The thesis deals with the proposal to introduce a quality management system. In the first part of the thesis is done a literature search in the field of quality management systems (QMS). In the second part is described possibility of implementation of QMS in company DIEFFENBACHER. In the third part is done SWOT analysis for company DIEFFENBACHER. There are defined strengths, weaknesses, opportunities and threats. Based on this analysis is elaborated a strategy. In the fourth part is described process of implementation of QMS. In the last part of the thesis is a feedback and overall efficiency of implementation of the entire QMS for company DIEFFENBACHER DCZ.

Práce se zabývá orientací na zákazníka v souvislosti s mezinárodními normami ISO. Obsahuje rozbor ČSN EN ISO 9001:2002, podpůrných norem řady ISO 10000 a ČSN ISO 20252:2007. Hledá vazbu mezi orientací na zákazníka a výsledky podniku podle ČSN ISO 10014:2007. V další části se zaměřuje na praktické využití tohoto souboru norem v oblasti marketingu.

Graduation Theses describes Management System of Quality (QMS) according to Needs of Standard Specifications ISO 9001:2000 in Antikvariát VŠE. Target was designed funktional QMS and implemented Survey of customer's Satisfaction with proposition of Quality Manual. This work is designed like Interpretation of single points ISO - Standards and with practical Solutions for Antikvariát VŠE.

With Chiptech’s current growth rate and size it has become necessary to develop a Quality Management System to enable repeatability, meet customer demands, and protect Chiptech from staff turnover. ISO 9001 was identified as a base for development, with the imperative that the system identified and developed must deliver value for Chiptech. Several frameworks were investigated, along with journal articles and discussions with industry members in order to determine the aspects that would deliver value, and determine the key success factors. Two factors were identified as critical: employee involvement, and the utilisation of metrics – both of which were leveraged for the project results and recommendations. The systems developed have already proved they offer benefits, however, in order to maintain performance Chiptech must a) keep evaluating the measured results, b) ensure that quality forms an integral part of the organisational culture and, c) continue the systematic approach of continual improvement.

Karel Dítě. The theoretical part of the work focuses on the history, where I explain the basic concepts and issues relating to the use of literature associated with the ISO 9001 Quality Management System. This section also describes the general process for implementing the standards within an organization. Following on from this, I have used the knowledge gained from the theoretical section within the practical part of the thesis. The practical part of my work includes a description of the selected organization from several perspectives - it is a focus, organizational structure, customer and supplier portfolio. This section also analyses the current state of the organization and includes a proposal to meet the ISO standards. The final section contains the documents necessary for the implementation of ISO 9001 within the organization, so that the quality management system corresponds to the prescribed standards.

Neste cenário, a análise do metaboloma utilizando apenas uma técnica analítica é por vezes inviável devido à magnitude química dos compostos bem como de sua faixa de concentração. Desta forma, o trabalho utilizou a combinação de HS-SPME (Microextração em Fase Sólida) aliada à técnica GC×GC-qMS (Cromatografia Gasosa Bidimensional Abrangente acoplada à Espectrometria de Massas Quadrupolar) para a obtenção do perfil metabólico volátil inédito de duas espécies de fungos com potencial ação para biocontrole e indução de resistência a fitopatógenos na agricultura, Curvularia sp e Memnoniella sp. Através de modelagem MPCA e HCA foi possível a obtenção de um perfil cinético característico da produção de compostos voláteis das espécies que delimitou o processo de identificação tentativa destes metabólitos. Hidrocarbonetos e álcoois foram responsáveis pela maior produção volátil de ambas as espécies avaliadas, com destaque para a produção de lactonas como y-octalactona, y-hexalactona e y-hexalactona por Curvularia sp e sesquiterpenos como acoradieno, ß-chamigreno, a-chamigreno, ß-elemeno e valenceno por Memnoniella sp. A partir do perfil volátil das espécies, os estudos poderão ser direcionados à ação biocontrole e indução de resistência na agricultura / Abstract: Volatile compounds produced by fungi as part of biocontrol strategies in preventing diseases caused by pathogens has been stimulating the market of biofungicides. So, the analysis of the metabolome using only one analytic technique is sometimes not feasible due to chemical compounds as well as the magnitude of its concentration range. Thus, the study used a combination of HS-SPME technique (Solid-Phase Microextraction) coupled with GC×GC-qMS (Comprehensive Two-Dimensional Gas Chromatography coupled to Quadrupole Mass Spectrometry) to obtain volatile metabolic profiles of two species of fungi with potential for biocontrol activity and induction of resistance to plant pathogens in agriculture, Curvularia sp and Memnoniella sp. Through modeling MPCA and HCA was possible to obtain a typical kinetic profile of volatile compounds from the production of species which delimited the process of identification of these metabolites. Hydrocarbons and alcohols were responsible for most volatile production of both studied species, with emphasis on the production of lactones such as y-octalactona, y-hexalactona e y-hexalactona by Curvularia sp and sesquiterpenes as acoradiene, ß-chamigrene, a-chamigrene, ß-elemene and valencene by Memnoniella sp. La détermination d’âges absolus est nécessaire pour calibrer la chronologie relative martienne basée sur la densité de cratères d’impact. Dans ce but, nous avons développé au laboratoire GEOPS un prototype pour la datation in-situ des surfaces planétaires par la méthode K-Ar. Cette installation, financée dans le cadre d’un programme de recherche CNES-CNRS, est basée sur l’ablation laser permettant la vaporisation d’un volume reproductible de roche. La composition chimique de la matière ablatée, et en particulier la teneur en potassium (K), est estimée par Laser Induced Breakdown Spectroscopy (LIBS) et la quantité d’argon (Ar) libérée est mesurée par spectrométrie de masse quadripolaire (QMS). La détermination de l’âge K-Ar requiert en outre la quantification précise de la masse d’échantillon ablatée. Après une phase importante de développement instrumental visant à améliorer la précision et la sensibilité de la technique, nos efforts ont porté sur la calibration du dispositif. Pour ce faire, une collection d’échantillons de référence (micas, feldspaths, amphibolite, basaltes) a été réalisée et analysée (chimie totale et mesures d’âges par les techniques K-Ar et ⁴⁰Ar/³⁹Ar classiques). Ces échantillons présentent des teneurs en K de 0,15 à 11% et des teneurs de 1 à 100x10¹⁴ atomes d’⁴⁰Ar* par gramme, équivalent à des basaltes martiens âgés de 700 Ma à 3 Ga pour 0,4%K. Les résultats obtenus ont permis de définir une corrélation entre le temps d'ablation et la masse ablatée selon le type de phase minérale ciblée. Afin de valider cette corrélation, des mesures ont été menées « en aveugle » sur plusieurs échantillons, en particulier un basalte microlitique (Trapps de Viluy) similaire aux basaltes composant la surface de Mars. L’âge K-Ar in-situ de 401 ± 41 Ma obtenu par notre approche est consistant avec l’âge de 381 ± 5 Ma mesuré indépendamment sur populations de minéraux. Plus généralement, cette étude a permis d’obtenir des âges K-Ar mesurés par notre méthode de datation in-situ avec une incertitude inférieure à 15%, y compris sur des roches peu potassiques et relativement jeunes comparées aux roches martiennes. / Absolute age determination is necessary to check and calibrate the relative Martian chronology presently available from meteoritic crater counting. For this purpose, we have developed an in-situ K-Ar dating prototype for planetary surfaces at GEOPS laboratory. This instrument, supported by a CNES-CNRS research program, is a laser ablation-based system built to vaporize a reproducible volume of rock or mineral. It quantifies potassium content (K) by laser-induced breakdown spectroscopy (LIBS) and argon (Ar) by quadrupole mass spectrometry (QMS). The K-Ar age determination also requires accurate quantification of the ablated sample mass. Following an important period of experimental set-up development to improve precision, accuracy and sensitivity of the technique, our efforts focused on the calibration of the different parts of this instrument. To that purpose, a collection of reference samples (micas, feldspars, amphibolite, basalts) was done and analyzed (total chemistry and age measurements by K-Ar and ⁴⁰Ar/³⁹Ar conventional techniques). These samples show a K content between 0,15 and 11% and an Ar content between 1 and 100x10¹⁴ ⁴⁰Ar* atoms per gram, corresponding to Martian basalt from 700 Ma to 3 Ga for 0.4% K. The results obtained evidence a correlation between the ablation time and the ablated mass depending on the mineral phase analyzed. In order to check this correlation, measurements were conducted on several samples, in particular a microlitic basalt (Viluy basalt) similar to Martian basalts. The K-Ar in-situ age of 401 ± 41 Ma obtained by our approach is consistent with the age of 381 ± 5 Ma measured indepedently on mineral populations. More generally, this study allows to obtain K-Ar ages measured by our in-situ dating method with uncertainties lower than 15%, on relatively low-K and young samples compared to Martian rocks.
