Métodos: inicialmente a amostragem de indivíduos foi de 49. O grupo controle (C, n= 16) realizou um programa de condicionamento leve (1 sessão/dia/ 2x/sem./12 m.min-1). Os outros ratos (n= 33) foram submetidos ao PO, com duração de 12 sem., prescrito para produzir um desequilíbrio entre as sessões de exercício e o período de recuperação. Em 8 sem. foram realizadas sessões de treino diárias, por 5 d consecutivos seguidas por 2 d de recuperação. Nas 4 sem. subsequentes, houve aumento da frequência de treino (2, 3, 4 e 4 x/d) e redução do tempo de recuperação entre as sessões (4, 3, 2 e 2 h). Para avaliação da aptidão física, os ratos foram submetidos a sete testes de esforço máximo (T-máx) durante e dois testes após o PO. Dois grupos foram identificados pela diferença da inclinação (α) obtida a partir da regressão linear dos desempenhos individuais nos Tmáx-3, 4, 5, 6 e 7. Ratos NFOR (n= 14) tiveram α= < -0.98. Analisou-se a recuperação passiva de 2 sem., sendo realizado testes de Campo Aberto (CA) e Labirinto em Cruz Elevado (LCE). Neste período, dois Tmáxs (8 e 9) foram realizados para verificação da continuidade das condições FOR e NFOR. Em Tmáx-2, Tmáx-6 e Tmáx-9 realizaram-se coletas sanguíneas para quantificação das concentrações plasmáticas de lactato ([lac]antes e [lac]após) e glicose ([glic]antes e [glic]após). Quantificou-se corticosterona durante o repouso e após os Tmáx-7 e 9, ([cort]repouso e [cort]após). Glândulas adrenais foram coletadas e pesadas. Para análise estatística aplicaram-se ANOVA de uma via e para medidas repetidas no tempo, testes t de Student pareado e não pareado e foi realizada correlação de Pearson (P< 0.05). Resultados: FOR e NFOR tiveram aumento de desempenho a partir do Tmáx-2 (303.8±13.8; 294.3±18.9 e 190.2±12.2 kg.m, respectivamente). O grupo NFOR teve o desempenho reduzido no Tmáx-7 (292.2±36.4 kg.m). No teste CA, ocorreu aumento da frequência de levantar para os grupos FOR e NFOR em relação ao grupo C. Houve redução da [lac]após para FOR e NFOR no Tmáx-6 em relação ao grupo C (3.61±0.38; 5.43±0.63; 7.64±0.43 mmol/L, respectivamente) sendo que NFOR obteve lactatemia maior que FOR. Ainda no Tmáx-6, a [glic]após foi menor em FOR em relação ao grupo C (5.66±0.17 vs. 6.52±0.21). Não houve diferença para as concentrações de corticosterona. Houve aumento do índice adrenosomático para o grupo NFOR no Tmáx-7 e redução deste após o Tmáx-9. Conclusões: "functional" ou "nonfunctional overreaching" provocou ansiedade e alterações nas concentrações de lactato e glicose em ambos os grupos bem como aumento do índice adrenosomático nos ratos que demonstraram a condição NFOR. / Purpose: To investigate the behavior, corticosterone and lactate concentrations of Wistar rats submitted to an overtraining protocol (OP) for the development of functional (FOR) or nonfunctional overreaching (NFOR). Methods: Sampling of rats was 49. The control group (C, n= 16) performed a mild conditioning program (1 session/day / 2x/wk/12 m.min-1). The other rats, n= 33, were submitted to 12-week OP prescribed to produce an imbalance between the exercise sessions and the recovery period. In 8 wks. daily training sessions were performed for 5 consecutive d followed by 2 d of recovery. In the following 4 wks. (2, 3, 4 and 4 x / d) there was an increase in training frequency and reduction of recovery time between sessions (4, 3, 2 and 2 h). To evaluate physical fitness, the rats were submitted to seven maximum tests (Tmax) during and two tests after the PO. Two groups were identified by the slope difference (α) obtained from the linear regression of individual performances at Tmax-3, 4, 5, 6 and 7. NFOR rats (n= 14) had α = -0.98. Passive recovery of 2 wks was analyzed, and Open Field (OF) and Elevated Plus Maze (EPM) tests were performed. In this period, 2 Tmaxs (8 and 9) were performed to verify the continuity of FOR and NFOR conditions. Blood samples were collected for Tmax-2, Tmax-6 and Tmax-9 for the quantification of plasma lactate concentrations ([lac]before and [lac]after) and glucose ([glic]before and [glic]after). Corticosterone was measured at rest and after Tmax-7 and 9, ([cort]rest and [cort]after). Adrenal glands were collected and weighed. For statistical analysis one-way ANOVA and repeated measures ANOVA, paired and unpaired Student's t-tests and a Pearson correlation were performed (P < 0.05). Results: NFOR and FOR had an increase in performance from Tmax-2 (303.8±13.8, 294.3±18.9 and 190.2±12.2 kg.m, respectively). The NFOR group had reduced performance in Tmax-7 (292.2±36.4 kg.m). In the OF test, there was an increase in the frequency of rearing for the FOR and NFOR groups in relation to the C group. There was reduction of [lac]after for FOR and NFOR in Tmax-6 compared to group C (3.61±0.38, 5.43±0.63 , 7.64 ± 0.43 mmol/L, respectively) and NFOR obtained lactate concentrations greater than FOR. Still in Tmax-6, [glic]after was lower in FOR compared to group C (5.66±0.17 vs. 6.52±0.21). In Tmax-7, resting was lower for the NFOR group compared to C. There was no difference in corticosterone concentrations. There was an increase in the adrenosomatic index for the NFOR group in the Tmax-7 and a reduction in the latter after the Tmax-9. Conclusion: functional and nonfunctional overreaching caused anxiety and changes in lactate and glucose concentrations in both groups as well as increase in the adrenosomatic index in rats that demonstrated the NFOR condition.

Maternal Separation in Rats : An Experimental Model for Long-Term Effects of Early Life Experiences on Neurochemistry, Voluntary Ethanol Intake and Exploration and Risk Assessment Behavior

The period of early life is important for the development of individual brain function and behavior. Human studies have shown altered vulnerability to develop psychopathology and/or excessive drug intake, possibly leading to dependence, as a consequence of early life experiences. In the present thesis, maternal separation (MS), an experimental model for studies of early environmental influences, was used to investigate long-term effects on neurochemistry, voluntary ethanol intake and exploration and risk assessment behavior in rats. Rat pups were assigned to one of three different rearing conditions: daily 15 min (MS15) or 360 min (MS360) of MS and normal animal facility rearing (AFR) during the first three weeks of life. Measurements of adult endogenous opioid peptide levels, opioid- and dopamine receptor density revealed minor MS-induced effects on the opioid system whereas interesting alterations were found in dopamine receptor density. Long-term effects on voluntary ethanol intake showed distinct MS-induced alterations in male Wistar and ethanol-preferring AA (Alko, Alcohol) rats. Female Wistar rats were unaffected, indicating sex differences in the effects of MS on ethanol intake. Male MS15 rats generally had a slower acquisition phase and a low subsequent ethanol intake whereas male MS360 rats had a high ethanol intake. MS15 is therefore suggested to protect against a high voluntary ethanol intake in male rats whereas MS360 may serve as a risk factor. The recently established concentric square field test indicated alterations in risk assessment as well as an increased exploratory drive and somewhat higher risk-taking behavior in adult MS360 rats, while minor effects were seen in MS15 rats. Altogether, these results demonstrate that environmental influences during the period of early life can have long-term effects on neurochemistry and behavior. Of special interest is the finding that MS altered the inherited high ethanol intake in adult ethanol-preferring AA rats.

The Impact of Growth Hormone and Gamma-Hydroxybutyrate (GHB) on Systems Related to Cognition

Drug dependence is a serious and increasing problem in our society, especially among adolescents. The use of the large variety of substances available can result in a range of physiological and psychological adverse effects on individuals and negative consequences on the society overall. Several different types of drugs induce neurotoxicological damages, which in turn can generate impairment in for example the reward system and affect cognitive parameters.  The drug gamma-hydroxybutyrate (GHB) is usually considered a harmless compound among abusers, but has now shown to be highly addictive. Furthermore, GHB can cause memory impairments in both humans and animals. On the contrary, growth hormone (GH) and its main mediator insulin-like growth factor 1 (IGF-1) have recently been suggested to improve memory and learning in several studies. The hormones exhibit certain neuroprotective capabilities and have also previously been demonstrated to reverse opioid induced apoptosis in hippocampal cells. These effects and the fact that GHB is shown to increase GH secretion, which attracted considerable attention among body builders, led us to initiate studies on GHB and its impact on relevant systems in the central nervous system (CNS). Thus, the main purpose of the present investigation was to elucidate some of the underlying mechanisms that could account for the effects exerted by GH and GHB in the CNS. We found that a) GH affects the density and functionality of GABAB-receptors and opioid receptors in the male rat brain, b) GHB induces cognitive deficits and down-regulates GABAB-receptors, c) GHB treatment creates an imbalance between the endogenous opioids Met-enkaphalin-Arg6Phe7 (MEAP) and dynorphin B and increases the levels of MEAP in regions of the brain that are associated with drug dependence, and d) GHB affects the expression of IGF-1 receptors but not the plasma levels of IGF-1. In conclusion, the present work demonstrates that GH interacts with both opioid and GABAB-receptors in the male rat CNS and that GHB has an impact on brain regions associated with cognition and the development of dependence. These observations may be of relevance in many aspects related to addiction and might be translated into humans.

Individual differences in behavior, neurochemistry and pharmacology associated with voluntary alcohol intake

Alcohol use disorder is a worldwide public health problem and is a disorder with substantial individual variation. There are suggested links between various behavioral traits, comorbid psychiatric diseases and excessive alcohol consumption. Moreover, the endogenous opioid system is involved in alcohol reward and reinforcement, and implicated in the action of alcohol. However, less is known about the complex associations between individual differences in behavior, alcohol consumption, pharmacotherapy response and related neurochemical mechanisms. Experimental animal models are critical for understanding the neurobiological underpinnings of alcohol use disorder. The overall aims of this thesis were: i) to study the association between behavior and voluntary alcohol intake in outbred rats; ii) to study the association of voluntary alcohol intake, behavior, opioid receptor density and response to naltrexone; and iii) to obtain detailed behavioral characterizations of the animals on the basis of their voluntary alcohol intake. The results revealed that the multivariate concentric square fieldTM (MCSF) test was a complementary method for understanding mechanisms underlying various mental states. The MCSF broadened the perspective on risk-related behaviors, including aspects of risk assessment. Individual differences in alcohol intake using the modified intermittent access paradigm enabled analyses of drinking patterns in high and low alcohol-drinking rats. There was an alcohol deprivation effect in high-drinking animals only. The behavior profiling of high alcohol drinking- rats before and after alcohol access suggested that this subgroup was consuming alcohol for its anxiolytic properties. Long-lasting changes were found in the mu and the delta opioid receptors after long-term, intermittent voluntary alcohol intake; some of these changes are in line with findings in humans. The voluntary alcohol consumption and the concomitant response to naltrexone were different for Wistar rats from different suppliers. Moreover, the Rcc Wistar rats may be more suitable for studies of alcohol use disorders due to increasing alcohol intake and the presence of a high-drinking subpopulation with increasing alcohol intake over time. The high-drinking subpopulation showed pronounced effects of naltrexone on alcohol intake. In conclusion, studies of individual differences increase understanding of variability in behavior, pharmacotherapy response and factors involved in vulnerability of alcohol use disorders.

Reversal of Morphine-induced Locomotion in M5 Muscarinic Receptor Knockout Mice with Food Deprivation but not Bilateral Infusions of VTA BDNF

Cholinergic inputs from mesopontine tegmentum activate midbrain dopamine (DA) neurons via M5 muscarinic receptors. The M5 receptor is important for mesopontine stimulation-induced accumbal or striatal DA efflux, brain stimulation reward or morphine-induced conditioned place preference (CPP). M5 receptor knockout (KO) mice show 40-50% less morphine-induced locomotion. Pedunculopontine tegmental nucleus (PPT) lesions in rodents block morphine CPP, but are ineffective after 18 hours food deprivation, opiate dependence, or intra-VTA BDNF. Based on these findings, we investigated whether acute food deprivation or intra-VTA BDNF alters morphine-induced locomotion (3 and 10 mg/kg, i.p.) in C57BL/6 M5 KO mice. Non-deprived M5 KOs showed reduced morphine-induced locomotion, suggesting M5 receptors partly mediate morphine-induced locomotion. Morphine-induced locomotion was reversed in food-deprived mice, suggesting the stimulant effects of morphine were altered to bypass the PPT. Unexpectedly, intra-VTA BDNF infusions were ineffective in altering morphine-induced locomotion. Additionally, M5 KOs receiving intra-VTA saline showed no deficits in morphine-induced locomotion.

O objetivo do presente estudo foi avaliar se ratos tratados com chá de ayahuasca apresentam i) alterações comportamentais no campo aberto e labirinto em cruz elevado (LCE); ii) alterações na expressão do transportador de glutamato (EAAC1) no córtex pré-frontal (CPF) e no hilus do giro denteado do hipocampo (HDG). Doze grupos de ratos Wistar machos (250g, n=10/cada) foram usados. Eles receberam 2 or 4ml/Kg de chá de ayahuasca ou água: dose única (agudo), 3 vezes/dia por 3 dias alternados (subcrônico) e 1 vez/dia por 15 dias (crônico). Trinta minutos após a última ingestão os animais foram submetidos aos testes comportamentais. Vinte e quatro horas após eles foram anestesiados, perfundidos e seus encéfalos seccionados (40-m) no hipocampo e CPF para os experimentos de imunoistoquimica para EAAC1. Comparações estatísticas entre cada grupo tratado com ayahuasca e seu respectivo controle foram feitas utilizando o teste t de Student e consideradas significantes quando p0,05. Apenas a ingestão subcrônica de ayahuasca induziu redução significante na atividade locomotora (27%) no campo aberto. No LCE nenhum dos tratamentos com ayahuasca induziu alterações significantes em ambos numero de entradas e tempo de permanência nos braços abertos. A ingestão subcrônica ou crônica de chá de ayahuasca induziu aumento significante na expressão de EAAC1 no HGD (20-67%). Em contraste, no CPF a expressão de EAAC1 foi significantemente reduzida em ratos tratados com 2 ou 4ml/Kg subcronicamente ou 4ml/Kg cronicamente (17-25%). A ingestão aguda de 2ml/Kg induziu discreto aumento na expressão de EAAC1 (16%). Estes resultados sugerem que i) Ayahuasca induz alterações nas atividades locomotora e exploratória de forma dependente da dose e frequência de ingestão; ii) Ayahuasca não tem efeito no nível de ansiedade; iii) A ingestão aguda, subcrônica e subcrônica de ayahuasca disparam distintos mecanismos no hipocampo e CPF envolvendo a modulação da neurotransmissão glutamatérgica. / This work aimed at investigating whether rats treated with Ayahuasca beverage show i) behavioral alterations in the open field and elevated plus maze (EPM); ii) alterations in the expression of glutamate transporter (EAAC1) in the prefrontal cortex (PFC) and in the hilus of dentate gyrus of the hippocampus (HDG). Twelve groups of male Wistar rats (250g, n=10/each) were used. They received 2ml/Kg or 4ml/Kg of ayauhasca beverage or water: only once (acute), 3 times/day for 3 days (sub-chronic) or once/day for 15 days (chronic). Thirty minutes after the last ingestion the animals were submitted to behavioural tests. After 24 hours they were anaesthetized, perfused and their brains sectioned (40-m) in the hippocampus and PFC for immunohistochemistry (IH) detection of EAAC1. Comparisons between ayahuasca and control groups used Student t test. Significance was set at p0.05. Only sub-chronic ingestion of Ayahuasca induced a decrease in locomotor (27%) activit in the open field. On the EPM all treatments with Ayahuasca induced no significant increase in both number of entries and time spent in the open arms. The sub-chronic and chronic treatments with Ayahuasca induced a significant increase in EAAC1 expression in the HDG (20-67%). In contrast, in the PFC the expression of EAAC1 was significantly decreased in rats treated with 2 or 4ml/Kg sub-chronically or 4ml/Kg chronically (17-25%). Acute ingestion of 2ml/Kg induced a smaller increase in EAAC1-IC (16%). These results suggest that i) Ayahuasca changes the locomotor and exploratory activities in a way depending the dose and frequency of ingestion; ii) Ayahuasca does not have effect on the level of anxiety; iii) Acute, sub-chronic or chronic ingestion of Ayahuasca beverage trigger distinct mechanisms in the PFC and hippocampus involving the modulation of glutamate neurotransmission.

Um estudo do comportamento social de duplas de ratos (Rattus norvegicus) / A study of social behavior in pairs of rats (Rattus norvegicus)

Uma das dificuldades de se estudar o comportamento social é que para que ele ocorra são necessários pelo menos dois sujeitos, o que dificulta medidas de um deles, sem que seja necessário levar em conta o comportamento do outro. O presente trabalho investiga o comportamento social de duplas de ratos separados por uma grade, para minimizar o efeito que o comportamento de um rato possa ter sobre o outro. Como equipamento, foi utilizado um campo aberto de madeira (120 x 120 x 40 cm) forrado de fórmica marrom escuro. Uma das paredes podia estar intacta ou apresentar uma abertura por onde se podia acoplar uma gaiola de pássaros (34 x 22 x 26 cm). Todas as sessões se iniciavam pela colocação de um rato no centro do campo aberto e da gravação de seu comportamento por 10 minutos (exceto quando mencionado diferente) por uma câmara de vídeo. Para o registro, o piso do campo aberto na tela da TV foi dividido em 36 quadrados de 20 cm. Para cada quadrado, foram registradas a frequência e a duração dos seguintes comportamentos: (1) entradas e tempo gasto nos quadrados do campo aberto (compondo as áreas: quadrado da gaiola, cantos, periferia e centro), (2) frequência e tempo gasto farejando, limpando-se, levantando-se, esticando-se, e roendo a grade da gaiola. Esse procedimento foi aplicado a sete experimentos: (1) comparação com outro teste da literatura (File e Hyde, 1978, registrado segundo os autores), no qual o comportamento social dos dois animais se confunde, (2) comparação do comportamento social de machos e fêmeas no campo aberto sem gaiola, com a gaiola vazia e ocupada por um co-específico, (3) habituação ao aparato antes do teste com o co-específico, (4) estabilidade das medidas registradas em sessões repetidas, (5) efeito da iluminação, (6) efeito de tratamento com clordiazepóxido, e (7) efeito da duração da sessão. Os resultados do primeiro experimento indicaram que o teste da literatura correlacionou-se muito pouco com suas próprias medidas e com as medidas do teste proposto, enquanto o teste proposto mostrou um grande numero de correlações entre si. O principal achado foi o de que os animais testados com a presença do co-específico na gaiola alocaram uma maior quantidade de tempo e executaram mais comportamentos na área da gaiola. O segundo experimento mostrou que a ocupação da área da gaiola depende da presença do co-específico, sendo menor (e menos frequentes os comportamentos) quando a gaiola estava vazia e menor ainda quando não havia gaiola. O terceiro experimento mostrou que a pré-exposição ao campo aberto com a gaiola vazia não alterou o tempo gasto na área defronte à gaiola em comparação com uma segunda sessão com o co-específico presente. O quarto experimento mostrou que submeter os ratos a cinco sessões sucessivas também não alterou o comportamento social dirigido ao co-específico. O quinto experimento mostrou que ratos testados no claro ou no escuro interagem com o co-específico de modo semelhante. O sexto experimento apresenta dados mostrando que o comportamento social aumentou com a administração de 3,0 e 5,6 mg/Kg de clordiazepóxido. Finalmente, o sétimo experimento mostrou que o comportamento social se altera muito pouco quando se compara o comportamento de ratos em sessões com 10 ou com 30 minutos de duração. De um modo geral, os dados demonstraram que o modelo proposto permite o estudo de duplas de ratos com foco no registro individual de um rato-alvo, sem que seja demasiadamente influenciado pelo co-específico, é estável em sucessivas sessões ou sessões com durações diferentes, é sensível à presença do co-específico, do sexo do animal-alvo e sensível a um tratamento farmacológico. Uma característica importante é que, ao contrário da literatura (File e Hyde, 1978) não é necessário isolar o(s) animal(is). / One of the difficulties of studying social behavior is that for it to occur at least two individuals are necessary, which makes it difficult taking measures of one of them without taking the behavior of the other into consideration. The present work investigated the social behavior of pairs of rats separated by a grid in order to minimize the effect the behavior of one rat may have upon the other. A wood open field (120 x 120 x 40 cm) lined with dark brown Formica was used. One of the walls could be intact or present an opening through which a bird cage (34 x 22 x 26 cm) could be fixed. All sessions started by placing a rat in the center of the open field and recording its behavior for 10 minutes (except when stated otherwise) with a video camera. For recording, the floor of the open field was divided into 36 20-cm squares on the TV screen. Frequency and duration of the following behaviors were recorded for each square: (1) entries and time spent in each square of the open field (grouped in the areas: cage square, corners, periphery and center), (2) frequency and time spent sniffing, grooming, rearing, stretching and gnowing the grid. This procedure was used in seven experiments: (1)comparison with a literature test (File and Hyde, 1978), in which social behavior of a pair of rats is mixed, (2) comparison of the social behavior of males and females in the open field without the cage, with an empty cage and with the cage with a co-specific, (3), habituation to the apparatus before the test with the co-specific present, (4) stability of the recorde measures along repeated sessions, (5) the influence of illumination during the test, (6) the effect of chlordiazepoxyde treatment, and (7) the effect of session duration. The results of the first experiment indicated that the measurements of the literature test did not correlate well, while the porposed test exhibited a large number of correlations. The main finding was that the rats tested with the co-specific in the cage allocated a larger amount of time to the cage square. The second experiment showed that the occupation of the cage square depends on the presence of the co-specific, and is smaller (as are less frequent the behaviors) when the cage was empty and even smaller when there was no cage. The third experiment showed the pre-exposure to the open field with an empty cage did not alter the time spent in the cage square in a second session with the co-specific present. The fourth experiment showed that submitting the rats to five daily sessions also did not alter the social behavior directed towards the co-specific. The fifth experiment showed that rats tested either in the light or in the dark interact with the co-specific in the same manner. The sixth experiment presents data showing that social behavior increased with the administration of 3.0 and 5.6 mg/Kg chlordiazepoxyde. Finally, the seventh experiment showed that social behavior alters very little when 10-min sessions are compared to 30-min sessions. In general, the data showed that the proposed model allows the study of pairs of rats focusing in the individual record of a target rat without its being too much influenced by the co-specific, is stable in successive sessions or sessions with different durations, is sensitive to the presence of a co-specific, to the gender of the target rat, and to a pharmacological treatment. An important characteristic is that, unlike the literature test (File and Hyde, 1978) it is not necessary to isolate the animals(s).

Este produto está relacionado a diversos efeitos controversos, podendo promover a cura de sintomas patológicos do aparelho digestório e respiratório ou causar efeitos psicoativos por consumo abusivo. No preparo de alimentos é empregada por seu sabor picante e caráter conservador (antimicrobiano e antioxidante). Este trabalho teve como objetivo caracterizar físico-químicamente a semente, enfatizando a determinação de miristicina, principal composto ao qual se atribui os efeitos danosos, e estudar o efeito de infusões consumidas cronicamente no comportamento de animais de laboratório. Para desenvolvimento do trabalho foram coletadas amostras de noz-moscada comercializadas na forma de semente e pó em duas regiões do país. Para caracterização físico-química foram adotados os procedimentos descritos pela AOAC (2000) para determinação de umidade, proteína, extrato etéreo, cinza e demais componentes por diferença. Foi adaptada uma metodologia para determinar miristicina em cromatografia gasosa. Para os seis lotes avaliados foram encontrados valores médios de 9,5%(+/-1,5); 2,1%(+/-0,5); 8,5%(+/-2,5); 31,9%(+/- 9,3) e 49,9 (+/-10,6) respectivamente para umidade, proteína, extrato etéreo e outros componentes. A miristicina foi determinada no extrato hidroalcoólico das sementes; da fração lipídica extraída a frio e da infusão. Os teores encontrados variaram entre 6,6 a 14 mg/g de semente no extrato hidroalcoólico e na infusão preparada, o que sugere que o maior risco de dano crônico de miristicina esta associado ao consumo de chás para diferentes fins. Numa segunda etapa foram estudados os efeitos crônicos do extrato aquoso, preparado com 0,5; 1,0 e 2,0% (p/v) de noz-moscada. Estas infusões das sementes foram avaliadas quanto a sua atividade psicoativa, administrando-se oralmente, ad libitum, durante 90 dias, aos animais. Os grupos tratados exibiram atividade ansiogênica no teste do labirinto em cruz elevado (LCE) e no teste de campo aberto (CA). Estes resultados sugerem e corroboram com efeitos centrais atribuídos ao consumo abusivo do vegetal. / Nutmeg, Myristica fragans, Houtt., is a specialty used to cook and to prepare and healing teas since the most remote times and more recently it started to be used in pharmaceutical and the food industry. This product is involved by controversial effects, since healing pathological symptoms of the stomach and the respiratory system or causing psychoactivity alteration due to it’s over use. Nutmeg is used as a spice due to it’s hot flavor and conservative characteristics (antimicrobiotic and antioxidant). This paper has as a objective characterizing physic-chemically the seed, giving emphasis to the determination of miristicyn, compound that causes the biggest damages, and studying its effects in lab animals with a diet rich in miristicyn. To develop the work samples of nutmeg seeds or powder where collected from two different regions of the country. For the physic-chemical characterization two procedures described by AOAC (2000) were adopted for the determination of humidity, protein, eter extract, ash and other compounds by difference. A methodology was adapted to determinate miristicyn in gas chromatography. The medium values for the six samples where; 9,5%(+/- 1,5) humidity, 2,1%(+/-2,5) ash, 8,5%(+/-2,5) protein,31,9%(+/-9,3) eter extract and 49,9%(+/- 10,6) of other compounds. The miristicyn was determined in hydroalcoholic extract from the seeds; from the cold lipid extracted portion and from the effusion. The found levels ranged from 6,6 to 14 mg/g seed in the prepared effusion hydroalcoholic extract, witch suggests a higher chronic damage to be associated to the use of multipurpose teas. In a second stage the chronic effects of a liquid solution prepared with 1,0 to 2,0% (p/v) of nutmeg. The seed effusions were evaluated for its psycho activity when ingested orally ad libitum trough 90 days in animals. The treated groups showed ansiogenic activity in the elevated plus maze (EPM) and in the open field test (OF). This results suggest and agree with central effects attributed to the abusive consume of this product.
