Mikroskopie pomalými elektrony ve studiu složitých krystalických struktur / STUDY OF COMPLICATED CRYSTAL STRUCTURES BY MEANS OF SLOW ELECTRONS

Methods for examination of the crystal structure of crystalline materials include the X-Ray, neutron and synchrotron-radiation diffraction, electron backscattered diffraction in the scanning electron microscope, scanning transmission electron microscopy, transmission electron microscopy and focused ion beam microscopy. The scanning low energy electron microscopy (SLEEM) is less known as yet but already has proven itself very powerful tool for studies of the crystal lattice. By means of very slow electrons reflected from the sample and effectively detected in their full angular and energy distribution the crystalline structure is imaged at high spatial resolution and high contrast is obtained between differently oriented grains in polycrystals. Because of high sensitivity of the image signal to the inner potential distribution in the sample even details like subgrains or twins as well as strain at the microstructural level can be visualized. The aim of this thesis is to demonstrate the scanning low energy electron microscopy as an effective tool for investigation of wide range of materials like steels, non-ferrous alloys and ultra-fine grained materials.

Pokročilé oxidační procesy v environmentálních aplikacích / Advanced oxidation processes in environmental applications

The aim of this thesis is the study of application of the synergistic effects of various advanced oxidation processes (AOP) used for decomposition of chemical compounds resistant to biodegradation. The synergistic effect of photocatalytically active material and low temperature plasma were tested in my thesis. Model substances with photocatalytically active material were exposed to atmospheric plasma discharge (Gliding Arc) in order to improve generation of the active hydroxyl groups and oxidation processes. The first chapter of the theoretical part summarizes the knowledge of advanced oxidation processes, their principles and utilization. Next chapters are dedicated to basic summary of titanium dioxide and types of creating the thin films. The last part of theoretical chapter describes the types of plasma discharges. The experimental part describes the equipment for decomposition of chemical compounds resistant to biodegradation using AOP. Next chapter of the experimental part is dedicated to decomposition of organic dye AO7 and antibiotic Verapamil hydrochloride. The last chapter summarizes the results of the experiments.

Arterial system is a system of vessels distributing blood. Ageing of arterial system leads to two distinct pathologies: atherosclerosis and arteriosclerosis - stiffening of arterial wall. These pathologies can coexist and interfere; however, they differ in their pathogenesis, location, scope and consequences. Progressive loss of elastic properties of large arteries is natural part of vascular ageing. It is directly responsible for several age dependent consequences, such as increase of central systolic pressure or prevalence of isolated systolic hypertension in the elderly. Clinically, central arteries stiffness manifests as aortic pulse wave velocity, which can be quantified, among other methods, using applanation tonometry. There is abundant evidence that aortic pulse wave velocity represents an independent predictor of cardiovascular mortality and morbidity. The most important mechanism in arterial stiffening is repeated mechanical damage which leads to fractures, fragmentation and thinning of elastin. Stiffening of large arteries can be accelerated by several other mechanisms, e.g. deposition of several substances (calcium, advanced glycation end-products, etc.), metabolic turnover of key elements of vascular extracellular matrix (collagen and elastin) or individual genetic susceptibility. Methods: The substances were tested using quantitative microdilution broth method on eight strains of yeast and fibrous fungi. These were Candida albicans, Candida krusei, Candida parapsilosis, Candida tropicalis, Aspergillus fumigatus, Aspergillus flavus, Absidia corymbifera and Trichophyton interdigitale. Results: The highest antimycotic activity was demonstrated by compound Ac-0a, where we recorded after 24 hours of incubation activity against all eight strains. However, it was not able to stop the growth of three strains after 48 hours of incubation, the strains concerned were Candida parapsilosis, Aspergillus fumigatus and Aspergillus flavus. Recently, an increasing problem in wastewater treatment is the insufficient removal of organic pollutants. These substances can be toxic to the environment already in a small amount, either acutely or chronically. The goal is therefore to develop of technologies that ensure their effective removal. One possibility is to use advanced oxidation processes. Advanced oxidation processes work on the principle of non-selective oxidation mediated by OH radicals. Several methods such as O3/H2O2 (Peroxone), UV/H2O2 or Fenton´s reaction can be used to generate them. The aim of this thesis was to evaluate the effectiveness of the used advanced oxidation processes in wastewater treatment via ekotoxicity tests. Wastewater samples treated with advanced oxidation processes on the pilot unit showed low values of acute toxicity for selected test organisms (D. magna, T. platyurus, S. alba, L. minor). For the sample of waste water that was treated with the UV/H2O2 on AOP laboratory unit, there was an increase in acute toxicity on the testing organism D. magna and L. minor. The similar effect was observer in the tests with the D. magna with the model samples treated with the same method, while the tests on the L. minor showed a reduction in toxicity.

Diagnostika herního výkonu statistickou analýzou v ledním hokeji / Diagnosis of game performance by statistical analysis in ice hockey

Aim of the thesis The aim of this thesis is to show usage of analysis of basic and advanced statistics to evaluate the game performance in ice-hockey. Using this analysis can be more accurate because it is based on facts. This statement is compared with evaluation of the game performance by the experts that used direct observing method. The main aim of this thesis is to bring this idea to the Czech ice-hockey community which can be helpful to improve the club hockey in Czech Republic or improve Czech ice-hockey itself. Methods of thesis The method of analysis of statistical data to evaluate the game performance is used in this diploma thesis. Statistical data were acquired by indirect watching of video records of the games. This analysis is compared with method of direct observing of the game to evaluate the game performance which was used by experts. Conclusion By analyzing current situation in evaluating the game performance it was found out that many teams are still using method of direct observing to evaluate the game performance. In practical part of the thesis the experts after using method of direct observing of the game came up with different conclusions. Pokročilé programové konstrukce a programovací techniky jazyka Swift / Advanced programming constructs and techniques in the Swift language

The thesis focuses on advanced programming constructs and techniques of the Swift programming language. At the beginning programming paradigms that are supported by the Swift design (object-oriented and functional) are introduced. Concepts of each paradigm are described and Swift is then evaluated by the level of support for these paradigms. Following programming constructs are described in-depth: working with variables, optional types, enumerated types, functions and closures. Creating of custom operators and subscripts. Operations with protocols, generic types and parallel program execution. Error handling with throwing functions. Detailed explanation of value types and their differences from reference type is given too. Thesis also explains nested types and all higher-order functions in Swift. At the end are described extensions; specifically extension of computed property, initialization function, function, subscript, nested type and protocol and generic types extensions. In the last section of the thesis these advanced programming techniques are described; function chaining and composition, structure saving, performance optimization of value types, reflection in the Swift programming language and how modules and namespaces work. At the end protocol-oriented programming is introduced.

Odstranění vybraných organických polutantů z vody s využitím pokročilých oxidačních procesů / Removal of selected organic pollutants from water using advanced oxidation processes

The aim of this diploma thesis is the determination of selected organic pollutants with a focus on artificial sweeteners in waste water from wastewater treatment plant (WWTP) Modřice and the subsequent degradation of the thus obtained concentrations in model water using advanced oxidation processes (AOP). Two sweeteners were determined, namely sucralose and saccharin found in waste water. Synthetic sweeteners are not sufficiently cleaned in the WWTP and penetrate the environment. Effective processes for removing organic substances, including alternative sweeteners, are AOP processes. These processes are based on reactions of highly reactive hydroxyl radical (OH·), which is produced by many chemical reactions. Generally, they use ozone (O3) and hydrogen peroxide (H2O2). On sampling, wastewater samples were treated with Solid Phase Extraction (SPE) and then evaluated by HPLC/MS (High Performance Liquid Chromatography). In observing the degradation of selected sweeteners, three AOP methods were used: O3/UV, H2O2/UV and O3/H2O2. For each of these methods, the efficiency of degradation of sucralose and saccharin was investigated. From the evaluated data it was found that the most efficient AOP process for removing artificial sweeteners from water is the O3/H2O2 method.
