most appropriate storage condition to A. leiocarpa seeds. / Apuleia leiocarpa ? uma esp?cie florestal, conhecida como gr?pia e garapa, que apresenta ampla distribui??o geogr?fica no territ?rio brasileiro, ocorrendo na mata atl?ntica, nas forma??es caracter?sticas da Floresta Estacional Semidecidual. Maturação in vitro de oócitos de caninos domésticos (Canis familiaris)

Este estudo foi realizado com o objetivo de: 1) determinar a influência da tensão de oxigênio na viabilidade das células do cumulus oriundas de oócitos caninos maturados em meio com alta concentração de glicose (11,0 mM); 2) determinar a taxa de maturação in vitro de oócitos caninos mantidos em meio contendo altas concentrações de glicose (11,0 mM), bem como avaliar o efeito da tensão de oxigênio sobre a taxa de maturação in vitro de oócitos caninos; 3) determinar a influência da adição de água de coco em pó (ACP-318®) ao meio de maturação com alta concentração de glicose (11,0 mM) na taxa de maturação in vitro de oócitos caninos. O meio de maturação usado nos experimentos foi TCM 199 suplementado com 26,19 mM de bicarbonato de sódio, 50 μg/ml de gentamicina, 0,20 mM de ácido pirúvico, 20 μg/ml de estradiol, 0,5 μg/ml de FSH e 0,03 UI/ml hCG. O meio de maturação era modificado de acordo com a proposta experimental apresentada. Os resultados do primeiro experimento mostraram diferença estatística nas taxas de apoptose nas células do cumulus entre os três grupos avaliados. Foi concluído que células do cumulus oriundas de COCs caninos cultivados em meio contendo altas concentrações de glicose apresentaram significativamente menos apoptose do que as cultivadas em meio com soro fetal bovino, e que a baixa tensão de oxigênio foi eficiente em reduzir a ocorrência de apoptose nas células do cumulus. No segundo experimento deste estudo, os índices mais elevados (p < 0,001) de oócitos degenerados foram obtidos, quando TCM 199 era suplementado com 10 % de FCS. Uma influência positiva sobre a retomada de meiose e aquisição de metáfase I (MI) foi observada quando os oócitos caninos foram maturados em meio livre de soro e com altas concentrações de glicose. Foi concluído que a adição de FCS ao meio de maturação de oócitos caninos resulta em altas taxas de degeneração dos oócitos e que a redução da tensão de oxigênio não resultou em melhora da taxa de maturação nuclear dos oócitos caninos. Os resultados obtidos no terceiro experimento mostraram que nos grupos experimentais cujo meio foi suplementado com ACP-318®, houve melhora significativa (p < 0,05) na taxa de maturação nuclear dos oócitos caninos. Oócitos maturados em meio suplementado com 5% de ACP-318® mostraram-se menos susceptíveis a alterações na configuração típica da cromatina que os maturados com 10% de ACP-318® no meio de maturação. Os resultados sugerem que tanto a integridade da morfologia nuclear, como a progressão da meiose dos oócitos caninos são positivamente influenciadas quando estes são expostos ao TCM 199 com alta concentração de glicose e suplementado com 5% de ACP-318®. / This study was designed: 1) determine the influence of oxygen tension on cumulus cell (CC) viability from canine oocytes in vitro matured in high glucose medium (11.0 mM); 2) determine the influence of two oxygen tensions on the nuclear maturation of canine oocytes in vitro matured in high glucose medium (11.0 mM); 3) determine the influence of powdered coconut water (ACP-318®) diluted in high glucose (11.0 mM) TCM199 in the achievement of nuclear in vitro maturation (IVM) of canine oocytes. Basic medium used in the experiments was TCM 199 suplemented with 26.19 mM sodium bicarbonate, 50 μg/ml gentamicin, 0.20 mM piruvic acid, 20 μg/ml oestradiol, 0.5 μg/ml FSH and 0.03 IU/ml hCG. Maturation medium was modified following the beyond described experimental proposals. The results of the first experiment showed that there was statistical difference in the rate of CCs apoptosis among the three groups. It was concluded that CCs of canine COCs cultured in high-glucose medium showed significantly less apoptosis than those cultured in medium with FCS. Low O2 tension was efficient in reducing apoptosis in canine CCs. In the second experiment of this study, the highest rates (p < 0.001) of degenerated oocytes were achieved when TCM 199 was added with 10% FCS. A positive influence on the meiosis resumption and on the MI acquisition rate was observed when canine oocytes were matured in defined high-glucose medium. It was concluded that the addition of FCS in the maturation medium of canine oocytes result in a high level of degenerated oocytes, and that the low level of oxygen tension did not improve the nuclear maturation of canine oocytes. The results achieved on the third experiment showed that in the experimental groups with the medium supplemented with ACP-318® (groups 2 and 3) enhanced the rates of oocytes’ nuclear maturation (p < 0.05). Ooocytes matured in medium added with 5% powdered coconut water (ACP-318®) were less prone to deviation of typical chromatin configuration than those matured in medium added with 10% ACP-318®. The results suggest that oocytes’ nuclear morphology integrity and meiosis achievement were positively influenced when exposed to high glucose TCM199 supplemented with 5% powdered coconut water.

Structural investigation of the archaeal replicative machinery by electron microscopy and digital image processing

Previous studies suggest a degree of homology between eukaryotic replication, transcription and translation proteins and archaeal ones. Hence, Archaea are considered a simplified model for understanding the complex molecular machinery involved in eukaryotic DNA metabolism. DNA replication in eukaryotic cells is widely studied. In recent years, DNA replication studies expanded on the archaeal DNA replication machinery. P. abyssi was the first archaeon whose genome was fully sequenced. Genome sequencing and comparative genomics have highlighted an MCM-like protein in P. abyssi. In this study, I report the biochemical and structural characterisation of PabMCM. PabMCM is explored as model for understanding more complex eukaryotic MCM proteins and unravelling the biochemical mechanism by which MCM proteins release their helicase activity. The crenarchaeon Sulfolobus solfataricus possesses a simplified toolset for DNA replication compared to Eukaryotes. In particular, S. solfataricus has a subset of the eukaryotic Okazaki fragment maturation factors, among which there are a heterotrimeric DNA sliding clamp, (the proliferating cell nuclear antigen, PCNA), the DNA polymerase B1 (PolB1), the flap endonuclease (Fen1) and the ATP-dependent DNA ligase I (LigI). PCNA functions as a scaffold with each subunit having a specific binding affinity for each of the factors involved in Okazaki fragment maturation. Here, the 3D reconstruction of PCNA in complex with the Okazaki fragment maturation proteins PolB1, LigI and Fen1 is reported.

Predicting physical activity behaviour across early adolescence

Physical activity (PA) has been labelled the miracle drug (Pimlott, 2010) and participating in regular PA has ample physical and mental wellbeing benefits. However, physical inactivity remains a critical public health concern, particularly across adolescence. In England the proportion of adolescents aged 13-15 years meeting the recommended guidelines for PA decreased significantly from those at a younger age (Health and Social Care Information Centre, 2009; 2012; 2015). The adolescent years (13 18 years) have been identified as the age of greatest decline in PA, although it is possible that large declines can also be seen at younger ages (Sallis, 2000). Among girls the decline in PA is greater at younger ages (9 12 years old) and among boys it is greater at older ages (13 16 years old) (Dumith, Gigante, Domingues, & Kohl, 2011). Thus, examining behaviour of early adolescents (aged 11-13 years) is a primary focus of this thesis. Researchers have called for a more comprehensive grasp of PA correlates and determinants and their impact on behaviour (Biddle & Mutrie, 2001, 2008). This broader picture needs to incorporate longitudinal study designs to accurately portray developmental changes (Evenson & Mota, 2011). This thesis aims to work towards a better understanding of associations among variables across aspects of the ecological model in relation to PA behaviour during early adolescence. Early adolescents within their first year of secondary school (year 7, aged 11 12 years) were recruited through schools across the East Midlands, United Kingdom (UK). These participants completed various measures across an 18 month period to compile all data required for the thesis. The thesis begins with a focus on active transport as a means of commuting to school which can significantly contribute to overall PA levels (Aibar, Bois, Generelo, Bengoechea, & Paillard, 2015; Slingerland, Borghouts, & Hesselink, 2012). The distance from home to school is an important influence on the decision to use active transport; however, ecological perspectives would suggest this variable may interact with individual, interpersonal and environmental factors. Therefore, the first study of this thesis investigates whether the relationship between distance to school and active transport is moderated by (i) gender, (ii) biological maturation, (iii) perceived family support for PA and (iv) multiple deprivation. Cross-sectional results from the baseline data collected demonstrated that the relationship between distance to school and the likelihood to actively travel to school is moderated by biological maturation, multiple deprivation and family support of PA in adolescents. Further analysis revealed that late-maturing children, those from less socio-economically deprived backgrounds and children with low family support of PA are less likely to actively commute to school as distance to school increases. Due to the interaction between these variables described above, the second study focused on the variables collectively using a person-oriented approach, which aimed to classify distinct profiles of early adolescents based on correlates of PA. The outcome variables were also broadened to include active transport and overall PA across two time points. Findings from this second study illustrate that the highly supported, shortest commuters produced the highest levels of self-reported PA and that affluent, short commuters were the most likely to use active transport to travel to school. The affluent, short commuters lived a relatively short distance to school in areas of the lowest deprivation and had relative moderate family support of PA. The highly supported, shortest commuters were characterised by the highest family support of PA and lived the shortest distance to school in areas of low deprivation. Study 1 evidenced an association between biological maturation and PA behaviour; however, study 2 displayed that biological maturation did not meaningfully contribute to the class characteristics, and were not a predictor of PA. Previous evidence as to whether early, average or late maturing adolescents are more likely to disengage from PA is mixed and tends to focus on one gender only (Sherar, Cumming, Eisenmann, Baxter-Jones, & Malina 2010; Bacil, Mazzardo, Rech, Legnani, & Campos, 2015). Thus for the third study a more focused inspection of biological maturity was undertaken. Biological maturity status was investigated as a predictor of PA behaviour at two subsequent time points (6 9 months after baseline and 12 18 months after baseline) and whether there was variation across genders. Findings displayed that biological maturity status does not predict subsequent PA, with no distinction across genders. To conclude, the final study examined additional forms of PA behaviour. For children to develop and maintain healthy PA behaviours, their PA during the school day, particularly during physical education (P.E) classes is important (Owen, Smith, Lubans, Ng, & Lonsdale, 2014). Self-reported PA was divided into school-time PA (during P.E. lessons, break and lunchtimes) and leisure-time PA (after school, during evenings and weekends). The final study fully utilised the longitudinal data collected and utilised longitudinal growth modelling to describe the changes in PA behaviour across 12-18 months during early adolescence. Results displayed that school-time PA and leisure-time PA are distinct. Males; those from less deprived backgrounds and individuals with higher family support of PA all separately reported more school-time PA than their counterparts (females, those from higher deprived backgrounds and individuals with lower family support of PA) at baseline. Males and those with higher family support of PA also reported more leisure-time PA than their respective counterparts at baseline. On average, both genders decreased in school-time PA across 18 months yet for leisure-time PA, on average, there was no change over time and no significant difference in the rate of change between genders. There were no observed significant differences in the rate of change between multiple deprivation status and biological maturation across the 18 months for both behaviours. For family support, on average school-time PA decreased over time and results showed significant difference in the rate of change between individuals with lower or higher levels of family support of PA across the 18 months. On average, there was no change over time for leisure-time PA yet there was a significant difference in the rate of change between individuals and their family support of PA across 18 months. Further analysis demonstrated if an individual s family support increases, so does their leisure-time PA and vice versa. These overall key findings demonstrate the complexity of PA behaviour throughout early adolescence. This thesis works towards predicting individuals, correlates and determinants that may be susceptible to physical inactivity and/or a decrease in activity over time. Results can be used to target and direct PA intervention work.
