This diploma work describes educational trial in small village school in the context of inclusive education. It is focused on teaching united classes and teaching pupils with special educational needs. Theoretical part contains characteristic of small village schools, describes their historical evolution, typical features of teaching and position assistant of teacher in these schools. In the following subsections are determined special learning disabilities, theirs causes of origin and regulars pedagogical intervention. And then is focused on theme of inclusive education. Targets of practical part were to find experiences of teachers at small village schools, to choose ideal didactical methods and organizational forms, which are suitable for differentiation and individualization in the teaching, after that to make a proposal of methodical support, that to verify in practise and to evaluate its. Congenital disorders of glycosylation (CDG) represent a rapidly growing group of rare inherited metabolic diseases with estimated prevalence as high as 1:20 000, which are caused by genetic defects that impair the process of glycosylation, i.e. the enzymatic addition of a specific saccharide structure onto a protein or lipid backbone. Due to non-specificity and variability of clinical symptoms in the patients, the medical diagnosis of CDG remains extremely challenging and significantly relies on accurate biochemical and genetic analyses. The overall goal of the present dissertation thesis was to study CDG at the biochemical and molecular genetic level in the context of the Czech and Slovak Republic, which involved three specific aims: A.) to introduce and optimize laboratory screening methods for CDG detection in a group of clinically suspected patients, B.) to determine the corresponding genetic defect in the positive patients selected via CDG screening and to study the pathobiochemical aspects of specific CDG types at the cellular level, and C.) to analyze glycosylation disturbances of non- CDG etiology. This diploma thesis deals with functional disorders of the musculoskeletal system. It discusses the possible causing factors of these disorders, their manifestations on the body, diagnostics and the possibilities of influencing them. Since this is a very common phenomenon on the human body, I chose mainly the muscle, fascia and joint structure, which I explored and described in more depth. The experimental part of this work aims to demonstrate the possible impact of the intervention of the 1.-3. ribs and claviculosternal joint as described by Ludmila Mojžíšová on the muscles of the neck (m. scalenus anterior, medius and sternocleidomastoideus). To obtain results, I used the ultrasound examination of the diameter of these muscles and Visual analogue scale (VAS) to obtain pain values before and after the intervention using this method. The thesis focuses on female university students and their body image in relation with their parents' body image. The satisfaction of the students with their own bodies depends on many elements such as the influence of the media and the judgements of their peers, family and authorities. A substantial part of our thesis is an unprecedented analysis of the possibility of a trans-generation transfer of the extent of satisfaction with one's own body from parents to their daughters. We have used the quantitative research strategy to assess the results gathered from questionnaires focusing on the students' as well as the parents of both male and female children's perception of their own bodily features. Physical parameters such as height and weight were also checked. The study also focused on whether the trans- generation transfer is affected by the way the parents treat their children during childhood and adolescence. Our results have shown that there is a positive relation between the student's and the mother's body image. This positive relation can be perceived as a consequence of the trans-generation transfer which is probably a mechanism of observational learning. The diploma thesis deals with the education of pupils with specific learning disabilities. The theoretical part deals with the definitions of specific learning disabilities, the needs of students and the possibilities of re-education of individual disorders. There are defined degrees and types of support, legislation that regulates the education of students with special needs, but also other opportunities for their education. The practical part deals with case studies or observations of students with SPU, then a questionnaire survey, which seeks to answer the question of how teachers worked with students with SPU at the time of distance learning, whether they succeeded educational goals and also with what difficulties pupils struggled. The questionnaire survey is supplemented by structured interviews with teachers who participated in the education of students during distance learning.

This diploma thesis deals with the topic of metaphorization of autism spectrum disorders (ASD) and their representation in common online media. The aim of the diploma thesis is to examine aspects of the relationship between metaphors and autism spectrum disorder. In terms of metaphor theory, it is based on cognitive linguistics and its approach to conceptual metaphor. The thesis lists the most common metaphors that are used in connection with autism spectrum disorders whereas the distinction between neurotypical and autistic metaphors is made. Then, it deals with medialization of autism spectrum disorders and presents possibilities of categorization of the way ASD is written about in common online media. Finally, the thesis discusses the overall problematics of metaphorization of autism spectrum disorders and possibilities of changes in the way they are represented.

This diploma thesis deals with the issue of mental anorexia (AN), clinical and laboratory manifestations of the disease. The theoretical part is focused on summarizing the existing knowledge about the diagnosis, complications and treatment of AN. Attention is paid to the effects of the disease on individual organ systems and clinical and laboratory consequences. The practical part deals with the evaluation of laboratory parameters and clinical symptoms of patients monitored at the Department of Pediatrics and Inherited Metabolic Disorders, 1st Faculty of Medicine, Charles University and General Hospital in Prague. The performed clinical examinations include anthropometric measurements, bioimpedance, measurement of resting energy consumption by indirect calorimetry, measurement of skin algae with a caliper and comparison of the results of methods for determining the proportion of adipose tissue using a caliper and bioimpedances. All patients underwent EKG and cardiological examination, blood pressure measurement, heart rate. This thesis deals with diagnostic methods for personality disorders - a questionnaire LPFS-BF 2.0 and a semi-structured interview STiP-5.1. Both methods assess the level of personality functioning from The DSM-5 Alternative Model for Personality Disorders. The main aim of that study is to compare a self-report method and a method based on clinical assessment on a sample of N = 30 non-psychiatric group and N = 30 clinical group. Diagnostic criteria for personality disorders in accordance with valid classification systems are presented in the theoretical part of the study, with an emphasis on benefits and limitations of the DSM-5 alternative model. Categorial and dimensional diagnostic approaches were compared and critically evaluated. Existing diagnostic methods for personality disorders were then discussed, structured interviews and questionnaires were analyzed more in detail. Specifics in use of self- report methods and structured interviews for assessing personality disorders were also discussed. The thesis deals with Autistic Spectrum Disorders in terms of their ethology and diagnostic criteria. The theoretical part further describes development of terminology of these disorders and historical concept of care of individuals with Autism. Included are also categories of disabilities and their symptoms. Another chapter discusses the specifics of self-help skills of children with Autism and characterizes motor development of preschool children with an emphasis on motor imitation, as one of the sub-kinetic abilities. The aim of the research work is to analyse the degree of influence of motor imitation on the development of self-help skills of children with Autism. It focuses on evaluation of the developmental level of the respondent's self-help skills and its development influenced by intensive imitation training. Determination of the actual developmental level allows easier orientation within self-help skills that can be practiced with the child. Their list is located in the attachments. Further research clarifies what techniques were the most successful during motor imitation training.
