Model za kontrolu površinske uniformnosti digitalnih otisaka / Print uniformity model for digital prints

<p>Površinska uniformnost je jedan od ključnih parametara za analizu<br />kvaliteta otisaka. Osnovni cilj istraživanja je definisanje modela<br />za kvantifikaciju površinske uniformnosti digitalno štampanih<br />otisaka. U nizu sprovedenih eksperimenata procenjen je uticaj ulaznog<br />uređaja za digitalizaciju otisaka i uticaj varijabilnih podešavanja<br />prilikom digitalizacije. U disertaciji su ispitane četiri metode za<br />merenje površinske uniformnosti koje su najčešće korišćene u<br />dosadašnjoj literaturi &ndash; GLCM, ISO 13660, Integralna i M-Score<br />metoda. Na osnovu dobijenih rezultata i zaključaka istraživanja<br />moguće je predložiti razvoj industrijski primenljivog rešenja za<br />merenje površinske uniformnosti digitalno štampanih otisaka.</p> / <p>Print uniformity is one of the key parameters for the analysis of print<br />quality.The basic aim of the research is to define a model for the<br />quantification of print uniformity of digitally printed samples. In the series of<br />carried out experiments, the influence of the input device for digitalisation of<br />prints and the influence of variable settings during digitization was evaluated.<br />The dissertation examined four methods for measuring the surface uniformity<br />most commonly used in the previous literature - GLCM, ISO 13660, Integral<br />and M-Score method. On the basis of the obtained results and conclusions of<br />the research, it is possible to propose the development of an industrially<br />applicable solution for measuring the surface uniformity of digitally printed<br />prints.</p>

Razvoj modela objektivne kontrole površinskih oštećenja premaznih papira u procesu savijanja / Development of a model for objective control of surface damages on coated papers in the folding process

<p>U disertaciji se predstavljaju istraživanja koja su rezultirala razvojem modela objektivne kontrole otpornosti premaznih papira prema površinskom oštećenju u procesima savijanja. Na bazi analize niza izabranih parametara procesa kontrole, ppedložena su tri obeležja digitalnih uzoraka premaznih papira za opis i ocenu površinskog oštećenja. Rezultati predloženih obeležja, kao i korelacione analize omogućuju primenu tih obeležja u funkciji kontrole kvaliteta kao osnove razvoja objektivne procesne kontrole premaznih papira u procesu savijanja.</p> / <p>The research presented in this dissertation resulted in development of the objective quality control model for fold cracking resistance of coated papers. Based on the analysis of chosen control process parameters, three different features of the digitalised coated paper samples were proposed for describing and classifying surface damages. The results of the proposed features along with their correlation analysis contribute to their usage in objective process quality control of coated papers in the folding process.</p>

Adsorpciona i emulgujuća svojstva proteinskog izolata i hidrolizata semena tikve (Cucurbita pepo) / Adsorption and emulsifying properties of pumpkin (Cucurbita pepo) seed protein isolate and hydrolysate

<p>Seme tikve (Cucurbita pepo) obiluje kako uljem tako i proteinima. Nakon izdvajanja ulja, proteini se koncentri&scaron;u u uljanoj pogači, sekundarnom proizvodu procesa proizvodnje ulja, gde njihov sadrţaj dostiţe do 65%. Proteini semena tikve su primamljiv sastojak za prehrambenu, farmaceutsku i kozmetičku industriju zbog svoje farmakolo&scaron;ke aktivnosti i visoke biolo&scaron;ke vrednosti. Pored toga, budući da su mnogi proizvodi ovih industrija po svojoj koloidnoj prirodi emulzije, proteini semena tikve bi se u njima mogli naći i kao prirodne povr&scaron;inski aktivne materije. Međutim, koloidna funkcionalnost proteina semena tikve se jo&scaron; uvek potcenjuje zbog globularne strukture za koju se vezuju slabija funkcionalna svojstva u odnosu na proteine sa fleksibilnijom strukturom. Prema tome, cilj ove disertacije je ispitivanje funkcionalnih osobina proteinskog izolata semena tikve, pre svega njegovih adsorpcionih i emulgujućih svojstava, kao i ispitivanje uticaja promene proteinske strukture putem enzimske hidrolize na ispitivana svojstva.<br />Pripremljeni su izolat proteina semena tikve (IPST) i dva enzimska hidrolizata, H1 i H2. IPST, H1 i H2 su okarakterisani određivanjem sadržaja vlage, proteina i pepela, zatim, određivanjem prinosa, molekulske mase i zeta potencijala. Ispitan je uticaj koncentracije proteina/peptida (0,0001&ndash;1 g/100 cm<sup>3</sup>), pH (3&ndash;8) i jonske jačine (0&ndash;1 mol/dm<sup>3</sup> NaCl) na rastvorljivost i adsorpciona svojstva: dinamički međupovr&scaron;inski pritisak (ulje/voda), statički povr&scaron;inski (vazduh/voda) i međupovr&scaron;inski (ulje/voda) pritisak, kinetiku adsorpcije i dilatacionu reologiju proteinskih adsorpcionih filmova. Nakon toga, ispitan je i uticaj pomenutih parametara na emulziona svojstva IPST, H1 i H2. Emulgujuća svojstva IPST, H1 i H2 su okarakterisana na osnovu prosečnog prečnika kapljica emulzija, raspodele veličina kapljica i stabilnosti emulzija.<br />Utvrđeno je da je prinos IPST veći od prinosa oba hidrolizata za oko 65 %. IPST ima najniţu rastvorljivost na pH=5, &scaron;to ujedno predstavlja i njegovu izoelektričnu tačku. Enzimskom hidrolizom IPST značajno se povećava rastvorljivost, posebno na pI=5. Povećanje jonske jačine je izazvalo salting&ndash;in ili salting&ndash;out efekat rastvorljivosti kod svih uzoraka u zavisnosti od pH. IPST, H1 i H2 poseduju povr&scaron;insku aktivnost pri čemu je povr&scaron;inski/međupovr&scaron;inski pritisak H1 i H2 manje zavistan od promene pH i jonske jačine u poređenju sa povr&scaron;inskim/međupovr&scaron;inskim pritiskom IPST. Adsorpcijom na granicu faza IPST i oba hidrolizata obrazuju adsorpcione filmove sa dominantnom elastičnom komponentom. Emulgujuća svojstva IPST, H1 i H2 zavise od koncentracije uzorka, pH vrednosti i jonske jačine kontinualne faze. Pri koncentraciji od 1 g/100 cm<sup>3</sup> i Ic=0 mol/dm<sup>3</sup> pripremljene emulzije su stabilne na svim pH osim emulzije IPST na pH 5. Sve emulzije podležu gravitacionoj nestabilnosti.</p> / <p>Pumpkin (Cucurbita pepo) seed is rich source of both, oil and proteins. Once the oil has been extracted, proteins concentrate in oil cake, a by&ndash;product of the oil<br />extraction process, where their content can reach up to 65%. Pumpkin seed proteins are desirable ingredient in food, pharmaceutical and cosmetic industry due to their pharmacological activities and high biological value. Moreover, since many of products of these industries are, in colloidal terms, emulsions, pumpkin seed proteins could serve as surface active materies. However, colloidal functionality of pumpkin seed proteins is still underestimated for their globular structure which entails inferior functional properties to functional properties of proteins with more flexible structure. Based on that, the aim of this dissertation is to investigate functional properties of pumpkin seed protein isolate, adsorption and emulsifying properties, in the first place, and then to investigate the influence of modification of the protein structure, by means of enzymatic hydrolysis, on the aforementioned properties.<br />Pumpkin seed protein isolate, IPST, and two enzymatic hydrolysates, H1 and H2, were prepared. IPST, H1 and H2 were characterized by determination of moisture, ash and protein content, then, by determination of protein recovery, molecular mass and zeta potential. Influence of the protein/peptide concentration (0.0001&ndash;1 g/100 cm<sup>3</sup>), pH (3&ndash;8) i ionic strength (0&ndash;1 mol/dm<sup>3</sup> NaCl) on the solubility and adsorption properties: dynamic interfacial (oil/water) pressure, static surface (air/water) and interfacial (oil/water) pressure, adsorption kinetics and interfacial dilatational properties, was<br />investigated next. In the end, influence of the aforementioned pharameters on the emulsifying properties of IPST, H1 and H2 was investigated. Emulsifying properties of IPST, H1 and H2 were discussed in terms of mean droplet diameter, droplet size distribution and emulsion stability.<br />Protein recovery of IPST was determined to be 65 % higher than recovery of H1 and H2. Solubility of IPST was the lowest at pH 5, what presents the isoelectric point. The enzymatic hydrolysis of IPST significantly increased solubility, especialy at the isoelectric point. Increase in the ionic strenght led to salting&ndash;in or salting&ndash;out effect depending on pH of the sample. Three investigated samples, IPST, H1 and H2 exhibited surface activity, however, sufrace/interfacial pressure of H1 and H2 were found to be less influenced by change in pH or ionic strenght of the solution in comparison to the IPST. Once adsorbed to the interface IPST and both hydrolysates form interfacial film with dominant elastic component. Emulsifying properties of IPST, H1 and H2 depend on the concentration, pH and ionic strength of the continuous phase. Stabile emulsions were formed at concentration of 1 g/100 cm3 and Ic=0 mol/dm<sup>3</sup> regardless of pH, with the exception of the IPST at pH 5. All emulsions were susceptibile to gravitational separation.</p>

Ispitivanje mehaničkih i površinskih svojstava stomatoloških nanostrukturisanih kompozitnih materijala na bazi smola / Determination of mechanical and surface properties of dental resin-based nanocomposites

<p>Uvođenje nanočestica u stomatolo&scaron;ke kompozitne materijale predstavlja poku&scaron;aj da se odgovori univerzalnim zahtevima za kvalitetom direktnog zubnog ispuna, i da se stvori materijal koji kombinuje visoku mehaničku otpornost sa dobrim estetskim karakteristikama i zadovoljavajućim kvalitetom poliranja. Cilj sprovedene studije je bio da se ispita uticaj nanočestica, i soft-start metode svetlosne indukcije polimerizacije na mehanička i povr&scaron;inska svojstva savremenih stomatolo&scaron;kih nanokompozita, dostupnih na trži&scaron;tu. Ispitana su četiri stomatolo&scaron;ka nanostrukturisana kompozitna materijala na bazi smola i jedan univerzalni mikrohibridni kompozit, kao referentni materijal (Filtek Z250, 3M ESPE). Kori&scaron;ćena su po dva reprezentativna materijala iz dve podgrupe nanokompozita: nanopunjenih (Filtek Ultimate Body, 3M ESPE i Filtek Ultimate Translucent, 3M ESPE) i nanohibridnih kompozita (Filtek Z550, 3M ESPE i Tetric EvoCeram, Ivoclar Vivadent - TEC). Uzorci su polimerizovani nakon svetlosne aktivacije polimerizacije, uz kori&scaron;ćenje dva svetlosna režima: konvencionalnog i soft start režima. Pritisna i zatezna čvrstoća (dobijena poprečnim sabijanjem valjka, engl. diametral tensile strength) testirane su na Univerzalnoj ma&scaron;ini, kidalici. Tvrdoća uzoraka merena je testom za određivanje tvrdoće po Vickersu. Povr&scaron;inska tekstura i parametri hrapavosti određeni su skeniranjem povr&scaron;ine mikroskopom atomskih sila. Uop&scaron;teno, TEC je pokazao najniže vrednosti pritisne i zatezne čvrstoće, i tvrdoće, i statistićki je značajno bio slabiji od drugih testiranih materijala kroz sve mehaničke testove. Suprotno, TEC je imao najniže vrednosti parametara hrapavosti među testiranim materijalima. Spoj prepolimerizovanog punioca i polimerne baze u ovom materijalu pokazao se kao njegova slaba tačka. Navedeni materijal je iz tog razloga pokazao značajno slabiju otpornost od ostalih na razvijene napone izazvane dejstvom mehaničkih sila. Čestice nanodimenzija u sastavu stomatolo&scaron;kih polimernih kompozita, samostalno, nisu imale značajan uticaj na pobolj&scaron;anje mehaničkih i povr&scaron;inskih svojstava testiranih kompozita. Zaključeno je i da se procentualna zastupljenost neorganskih čestica u polimernoj bazi ne može smatrati apsolutnim kriterijumom kvaliteta kompozita, u pogledu njihovih mehaničkih svojstava. Samo srodni materijali, izrađeni istim tehnolo&scaron;kim postupkom, koji imaju veoma sličan ili isti hemijski sastav, pokazali su se kao mehanički superiorniji ukoliko su sadržali veći procenat neorganske komponente u svom sastavu. Nano prefiks u nazivu klase materijala ne garantuje sigurnu prednost stomatolo&scaron;kih nanokompozita nad univerzalnim mikrohibridnim kompozitima.</p> / <p>The introduction of nanoparticles in dental composite materials was an attempt to respond to the universal quality requirements for a direct dental restoration, and to create a material that meets the needs of high mechanical resistance, good aesthetic characteristics and surface properties of tooth restoratives. The aim of this study was to investigate the effect of nanoparticles in materials composition, and the soft start photoactivation method on the mechanical and surface properties of contemporary dental nanocomposites, available in the market. Four dental resin based nanostructured composites were tested along with a universal microhybrid one, as reference material (Filtek Z250, 3M ESPE). Two representative materials from the two classification subgroups were tested, nanofilled (Filtek Ultimate Body, 3M ESPE and Filtek Ultimate Translucent, 3M ESPE) and nanohybrid composites (Filtek Z550, 3M ESPE and Tetric EvoCeram, Ivoclar Vivadent, TEC). Polymerization of the samples was light activated using two light modes: conventional and soft start. Compressive and diametral tensile strength were tested on the Universal testing machine. The Vickers hardness was also determined. Surface texture and roughness parameters were examined by atomic force microscopy. Generally, TEC showed the lowest values of compression, tensile strength and hardness, and was statistically different from the other tested materials throughout all mechanical tests. In contrast, TEC had the lowest values of roughness parameters among the tested materials. In this material, contact zone of prepolimeryzed filler and polymer matrix appeared to be a weak point. For this reason, this material showed significantly lower resistance than the others on mechanically developed stresses. Nanosized particles within the dental polymer composites, individually, did not have a significant influence on improving the mechanical and surface properties of tested composites. It is concluded that the inorganic volume fraction in composites cannot be considered as an absolute criterion of their quality, regarding their mechanical properties. Only similar materials, made using the same technological process, which have very similar or the same chemical composition, and similar technological method of synthesis and optimization of organic and inorganic components, showed improved mechanical strength, if they contained a higher percentage of inorganic components in their composition. Nano prefix in the name of material&rsquo;s class does not guarantee the pure advantage of dental nanocomposites in comparison with the universal microhybrid composites.</p>

Model procene uticaja raspodele katjona toksičnih metala u heterogenom sistemu tečno/čvrsto na kvalitet površinskih voda / Impact assessment model of toxic metal cations partitioning in heterogeneous system liquid/solid sistem for surface water quality

<p>U doktorskoj disertaciji evaluirani su interkorelacioni rezultati odnosa ključnih fizičko-hemijskih parametara i katjona metala u površinskoj vodi Dunava, Tise (na po tri selektovana reprezentativna lokaliteta) i kanala Dunav-Tisa-Dunav (na dva lokaliteta). U<br />Kanalu Dunav-Tisa-Dunav praćene su i rezidualne katjonske koncentracije metala u sedimentu. Na osnovu podataka prisutnosti katjona metala u heterogenom sistemu tečno/čvrsto izračunate su po prvi put konstante podeonih procesa za ispitivane katjone metala. Primenom multivarijantnih statističkih metoda analizirani su korelacioni odnosi između ključnih fizičko-hemijskih parametara i koncentracionih nivoa rezidula katjona teških, lakih i semimetala. Faktorskom analizom (PCA) i klaster analizom sa dendogramima tumačeni, analizirani i diskutovani su dobijeni rezultati. Za optimalnog uočavanja korelacionih odnosa primenjeno je unapređeno rangiranje određenih skupova podataka (interkorelacije ključnih fizičko-hemijskih parametara i rezidua katjona metala) i grafički prikazanih Haseovih dijagrama primenom DART softvera (Decision Analysis by Ranking Techniques) za definisani model procene uticaja raspodele katjona toksičnih metala u heterogenom sistemu tečno/čvrsto na kvalitet površinskih voda.</p> / <p>In the doctoral thesis are evaluated inter correlational results of key physicochemical parameters and metal cations in surface water of Danube, Tisa (on three selected representative sampling points) and canal Danube-Tisa-Danube (on two sampling sites). In the canal Danube-Tisa-Danube are also followed residual metal cations concentrations in sediment. Based on metal cations presence data in heterogeneous system liquid/solid for the first time partitioning processes constants are calculated for<br />selected metal cations. Appling multivariate statistical methods correlations between key physicochemical parameters and concentration levels of residual heavy, light and semimetals cations are analysed. Using factorial analyse (PCA) and cluster analyse with dendograms obtained results were interpreted, analysed and discussed. For optimal comprehension of correlations, advanced method ranking of finite order sets is applied (inter correlations key physicochemical parameters and residual metal cations) and graphically shown Hasse diagrams obtained by DART software (Decision Analysis by Ranking Techniques) for defined impact assessment model of toxic metal cations partitioning in heterogeneous system liquid/solid for surface water quality.</p>

Uticaj antiseptika i antibiotika na formiranje bakterijskog biofilma na različito teksturisanim silikonskim implantatima za dojku / The influence of antiseptic and antibiotic irrigation on bacterial biofilm formation on silicon breast implants

<p>Najče&scaron;ća komplikacija nakon ugradnje silikonskih implantata za dojku je kontraktura fibrozne kapsule (KK), koja se normalno stvara oko implantata u sklopu reakcije oko stranog tela. Najozbiljnija komplikacija nakon ugradnje silikonskih implantata za dojku je anaplastični krupnoćelijski limfom koji se javlja isključivo kod pacijentkinja koje imaju ugraĎene implantate (eng. Breast-implant associated anaplastic large cell lymphoma &ndash; BIA ALCL). Uzrok nastanka ovih komplikacija ostaje nepoznat. Ustanovljeno je da se KK manje javlja kod implantata koji imaju makroteksturisanu povr&scaron;inu i kod onih koji su presvučeni poliuretanskom penom. S druge strane, BIA-ALCL se če&scaron;će dijagnostikuje kod pacijentkinja kojima su ugraĎeni upravo makroteksturisani implantati. Subklinička infekcija koja predstavlja odgovor organizma na postojanje biofilma na ugraĎenim implantatima, predstavlja jedan od najznačajnijih etiolo&scaron;kih faktora za nastanak KK i BIA-ALCL. Biofilm je konglomerat mirkoorganizama uronjenih u matriks koji ih &scaron;titi od dejstva antibiotika i antiseptika. Kako je nemoguće delovati medikamentozno na eradikaciju biofilma, brojni autori daju razne preporuke u cilju izbegavanja kontaminacije implantata tokom operativnog zahvata, a time i formiranja biofilma. Pored brojnih mera, savetuje se i ispiranje džepa u koji će se plasirati proteza kao i same proteze, nekim od antiseptičkih ili antibiotskih rastvora. Do sada ne postoje prihvaćene jasne preporuke o načinu ispiranja različitih implantata, objavljena su samo lična iskustva raznih autora. Ciljevi ovog istraživanja su bili da se ustanovi mogućnost formiranja biofilma četiri različite bakterije (Staphylococcus epidermidis, Staphylococcus aureus, Pseudomonas aeruginosa i Ralstonia pickettii) na tri različito teksturisana silikonska implantata za dojku (sa porama veličine 70-150 &mu;m, 50&ndash;900 &mu;m i 13 &mu;m) u in vitro uslovima; da se ispita da li ispiranje antisepticima (oktenidindihidrohloridom i povidon jodom), ili antibiotikom (cefuroksimom) ili istovremeno me&scaron;avinom povidon joda i antibiotika pre bakterijske kontaminacije sa četiri različite bakterije ima uticaja na formiranje biofilma na tri različito teksturisana implantata za dojke u in vitro uslovima; i da se ispita efekat antiseptika u odnosu na efekat antibiotika na formiranje bakterijskog biofilma na tri različito teksturisana silikonska implantata za dojku. Istraživanje je koncipirano kao prospektivna studija u vidu eksperimenta koji je izveden u Laboratoriji za mikrobiologiju, Instituta za javno zdravlje Vojvodine u Novom Sadu. Za izvoĎenje eksperimenta kori&scaron;ćeni su uzorci tri vrste silikonskih implantata za dojku sa različito teksturisanom povr&scaron;inom, odnosno porama različite veličine: 70-150 &mu;m, 50&ndash;900 &mu;m, i 13 &mu;m. Od svakog od navedenih implantata su pravljeni uzorci, sečenjem kapsula implantata na komadiće veličine 1x1 cm. Ukupno je bilo 1440 uzoraka. Na osnovu teksture uzorci su podeljeni u tri grupe: Grupa 1 (pore veličine 70-150 &mu;m), Grupa 2 (pore veličine 50&ndash;900 &mu;m) i Grupa 3 (pore veličine 13 &mu;m). Svaka od ovih grupa je dalje podeljena u jednu kontrolnu grupu i po četiri ispitivane grupe. Nakon sterilizacije uzoraka svaka kontrolna grupa je kontaminirana sa po 100&mu;l bakterijskog bujona Staphylococcus epidermidis (n=30), Staphylococcus aureus (n=30), Pseudomonas aeruginosa (n=30) i Ralstonia pickettii (n=30). Ispitivane grupe se bile podeljene prema načinima ispiranja na one u kojima su uzorci prvo ispirani: oktenidin &ndash; dihidrohloridom ili povidon jodom ili cefuroksimom ili kombinacijom povidon joda i dva antibiotika, pa potom kontaminirani sa po 100&mu;l bakterijskog bujona Staphylococcus epidermidis (n=30), Staphylococcus aureus (n=30), Pseudomonas aeruginosa (n=30) i Ralstonia pickettii (n=30). Po zavr&scaron;enoj kontaminaciji, uzorci su se inkubirali na temperaturi od 37&deg;C u trajanju od 96h, čime su stvoreni uslovi za formiranje biofilma. Nakon inkubacije, svaki pojedinačni uzorak je uronjen u sterilan tripton soja bujon, izlagan soničnoj energiji u trajanju od 1minuta i zatim vorteksiran 1 minut, čime je omogućeno odvajanje nastalog biofilma od implantata. Za ispitivanje sposobnosti formiranja biofilma kori&scaron;ćena je modifikovana tehnika sa mikrotitar pločom po Stepanoviću. Rezultati su pokazali da sve četiri ispitivane bakterije S. epidermidis, S. aureus, P. aeruginosa i Ralstonia pickettii statistički značajno vi&scaron;e stvaraju biofilm na implantatima sa porama veličine 50&ndash;900 &mu;m u odnosu na pore 70-150 &mu;m i u odnosu na pore veličine 13 &mu;m. Biofilm se statistički značajno vi&scaron;e stvara na porama veličine 70-150 &mu;m u odnosu na pore 13 &mu;m. Jedini izuzetak je Pseudomonas aeruginosa kod kojeg ne postoji statistični značajna razlika u produkciji biofilma na teksturisanim implantatima sa porama veličine 70-150 &mu;m u odnosu na one sa porama 13 &mu;m. TakoĎe, sve četiri ispitivane bakterije statistički značajano manje stvaraju biofilm nakon ispiranja povidon jodom, oktenidin-dihidrohloridom ili rastvorom antibiotika u sve tri grupe implantata, u odnosu na povr&scaron;ine koje nisu ispirane. Izuzetak je S. epidermidis u Grupi 3 kod kojeg nije utvrĎeno statistički značajno manje formiranje biofilma nakon ispiranja oktenidin dihidrohloridom u odnosu na neispiranje. Cefuroksim je bio efikasniji u sprečavanju formiranja biofilma sve četiri ispitivane bakterije u odnosu na neispiranje u Grupi 1, kao i za S. epidermidis i Ralstoniu Pickettii u Grupi 2. Cefuroksim se nije pokazao statistički značajno efikasnim u sprečavanju formiranja biofilma S. aureus i P. aeruginosa u Grupi 2, kao ni kod jedne bakterije u Grupi 3. Dalje je dokazano da su antiseptici (oktenidin-dihirohlorid i povidon jod) kao i me&scaron;avina povidon joda i dva antibiotika (cefuroksim i gentamicin), statistički značajno efikasnji od ispiranja samo antibiotikomcefuroksimom u smanjenju formiranja biofilma sve četiri ispitivane bakterije kod sva tri ispitivana, različito teksturisana silikonska implantata. Rezultati su pokazali da je ispiranje povidon jodom statistički značajno efikasnije u prevenciji stvaranja biofilma kod skoro svih ispitivanih bakterija od ispiranja oktenidin- dihidrohloridom u sve tri grupe implantata. Statistički značajna razlika nije utvrĎena u prevenciji stvaranja biofilma Staphylococcus aureusa kod sve tri grupe implantata prilikom ispiranja povidon jodom u odnosu na oktenidin- dihidrohlorid, kao i kod Ralsotnia pickettii u Grupi 2. Na osnovu rezultata ove studije, preporuka je da se koriste mikroteksturisani implantati kao i da se oni, pre ugradnje isperu povidon jodom ili me&scaron;avinom povidon jod i dva antibiotika (cefuroksim i gentamicin), u cilju prevencije stvaranja biofilma, a time i postoperativnih komplikacija koje mogu nastati nakon ugradnje implantata.</p> / <p>The most common complication after breast implant surgery is contracture of capsule, which is normally formed around implants as part of foreign body reaction. The most sincere complication after this kind of surgery is breast implant associated anaplastic large cell lymphoma (BIA-ALCL). The cause of these complications is still unknown. It is evident that capsular contracture (CC) is seen less frequently in patients with macro-textured implants and in those with implants covered with polyurethane foam. On the other hand, BIA-ALCL is diagnosed more frequently in patients with those, macro-textured implants. Subclinical infection, defined as an response of organism on presence of biofilm on the implant, is considered to be one of the most important etiologic factors for CC and BIA-ALCL. Biofilm is a conglomerate of microorganisms immersed into matrix, which protects them from influence of antibiotics and antiseptics. As it is impossible to eradicate biofilms with medicaments, many authors suggest different steps in order to avoid contamination of the implant during the operation and therefore, prevent the formation of biofilm. Among many tips, it is recommended to irrigate the pocket for breast implant and the implant itself, with some antiseptic or antibiotic solution. Up till now, there is no agreed consensus on the type of irrigation for different implants. Only personal experiences of a few authors have been published. Aims of this research were: to establish the possibility of biofilm formation of four different bacteria (Staphylococcus epidermidis, Staphylococcus aureus, Pseudomonas aeruginosa and Ralstonia pickettii) on three differently textured breast implants (with pore diameter of 70-150 &mu;m, 50&ndash;900 &mu;m and 13 &mu;m) in vitro; to examine whether the irrigation of implant with antiseptics (povidone iodine and octenidine dihydrochloride), antibiotics (cefuroxime) or mixture of povidone iodine and two antibiotics, before the contamination with bacteria, has an influence on the incidence on biofilm formation on three differently textured implants; and to examine the effect of antiseptics in contrast to the effect of antibiotics on biofilm formation on three differently textured breast implants. The study was conducted as a prospective research that took place at the Laboratory for microbiology, at the Institute of public health of Vojvodina in Novi Sad. For the experiment, three types of silicone breast implants were used with different pore sizes: 70-150 &mu;m, 50&ndash;900 &mu;m and 13 &mu;m. Samples were made by cutting each of these types of implants into pieces sized 1x1cm. There were 1440 samples in total. According to texture, samples were divided it three groups: Group 1 (pore size 70-150 &mu;m), Group 2 (pore size 50&ndash;900 &mu;m) and Group 3 (pore size 13 &mu;m). Furthermore, each of these groups was divided in one control and four test groups. After sterilisation of samples, every control group was contaminated with 100&mu;l of bacterial broth of Staphylococcus epidermidis (n=30), Staphylococcus aureus (n=30), Pseudomonas aeruginosa (n=30) and Ralstonia pickettii (n=30). Tested groups were divided according to type of irrigation into those where samples were firstly irrigated with either: octenidine dihydrochloride of povidone iodine or cefuroxime of mixture of povidone iodine with two antibiotics, and after the irrigation, contaminated with 100&mu;l bacterial broth of Staphylococcus epidermidis (n=30), Staphylococcus aureus (n=30), Pseudomonas aeruginosa (n=30) and Ralstonia pickettii (n=30). After contamination, samples were incubated on 37&deg;C for 96h, which created excellent conditions for biofilm formation. After incubation, each sample was dipped into sterile tripton soy broth, and then exposed to sonic energy for 1 minute and vortexed for 1 minute, which made biofilm separate from the implant. For testing the capability of biofilm formation, modified technique with microtitar plates described by Stepanović was used. Results show that all four examined bacteria S. epidermidis, S. aureus, P. aeruginosa and Ralstonia pickettii form more biofilm on implants with pore sizes 50&ndash;900 &mu;m compared to implants with pore size 70-150 &mu;m and those with 13 &mu;m. Statistical significance was found in biofilm formation on implants with pores 70-150 &mu;m compared to implants with pores 13 &mu;m. Furthermore, all four examined bacteria form statistically less biofilm after the irrigation with any of used solutions: povidone iodine, octenidine dihydrochloride, antibiotic solution of mixture of povidone iodine and two antibiotics, in all three groups of implants compared to surfaces that were not irrigated. The exception is S. epidermidis in Group 3, where no statistical significance was found on biofilm formation after the irrigation with octenidine dihydrochloride compared to non-irrigation. Cefuroxime was more efficient in biofilm prevention for all four tested bacteria compared to non-irrigation in Group 1 and for S. epidermidis and Ralstonia pickettii in Group 2. There was no statistical significance found in prevention of S. aureus i P. aeruginosa biofilms when irrigating with cefuroxime in Group 2, as well as for all tested bacteria in Group 3. Furthermore, it was verified that antiseptics (octenidin dihydrochloride and povidone iodine) and mixture of povidone iodine and two antibiotics (cefuroxime and gentamycin), were statistically more efficient in biofilm prevention of all four examined bacteria in all groups of implants, compared to irrigation with antibiotic-cefuroxime alone. Results show that irrigation with povidone iodine is statistically more efficient in biofilm prevention of almost all examined bacteria compared to irrigation with octenidine dihydrochloride in all groups of implants. There was not found any statistical significance in prevention of Staphylococcus aureus biofilm when irrigating with povidone iodine compared to octenidine dihydrochloride in all groups of implants, and also in biofilm prevention of Ralsotnia pickettii in Group 2. According to results of this research, it is recommended to use micro-textured implants and to irrigate them with povidone iodine or mixture of povidone iodine and two antibiotics (cefuroxime and gentamycin) prior the implementation, in order to prevent biofilm formation which is most probable cause of postoperative complications after implant surgery.</p>
