Mise au point des espèces du genre Stephania au Cambodge : études systématique, phytochimique et pharmacologique / Update on the species of the genus Stephania in Cambodia : systematic, phytochemical and pharmacological studiesDary, Chhavarath 18 July 2016 (has links)
The thesis aims to provide update on species of the genus Stephania recorded in Cambodia by systematic, phytochemical and pharmacological studies. The systematic study allowed to list seven species and two varieties of Stephania in Cambodia: S. cambodica, S. japonica var. discolor and var. timoriensis, S. oblata, S. pierrei, S. rotunda (type species), S. suberosa and S. venosa. The presence of S. japonica var. timoriensis and S. oblata is reported for the first time in Cambodia. According to the phylogenetic results, only S. pierrei is monophyletic, which represents less than 10% of species in the genus Stephania. Chemical studies on the tubers of five species were carried out using chromatographic methods (HPLC, UHPLC) and spectroscopic methods. Different isolated alkaloids have been proposed as markers: palmatine, roemerine and tetrahydropalmatine for S. cambodica and S. rotunda, cepharanthine, tetrahydropalmatine, and xylopinine for S. suberosa, cepharanthine for S. pierrei, crebanine, and tetrahydropalmatine for S. venosa. Nine alkaloids belonging to five classes were isolated from the tuber of S. cambodica and a new compound namely "angkorwatine" was described. Ultrasound assisted extraction of alkaloids was developed and optimized by Design of Experiment for this species. This innovative extraction is a good alternative to conventional methods. An analytical UHPLC method was developed and validated for the quality control of S. cambodica. Results show that hydroethanolic extracts obtained by maceration of S. suberosa, S. rotunda and S. venosa exhibit significant anti-inflammatory activity with high anti-inflammatory ratio (selectivity index).

From Transformation to Therapeutics : Diverse Biological Applications of Shock WavesGanadhas, Divya Prakash January 2014 (has links) (PDF)
Chapter–I Introduction
Shock waves appear in nature whenever the different elements in a fluid approach one another with a velocity larger than the local speed of sound. Shock waves are essentially non-linear waves that propagate at supersonic speeds. Such disturbances occur in steady transonic or supersonic flows, during explosions, earthquakes, tsunamis, lightening strokes and contact surfaces in laboratory devices. Any sudden release of energy (within few μs) will invariably result in the formation of shock wave since it is one of the efficient mechanisms of energy dissipation observed in nature. The dissipation of mechanical, nuclear, chemical, and electrical energy in a limited space will result in the formation of a shock wave. However, it is possible to generate micro-shock waves in laboratory using different methods including controlled explosions. One of the unique features of shock wave propagation in any medium (solid, liquid or gases) is their ability to instantaneously enhance pressure and temperature of the medium. Shock waves have been successfully used for disintegrating kidney stones, non-invasive angiogenic therapy and osteoporosis treatment. In this study, we have generated a novel method to produce micro-shock waves using micro-explosions. Different biological applications were developed by further exploring the physical properties of shock waves.
Chapter – II Bacterial transformation using micro-shock waves
In bacteria, uptake of DNA occurs naturally by transformation, transduction and conjugation. The most widely used methods for artificial bacterial transformation are procedures based on CaCl2 treatment and electroporation. In this chapter, controlled micro-shock waves were harnessed to develop a unique bacterial transformation method. The conditions have been optimized for the maximum transformation efficiency in E. coli. The highest transformation efficiency achieved (1 × 10-5 transformants per cell) was at least 10 times greater than the previously reported ultrasound mediated transformation (1 × 10-6 transformants per cell). This method has also been successfully employed for the efficient and reproducible transformation of Pseudomonas aeruginosa and Salmonella Typhimurium. This novel method of transformation has been shown to be as efficient as electroporation with the added advantage of better recovery of cells, economical (40 times cheaper than commercial electroporator) and growth-phase independent transformation.
Chapter – III Needle-less vaccine delivery using micro-shock waves
Utilizing the instantaneous mechanical impulse generated behind the micro-shock wave during controlled explosion, a novel non-intrusive needleless vaccine delivery system has been developed. It is well established, that antigens in the epidermis are efficiently presented by resident Langerhans cells, eliciting the requisite immune response, making them a good target for vaccine delivery. Unfortunately, needle free devices for epidermal delivery have inherent problems from the perspective of patient safety and comfort. The penetration depth of less than 100 µm in the skin can elicit higher immune response without any pain. Here the efficient utilization of the device for micro-shock wave mediated vaccination was demonstrated. Salmonella enterica serovar Typhimurium vaccine strain pmrG-HM-D (DV-STM-07) was delivered using our device in the murine salmonellosis model and the effectiveness of the delivery system for vaccination was compared with other routes of vaccination. The device mediated vaccination elicits better protection as well as IgG response even in lower vaccine dose (ten-fold lesser), compare to other routes of vaccination.
Chapter – IV In vitro and in vivo biofilm disruption using shock waves
Many of the bacteria secrete highly hydrated framework of extracellular polymer matrix on encountering suitable substrates and get embedded within the matrix to form biofilm. Bacterial colonization in biofilm form is observed in most of the medical devices as well as during infections. Since these bacteria are protected by the polymeric matrix, antibiotic concentration of more than 1000 times of the MIC is required to treat these infections. Active research is being undertaken to develop antibacterial coated medical implants to prevent the formation of biofilm. Here, a novel strategy to treat biofilm colonization in medical devices and infectious conditions by employing shock waves was developed. Micro-shock waves assisted disintegration of Salmonella, Pseudomonas and Staphylococcus biofilm in urinary catheters was demonstrated. The biofilm treated with micro-shock waves became susceptible to antibiotics, whereas the untreated was resistant. Apart from medical devices, the study was extended to Pseudomonas lung infection model in mice. Mice exposed to shock waves responded well to ciprofloxacin while ciprofloxacin alone could not rescue the mice from infection. All the mice survived when antibiotic treatment was provided along with shock wave exposure. These results clearly demonstrate that shock waves can be used along with antibiotic treatment to tackle chronic conditions resulting from biofilm formation in medical devices as well as biological infections.
Chapter – V Shock wave responsive drug delivery system for therapeutic application
Different systems have been used for more efficient drug delivery as well as targeted delivery. Responsive drug delivery systems have also been developed where different stimuli (pH, temperature, ultrasound etc.) are used to trigger the drug release. In this study, a novel drug delivery system which responds to shock waves was developed. Spermidine and dextran sulfate was used to develop the microcapsules using layer by layer method. Ciprofloxacin was loaded in the capsules and we have used shock waves to release the drug. Only 10% of the drug was released in 24 h at pH 7.4, whereas 20% of the drug was released immediately after the particles were exposed to shock waves. Almost 90% of the drug release was observed when the particles were exposed to shock waves 5 times. Since shock waves can be used to induce angiogenesis and wound healing, Staphylococcus aureus skin infection model was used to show the effectiveness of the delivery system. The results show that shock wave can be used to trigger the drug release and can be used to treat the wound effectively.
A brief summary of the studies that does not directly deal with the biological applications of shock waves are included in the Appendix. Different drug delivery systems were developed to check their effect in Salmonella infection as well as cancer. It was shown for the first time that silver nanoparticles interact with serum proteins and hence the antimicrobial properties are affected. In a nutshell, the potential of shock waves was harnessed to develop novel experimental tools/technologies that transcend the traditional boundaries of basic science and engineering.

Etude comparative et optimisation de prétraitements des écorces de bois pour l'extraction des composés phénoliques / Comparative study and optimization of pre-treatment of wood bark for the extraction of phenolic compoundsBouras, Meriem 18 December 2015 (has links)
This research work focuses on the intensification of polyphenols extraction from Quercus robur and Norway spruce barks using innovative technologies : pulsed electric field (PEF), ultrasound assisted extraction (UAE) and microwave assisted extraction (MAE). These pre-treatments enhance the release of intracellular compounds by damaging the membrane and/or cell wall. The effect of each of these technologies (PEF, UAE and MAE) on polyphenols extraction has been highlighted in a quantitative and qualitative optimization analysis by evaluating the polyphenols concentration, antioxidant activity and by the chemical characterization of the extracted compounds.A comparative study of pre-treatments (PEF, UAE and MAE) coupled with an extraction step in an alkaline hydro-alcoholic medium allowed us to better understand the involved phenomena. The induced process efficiency is independent of the tree family to which the bark belongs (coniferous and hardwood). For bark valorization, the application of PEF treatment (20 kV/cm, 200 pulses, 3.2 kJ/g bark) followed by a diffusion step seems to be an alternative to ultrasound and microwave assisted extractions and to the extraction from grinded barks. In fact, the electrical and mechanical effect of PEF treatment allow to obtain an extract rich in polyphenols (10.5 g GAE/100g DM and an efficiency of 83 % for Norway spruce and 5 g GAE /100g DM and an efficiency of 82 % for Quercus robur bark).

In this work two multiresidual methods for extracting contaminants from sediments were developed. In the first part of this study, a method was developed for extracting simultaneously two groups of oxygenated metabolites of polycyclic aromatic hydrocarbons (PAHs), quinones and hydroxylated PAHs (hydroxy-PAHs). A chemometric approach allowed us to determine the influential parameters on microwave assisted extraction (MAE), and a compromise could be found for extracting quantitatively both families of compounds. Two chromatographic analytical methodologies were developed and validated for analysing the extracts: high performance liquid chromatography coupled with fluorimetric and ultraviolet detection (HPLC-UV-FLD) and gas chromatography coupled with an electronic impact mass spectrometer (GC-MS). Using GC-MS, reactions of silylation of hydroxy-PAHs and of acetylation of quinones had to be developed, to decrease detection limits (LOD), particularly for ortho-quinones. Using HPLC-UV-FLD, LODs were lower than using GC-MS, particularly for hydroxy-PAHs detected by FLD, and the analysis was faster, without derivatization; but the detectors were not selective, and identification of analytes was doubtful. Choice was done to favour GC-MS for a more reliable analysis of the two families of compounds extracted from naturally contaminated sediments. In the second part of this thesis work, a new fast and simple extraction methodology was developed and validated, based on matrix solid phase dispersion (MSPD), capable of extracting and purifying simultaneously sediment samples. Two families of compounds were simultaneously extracted from sediments, PAHs and polychlorobiphenyls (PCBs). Many parameters were optimized, as the nature of dispersing agents, the time of grinding, the volume and nature of elution solvent mixtures. In a second step, hydroxy-PAHs were introduced in the analytical process, which led us to add another more selective extraction/purification method to MSPD, based on molecularly imprinted polymers (MIPs). Indeed polar interfering compounds, trapped by the polar dispersant in the first cartridge containing the blended sediment, had to be eluted to release hydroxy-PAHs, which in turn had to be selectively retained by the polymer, imprinted for phenols, to provide a final eluate free from other polar compounds. It was demonstrated that those MIPs could selectively extract low and high molecular weight hydroxy-PAHs, but appropriate percolating solvents had to be chosen to avoid polymer damages. However, the main difficulty was to desorb hydroxy-PAHs strongly retained by the sediment matrix through hydrogen bonds. It could be achieved for light hydroxy-PAHs, using a mixture of eluting solvents with salting-out effect, but not for heavy hydroxy-PAHs which stayed strongly sorbed on the sediment matrix. Furthermore we needed to use high amounts of imprinted polymer because of the competition for recognition sites between hydroxy-PAHs and phenolic compounds.

Savremene metode ekstrakcije očajnice (Marrubium vulgare L.) i sušenje odabranih ekstrakata, hemijski sastav i biološke aktivnosti / Contemporary methods of extraction of white horehound (Marrubium vulgare L.) and drying of selected extracts, chemical composition and biological activitiesGavarić Aleksandra 11 December 2020 (has links)
Within the scope of this dissertation, the possibilities of applying modern methods for obtaining liquid (ultrasonic extraction, microwave extraction and subcritical water extraction) and lipophilic extracts (extraction with supercritical carbon dioxide) rich in polyphenols from the aboveground part of white horehound (Marrubium vulgare L.) were examined. Liquid extracts were optimized using an experimental design and response surface method, and subjected to spray drying. Liquid and dry extracts have been extensively examined for their chemical composition and biological activity, antimicrobial and in vitro antihyperglycaemic activities.<br />For each of these extraction techniques, the influence of the main parameters was examined: ultrasonic extraction (temperature, extraction time and ultrasound power), microwave extraction (ethanol concentration, extraction time and microwave power) and subcritical water extraction (temperature, extraction time and HCl concentration in the extractant). Liquid extracts obtained by ultrasonic and microwave extraction under optimized conditions revealed the presence of chlorogenic acid and quercetin, which were not detected in macerates. The antimicrobial activity of both extracts was examined and B. cereus was found to be the most sensitive bacterial strain, whereas in the case of yeast S. cerevisiae was particularly sensitive to both extracts. In addition to this activity, the in vitro antihyperglycaemic activities of both extracts against α-amylase and α-glucosidase were investigated, with ultrasound extract showing higher antihyperglycemic activity (50.63% inhibited α-amylase and 48.67% inhibited α-glucosidase) which could potentially be explained by the presence of chlorogenic acid (33.11 μg/mL) and quercetin (34.88 μg/mL). In the subcritical extract, obtained under optimal conditions, only the presence of hydroxy methylfurfural was detected, and this extract was not subjected to antimicrobial and antihyperglycemic activity.<br />For the extraction of lipophilic components of M. vulgare, distillation of essential oil (0.05%), Soxhlet extraction (3.23%) and extraction with supercritical carbon dioxide at pressures of 100, 200 and 300 bar and at temperatures of 40, 50 and 60 ° C were applied. The most dominant component in all the supercritical extracts obtained is the chemotaxonomic marker marubiin, which was not identified in the Soxhlet extract.<br />The liquid extracts obtained by the ultrasound and microwave extraction under optimized conditions and the macerate were spray dried (with the addition of 10 and 50% maltodextrin) to obtain a more stable form of the dry extract. Dry extracts underwent determination of the most important physical properties and chemical composition as well as antimicrobial and antihyperglycemic activities to evaluate their incorporation into functional foods, dietary supplements or in the formulation of various finished dosage forms that are administered preventively or to improve human health.</p>
