The diploma thesis is focused on the use of the ISSR and SSR methods for the purpose of genetic analysis of 9 varieties of pepper (Capsicum annuum). The literature review describes the basic characteristics of the pepper. The next part is focused on the description of molecular methods. The aim was to find suitable ISSR primers and SSR markers that can distinguish individual varieties of pepper. Based on the results dendrograms of genetic related of the studied varieties were created.

The genetic variability and diversity of the Old Kladruber horse breed is evaluated in this master thesis. The aim is to evaluate the variability and diversity in the breed, between the sire lines and inside these lines. The detailed information about the Old Kladruber horse breed is presented in the literary overview with the breed history including the regeneration process. Basic indexes of the genetic variability are presented in the next part. 17 microsatellite markers were used to evaluate the genetic variability and diversity with further statistical analyses performed in GENEPOP 4.2 and ARLEQUIN 3.5 programs. The allele and genotype frequencies were calculated together with the number of alleles per loci and Hardy -- Weinberg equilibrium was tested for the population. Polymorphic information content, observed and expected heterozygosity and fixation coefficients were calculated. The results show surprisingly high degree of genetic variability in the Old Kladruber horse, increased probably due to introduction of other breeds during the regeneration process.

@Adansonia digitata@L. (Malvaceae) is a huge multipurpose tree of the savannahs of sub-Saharan Africa with high economic potential for local communities. The edible fruits and leaves are known for their high nutritional values and can be used fresh or processed. However, a high intra-specific variability regarding morphology, genetics and nutritional content of baobab and its products is documented for several African regions, while data for Kenya is largely lacking. This study aimed at documenting the genetic and morphological variability of baobab accessions in Kenya and at checking the presence of the newly described diploid baobab species @Adansonia kilima@. Samples were collected from 204 baobab trees from seven populations defined by geographical distance in South-eastern and Coastal Kenya at altitudes of 6-1,058 m asl. Leaf or bark samples for genetic diversity assessment were collected from all 204 trees, while leaves only from 65 and fruits from 76 trees (all in inland locations) for morphological analyses based on the publication Descriptors for Baobab. Nine microsatellite loci were used to assess genetic variation and results analysed with specific software because of the tetraploid nature of baobab. Overall genetic diversity was high and all loci were polymorphic. The mean gene diversity was 0.803 and observed heterozygosity was 0.907. Analysis of molecular variance (AMOVA) revealed low variation among populations (12.4%) and high variation within populations (87.6%). Bayesian clustering and Principal Coordinate Analysis divided the accessions into two clusters, one with only inland and one with coastal accessions. Although the presence of @Adansonia kilima@ was previously postulated for Kenya, flow cytometry did not detect any among the analysed samples as only tetraploids were observed. Regarding morphological characteristics, no differences among the fruit accession from inland populations were found (no fruits were collected in coastal areas). Leaf morphological data showed significant differences between inland and coastal populations with longest leaflets and leaf petioles in accession from the Coast, thus confirming the results obtained for genetic analysis. This study contributes to the overall knowledge of the genetic diversity of baobab in Kenya and can contribute to the development of germplasm conservation strategies and domestication programs for baobab.

A switch from hunting to beginnings of farming and agriculture was a crucial step for humans and for developing civilization. This period was suitable for domestication processes. Dogs were the first domesticated animals at all. Their breeding was connected with settlement and cattle protection and they were used as shepherd and hunting dogs. In some cultures they are even used as food source. Recent trends lead to breeding of new breeds and division of original gene pool into smaller closed groups which lead to an increase of homozygosity and to display recessively inherited diseases. These effects are mostly visible in small local breeds that have a small population size. Global protection of genetic diversity in cattle, poultry and pigs was already addressed in many studies, probably because of the high economic value of these animals. Impact of domesticated carnivores to humans is comparable and due to the growing market in veterinary services it is needed to protect sufficient genetic variability of various unique races. The aim of this work is to compare selected local breeds of dogs from different regions of origin, to evaluate their overall genetic diversity, to determine the occurrence of candidate types of diseases and to compare the frequency of these diseases in selected breeds. Main output is to recommend appropriate methods to protect the gene pool of these unique races. Central European breed (e.g. Český Fousek), subtropical breed (e.g. Sarplaninec), tropical continental breed (eg. Basenji), tropical island breed (e.g. The New Guinea Singing Dog) will be selected. The material will be collected at exhibitions and in cooperation with local farmers. The samples will be processed by classical methods of population genetics in the laboratory of molecular genetics FTZ ČZU.

Diploma thesis deals with genetic variability and evolutionary relationships among selected species of the group Chenopodium album and is especially focused on the origin of hexaploid species C. album in the Czech Republic. The main aim of the study was detection of putative recent emergence of hexaploid C. album s. str. in mixed populations of diploid (C. ficifolium and C. suecicum) and tetraploid (C. strictum and C. striatiforme) species. To assess phylogenetical relationships I performed an analysis of nuclear microsatellite loci, that are suitable for detection parental lines within polyploid species. As long as microsatellite primers have been developed for studied group, therefore they had to be cross-amplified from closely related species C. quinoa. Three PCR multiplexes were assembled by cross-amplification of microsatellite primers. These PCR multiplexes were tested on representative sample sets to estimate genetic variability of individual microsatellite loci. A total of 911 individuals were analyzed from five localities from the Czech Republic. Representatives of family Bovidae are subjects of many studies concerning with their phylogeny, phylogeography, time of divergence or genetic diversity. Taxonomy is solved by comparison of morphological characteristics or by genetic approaches, genetic diversity could be solved by pedigree or by genetic analyses too. Tragelaphinae number nine species of two genera, Tragelaphus sp. and Taurotragus sp. The antelopes of the genus Taurotragus (T. derbianus and T. oryx) belong to the largest antelopes of the world. Derby eland (Taurotragus derbianus) has two subspecies, Western Derby eland (T. d. derbianus) and Eastern Giant eland (T. d. gigas), which are distinguished on the basis of morphological characteristics. Western subspecies (T. d. derbianus) is classified as critically endangered. There lives the only population in Niokolo Koba National Park in Senegal, which numbers fewer than 200 individuals. For the conservation, the semi-captive breeding programme has been established in 2000. It was created by six founders (one male and five females), which are presumed to be non-related. The population within this programme had 95 living individuals in 2013, living in seven herds in Bandia and Fathala reserves in Senegal. The population is under breeding management, which efforts to minimize kinship of the individuals. Studbook was established for the Western Derby eland (T. d. derbianus) in 2008 and is published annually. It acts about small population with low number of founders and no gene flow, which is threatened by inbreeding and genetic drift. Genetic diversity of the population was evaluated by means of microsatellite markers and the results were compared with the results of pedigree analysis. Pedigree analysis showed the highest genetic diversity in the generation of founders (FOUNDERS). It decreased in the generation of founders' offspring (OFFSPRING 1; born in season 2007/2008), due to the fact, that the only male took part in the reproduction. And it increased again in the generation of offspring of founders' offspring (OFFSPRING 2; born in the season 2009/2010), because more individuals were included into the reproduction. Fifteen individuals and five polymorphic microsatellite loci (from the total number of 13 tested loci) were chosen for the genetic study. The parameters of genetic diversity (HE and HO, Ar and deviations from Hardy-Weinberg equilibrium, and FIS and FST) were evaluated. Not any deviations from Hardy-Weinberg equilibrium were found out. The results of genetic analysis confirmed the highest genetic diversity in the population of founders (Ar = 2.79; HE = 0.664; HO = 0.750; FIS = --0.154). In both generations of offspring values of allelic richness and observed and expected heterozygosity decreased (Ar = 2.15; HO = 0.580; HE = 0.586 in OFFSPRING 1 and Ar = 2.14; HO = 0.370; HE = 0.480 in OFFSPRING 2). Contrary to the results of pedigree analysis, there was not been observed any improvement in OFFSPRING 2. The resultant values of genetic diversity parameters were quite satisfactory, despite of the low number of founders and mating of related individuals.

The use of candidate genes has become a widespread approach in the study of behavioral adaptations. Gene function is often maintained in very distant evolutionary lines. This approach allows us to extend knowledge about non-model species. I studied the influence of selection on candidate behavioral adaptation genes associated mainly with biorhythms. I was interested in comparing genetic variability between closely related species of songbirds from tropical and temperate zones. These environments differ mainly in the conditions stability. Tropical species live in very stable conditions with generally low seasonality, unlike temperate species. Timing of events of cirkanual cycle of temperate birds is essential because of the alternating of conditions of the environment. I therefore assumed a reduction in genetic variability in migrating temperate zone species compared to tropical species due to stabilizing selection. The study is based on analyzes of microsatellite loci in the exon region. I involved analysis of neutral microsatellites as a control for possible genetic variability reduction coming from different reasons than selection. Arthroderma benhamiae has been almost unknown among clinical mycologists but it is a cause of around 40 % of dermatophytic infections according to current studies in the Czech Republic. The species is primarily transmitted to humans from guinea pigs and other rodents. The epidemiological situation is similar in other countries in Central and Western Europe. The reason of significant increase in the incidence of infections has not been identified yet, and no sufficiently informative molecular markers have been developed for typification of the species that could help to resolve the cause of this problem. For the purposes of this study, isolates (n=268) from cases of human and animal dermatophytoses from the Czech Republic, Germany, Belgium, Switzerland, Italy, Japan and USA were selected. Ten variable microsatellite markers were developed and sequence analysis of two genetic loci (ITS rDNA and gpdh gene) were performed to reveal intraspecific variation. Phenotype was also studied at the level of micro- and macromorphology of the strains and growth parameters at several temperatures and on several cultivation media.
