Title of diploma thesis: Investigating the role of zinc transporter ZIP 6 and STAT3 in mitosis It has been shown that STAT3 (signal transducer and activator of transcription 3) plays a role in the development of cancer. ZIP6 is the downstream target of this transcription factor. Previous research has focused mainly on the activation of STAT3 by tyrosine phosphorylation, while the effect of phosphorylation at a second site, serine 727, remained relatively uninvestigated. In this study, it is proposed that serine-phosphorylated STAT3 is activated throughout mitosis in tamoxifen-resistant (TamR) breast cancer cells and that zinc transporter ZIP6 and serine-phosphorylated STAT3 are involved in a zinc-mediated mitotic mechanism. After using nocodazole to induce mitotic arrest, the expression of tyrosine- phosphorylated STAT3 protein was observed to be reduced while the expression of serine- phosphorylated STAT3 was increased. The differences between signalling pathways in MCF7 cell line and TamR cells were evaluated with a special focus on a role of MAP kinase, which activation is believed to be linked with malignant diseases. An effect of zinc on various cellular kinases in 0, 2, 5, 10, 15 and 20 minute of zinc treatment was analyzed in MCF7 cells transfected by wild and mutant type of ZIP 7, TamR cells and TamR cells pre-treated with MAP kinase inhibitor (PD) using the methods of western blotting and fluorescent microscopy. We show here the dependence of activation of pMAP kinase and other important oncogenic kinases (such as Lyn, Src and STAT3) on zinc release into cytoplasm. According to our results, MAP kinase is activated very upstream and it can stimulate many important protein kinases as Src Y418 , STAT3 S727 and Lyn Y396 in tamoxifen-resistant breast cancer...

The aim of the diploma thesis is the fattening of the layer males hybrid combinations type Dominant. It is very unethical that the layer males are put to death immediately after hatching and regarded as a waste in most countries with advanced the poultry industry. One of the options how to prevent this putting to death is to use alternative forms of fattening. An experiment was conducted within the scope of the diploma thesis. 200 pieces of cockerels 4 hybrid combinations were fattened during the experiment. The cockerels were fattened on a litter as well as in cages untill the ages of 12 weeks. The life weight and feed conversion were located by cockerels until the age of 12 weeks by the layer males during fattening. In the age of 12 and 18 weeks the live weight, carcass weight, carcass value, the percentage yield of breast muscle and yield leg muscle of the carcass and the percentage of the heart, liver and thigh bones from the carcass were determined. The dry matter content, fat content, and also the colour of L *, a * and b * from breast and thigh muscle were evaluated. Regardless of the hybrid combination the life weight was always higher by cockerels housed on a litter. The feed conversion by hybrids in cages was not demonstrably different, but the conversion by the hybrid housed on litter in the hybrid D - 109 was lower than by other hybrid types. The highest conversion during the whole fattening period was measured by the hybrid D -- 459. The hybrid combination had a statistically demonstrative influence (P < 0,05) on the slaughter value in each of the ages. The fat content of breast and thigh meat increased by all hybrid combinations with the age.

The aim of diploma thesis was monitoring the influence of grass in feed on content of fatty acids in chichen thighs and breast meat. For experiment there were used 50 chickens divided to three groups. The first one was fast growing hybrid Ross 308, the second one was middle growing hybrid JA 757 and the last one was combined hybrid ISA Dual. The fattening was specified for each hybrid from 42 till 90 days. Feedin and water supply was ad libitum. The fortified feed was intaken 2/3 of whole fattening for every hybrid. There were statistically significant differences (P < 0,05) between breeds. There was increase of fatty acids in hybrid thighs and breasts of Ross 308. This hybrid has the highest amount of fatty acids. Between males and females there was no statistically significant differences (P < 0,05).

The topic of this thesis is the quality of poultry meat in relation to the production method. In the literature review, poultry meat is described, then its consumption in the Czech Republic and its chemical composition. Selected methods of sensory analysis are also described. Types of tested meats are characterized in the practical part. This thesis is also focused on the experimental evaluation of the chemical composition of chicken breast muscle from the Loužná farm using the NIR method and comparing the composition changes depending on the age of the chickens. It is followed by selected sensory methods aimed at recognizing differences in chicken meat from three different productions. In addition, there was published a questionnaire to determine citizen's preferences for products (chicken meat) from different production methods.

Evaluation of water use efficiency of short rotation poplar coppice at Bohemian-moravian highlands

The diploma thesis provides basic characteristics of short rotation coppice plantations, then the most significant short rotation coppice species cultivated in the Czech Republic are described. The detailed description of particular Populus L. clone J-105 grown at the plantation of the Test Station Domanínek Ltd. follows. Afterwards, key determinants of short rotation coppice yield are presented in the text. The last theoretical part of the thesis deals with water use efficiency (WUE) with emphasis on the method used at the experimental plantation in Domanínek. The methods of sap flow measurements and biomass determinations are described for this purpose. The main aim of the practical part of the thesis is to determine WUE of 16 measured trees where sap flow measuring systems were installed. These trees are divided into 3 cohorts according to diameters at breast height (DBH) at the beginning of the growing season 2013. The additional measurements were carried out: phenological imaging, soil moisture measurements, LAI measurements and sapwood area determination. The statistical analysis of WUE did not show any statistically significant differences between values of WUE for individual cohorts. Only in May, the statistically significant difference between cohort 1 (trees with the thickest stems) and cohort 3 (trees with the lowest values of DBH) appeared.
