This thesis is based on physical properties of fish meat and fish products. Texture of fish meat is considered as one of the most perceived character. Major influence on the textural properties of fish meat has chemical composition of fish meat and fish meat structure. Fish products are changed during production by influence of low temperature during frozen storage and influence of high temperature during cooking. Changes in the textural properties of fish muscle caused during the autolytic process after slaughtering. The aim of the thesis was compared different methods for measurement textural properties of fish and fish products. Surimi sticks and pre-fried fish sticks were tested. Common carp (Cyprinus carpio), Rainbow trout (Oncorhynchus mykiss) and Tench (Tinca tinca) were chosen for testing by penetration test (penetration of the probe into the sample) and Warner-Bratzler shear test (sample cutting by Warner-Bratzler shear).

Změny texturních vlastností hovězího masa v průběhu zrání

This diploma thesis is focused on textural properties of beef meat during aging. Round and sirloin from three heifers of Czech Fleckvieh breed were used for the experiment. Samples were analyzed by Warner-Bratzler test, compression test and penetration test in 4th, 6th, 7th and 8th week of aging. Results of Warner-Bratzler test ranged from 54,72 – 59,96 N and there was not found statistically significant difference. Results from compression test TPA1 were in range 187,75 – 277,05 N and TPA2 results were in range 153,74 – 225,55 N and there was found statistically highly significant difference in most cases. Penetration test results of beef round were in range 5,58 – 6,18 N and there was not found statistically significant difference. Penetration test results of beef sirloin were in range 6,39 – 9,49 N and the difference was not statistically significant in most cases.

Katepsiny L cerkárií Diplostomum pseudospathaceum / Cathepsins L of Diplostomum pseudospathaceum cercariae

This study is focused on cercarial cysteine peptidases of the trematode Diplostomum pseudospathaceum. It follows previous research which confirmed the presence of a 24kDa cysteine peptidase in cercariae biochemically and by mass spectrometry. It was postulated, that the function of this peptidase is histolytic, when cercariae penetrate the tissues. During an attempt to purify this peptidase and characterize its peptidolytic activity, it was found out that the cercarial homogenate containsmore different peptidases varying in their pI. Tests of peptidolytic activity and inhibition have shown that these peptidases are cathepsin L-like. They are active over a broad spectrum of pH with optima of activities in weakly acidicor neutral pH. Using degenerate primers based on conserved motifs of cysteine pepridases, partial sequences of three genes for cathepsin L of D. pseudospataceum (DpCL1, 2 a 3) were obtained. Then the complete sequences of DpCL2 and 3 genes and partial sequence (without 5'end) of DpCL1 were obtained by RACE PCR. To confirm function of these peptidases we tried to immunolocalize them. IMQ is currently used in clinical practice in the form of cream Aldara® containing 5% of active substance, which is unstable and irritating and after removal from the skin IMQ poses an ecological load for the environment. The aim of this thesis was preparation of liposomes for topical administrativ containing lower - 1% amount of IMQ and evaluation of penetration of IMQ into human skin in vitro. To improve the properties of liposomes and promote patency of the active ingredient through the skin barrier to the deeper skin layers various additives were used. This diploma thesis deals with the creation of penetration of cast iron with graphite casting cast into bentonite molds. In the first part of the thesis is theoretically described the formation of penetration, its main causes, consequences and the elimination of this defect. The second part analyzes the causes of the defect with the Minitab statistical software and shows the possible connections of the input materials to the occurrence of the defect. Next, the experimental finding of the cause of the defect, which is focused on the measurement of the pressures in the form of the casting, is devoted to it.

The main subject of this thesis is research work of scholarly articles focused on the penetration of resins and polymers into the cell wall of the wood. In contains a description of the principles on which the used methods are based, and a summary of the results achieved. In the experiment, several method were used to investigate the possible penetration of selected methods adhesives into the structure of the cell wall. These are fluorescence microscopy, confocal microscopy, scannin electron microscopy (SEM) and transmission electron microscopy (TEM).

This work deals with the transmittance of a family of non-steroidal anti-inflammatory drugs across biological membranes in combination with the use of hyaluronan. Hyaluronic acid (hyaluronan, HYA) is a linear polysaccharide formed from disacharide units containing N-acetyl-D-glucosamine and glucuronic acid. HYA is present in almost all biological fluids and tissues, so there is an assumption that could affect the penetration of substances through the skin. Standards were prepared by anti-inflammatory drugs in admixture with various concentrations hyaluronan and subsequently measured transmittance of such substances through the skin. Standards were applied to the skin obtained from pig auricle. The drug content was determined by HPLC.

Trichobilharzia spp. are avian schistosomes related to medically important human parasites of the genus Schistosoma. Penetrating cercariae are well known as causative agent of cercarial dermatitis in humans. Cercariae actively penetrate the skin of definitive hosts and transform into schistosomula. This process is preceded by cercarial tail detachment and includes emptying of penetration glands and extensive surface changes. One of these changes is the loss of highly immunogenic glycocalyx which represents a protective coat in the aquatic environment. The glycocalyx has specific composition of saccharide molecules which are bound to lipids or proteins on the membrane of cercarial tegument. There is only limited information about the mechanism of shedding. Hypotheses based on indirect evidences suggest that peptidases or (phospho)lipases from penetration glands could be involved. This work describes the changes in surface glycosylation during transformation of cercariae into schistosomula by fluorescently labelled lectins and monoclonal antibodies against Lewis X antigen. Lectins UEA-I, LTA and PNA have been chosen as markers of transformation of T. regenti. Further, our experiments have been focused on shedding of cercarial glycocalyx. The practical part deals with the RTFOT aging and with the influence of the duration of this test. The comparison of test results is conducted before and after the aging by values of needle penetration, elastic recovery, softening point and resilience tests are evaluated and compared.
