Previous issue date: 2018-02-21 / Ao considerar as brincadeiras de pular corda na perspectiva cultural, estaremos favorecendo para a sua preservação enquanto patrimônio histórico das culturas infantis e contribuindo para que a criança se sinta parte do processo dessas brincadeiras. Nesse sentido foi firmada uma problemática: “Como os professores têm trabalhado as brincadeiras de pular corda, pertencente aos Jogos e Brincadeiras Tradicionais, considerando-a como patrimônio histórico cultural da infância ao receber as crianças no Ensino Fundamental/Anos Iniciais?” Assim, objetivou-se analisar e disseminar as brincadeiras de pular corda na infância das crianças das Escolas de Ensino Fundamental/Anos Iniciais. Como objetivos específicos foram: analisar as percepções dos professores de Educação Física em relação às crianças ingressantes no EF/AI, ao introduzir as brincadeiras de pular corda e evidenciar o processo de ensino aprendizagem desses professores; constatar como foram essas brincadeiras na infância dos avós que possuem vínculo com os alunos do 1º ano e 2º ano de uma das escolas; elaborar um vídeo, contendo a história de vida dos avós participantes sobre suas brincadeiras de infância, depoimentos de profissionais envolvidos com o processo educacional e com orientações pedagógicas no ensino e aprendizagem das brincadeiras de pular corda, valorizando a cultura evidenciada pelos avós e contribuições dos professores. A abordagem metodológica adotada para esta pesquisa foi a qualitativa. Para tanto, realizou-se a revisão de literatura, utilizando como instrumentos de coleta de dados a entrevista semiestruturada para treze professores de Educação Física e o questionário para trinta e um avós. Na construção do vídeo foi utilizado um roteiro de filmagem e câmera de vídeo. Esse estudo foi realizado em uma cidade do interior do estado de São Paulo, e a coleta de dados foi dividida em duas etapas: na primeira foi feita as entrevistas com os professores de Educação Física e na segunda, aplicação dos questionários aos avós. Após o tratamento, análise e discussão dos dados, concluiu-se, com relação aos Jogos e Brincadeiras Tradicionais, bem como as brincadeiras de pular corda, que os professores de Educação Física que participaram da pesquisa, apresentam conhecimento com esse conteúdo em diversas manifestações corporais, entretanto, alguns foram indicando dificuldade em seu planejamento e prática pedagógica alicerçada na valorização histórica cultural das brincadeiras de pular corda. Nesse contexto, convidamos os professores a pensarem as manifestações da cultura corporal numa perspectiva cultural, oportunizar aos alunos experiências que valorizem o patrimônio cultural corporal das culturas infantis e se compreendam como pertencentes a eles. A partir das conclusões da presente pesquisa, foi elaborado, como produto final do mestrado, um vídeo com a intenção de subsidiar os professores no ensino das brincadeiras de pular corda numa perspectiva cultural, intitulado como “Vamos Brincar de Pular Corda?”, contando com a participação de avós, alunos e professores e profissionais da área da educação, no qual ele é composto por quatro esferas do aprender: Pesquisa/acervo; Ampliação; Aprofundamento e Disseminação do conhecimento. Acreditamos que o produto educacional contribuirá para disparar discussões/reflexões em cursos de formações iniciais e continuadas de professores e que repercuta na prática pedagógica fundamentada, voltada para valorização do brincar na escola. / When considering jump rope jokes in the cultural perspective, we will be favoring its preservation as a historical heritage of children's cultures and contributing to the child feeling part of the process of these games. In this sense, a problem was established: "How have teachers played the jumping rope games belonging to Traditional Games and Games, considering it as a historical cultural patrimony of childhood when receiving children in elementary school?" , the objective was to analyze and disseminate the jumping rope play in the childhood of elementary school children. The specific objectives were: to analyze the perceptions of Physical Education teachers in relation to incoming children in EF / AI, by introducing the jumping jump rope and to highlight the learning process of these teachers; to verify how these games were in childhood of the grandparents who have bond with the students of the first year and the second year of one of the schools; to elaborate a video, containing the life history of the participating grandparents about their childhood games, testimonials of professionals involved with the educational process and with pedagogical guidelines of the process of teaching and learning the jumping rope jokes, valuing the culture evidenced by the grandparents and contributions of teachers. The methodological approach adopted for this research was qualitative. For that, the literature review was performed, using as data collection instruments the semi-structured interview for thirteen Physical Education teachers and the questionnaire for thirty-one grandparents. In the construction of the video was used a script of filming and camera of video. This study was carried out in a city in the interior of the state of São Paulo, and the data collection was divided into two stages: the first was the interviews with Physical Education teachers and the second, the application of the questionnaires to the grandparents. After the treatment, analysis and discussion of the data, it was concluded, in relation to the Games and Traditional Games, as well as the games of skipping rope, that the Physical Education teachers who participated in the research, present knowledge with this content in diverse corporal manifestations , however, some were indicating difficulty in their planning and pedagogical practice based on the historical cultural valorization of jumping rope jokes. In this context, we invite teachers to think about the manifestations of body culture in a cultural perspective, to give students experiences that value the corporal cultural patrimony of children's cultures and understand themselves as belonging to them. Based on the conclusions of the present research, a video was created as a final product of the master's degree, with the intention of subsidizing the teachers in the teaching of jumping jump rope in a cultural perspective, titled "Let's Play Jumping Rope?" the participation of grandparents, students and teachers and professionals in the area of education, in which it is composed of four spheres of learning: Research / collection; Enlargement; Deepening and Dissemination of knowledge. We believe that the educational product will contribute to trigger discussions / reflections in initial and continuing teacher training courses and that it will have repercussions on the pedagogical practice based on the value of playing in school.

A necessidade de exploração de petróleo em águas profundas exige o emprego de dutos flexíveis com desempenho em fadiga adequado, sendo as armaduras de tração deste tipo de duto as camadas mais críticas para este tipo de solicitação. Atento a isto, este trabalho mostra o desenvolvimento de um novo conceito de armaduras de tração, que são construídas com cabos de aço. Para avaliar o desempenho dos cabos de aço na armadura tração foi elaborada uma configuração de armadura, com a seleção e aplicação de cabos de aço disponíveis no mercado nacional. Além da caracterização metalúrgica, dimensional e mecânica, foram feitos ensaios de fadiga ao ar e em água do mar sintética no cabo de aço selecionado. Ensaios em fadiga de uma seção da conexão foram feitos para avaliar as características da falha de fadiga, comparando-as com os tendões maciços. Também foi executada uma análise por elementos finitos para avaliar a distribuição de tensões no interior da resina, sendo validada pelo uso de extensômetros no cabo. Foi projetado e construído um duto com a configuração proposta, além das conexões. Este duto foi ensaiado em tração e fadiga e comparado com um duto de referência, em termos de peso, tensões e rigidez axial e a flexão. Os resultados sugerem um grande aumento na vida em fadiga nas conexões e no ambiente com água do mar. Além disso, o duto construído suportou com sucesso todos os modos de carregamento impostos ao duto de referência, superando o número de ciclos deste. As análises de falha indicaram que o duto desenvolvido apresentou o modo de fratura dúctil, enquanto que os ensaios com as seções da conexão indicaram sinais de fadiga em regiões com falha superficial dos fios. A rigidez axial é similar ao do duto de referência e a rigidez a flexão calculada é superior ao do duto de referência devido ao maior diâmetro das camadas das armaduras. Apesar do maior peso do duto desenvolvido, a resistência específica é similar. / The need for oil exploration in deep water requires the use of flexible pipes with better fatigue performance, and the tensile armor of this type of product layers critical for this type of application. Aware of this, this work shows the development of a new concept of tensile reinforcement, which are built with wire ropes. To evaluate the performance of wire ropes in tensile armor, an armor configuration was developed, with the selection and application of steel cables available in the domestic market. Besides metallurgical characterization, dimensional and mechanical fatigue tests were carried out in air and in synthetic sea water on wire rope selected. Fatigue tests on end-fitting section were made to evaluate the characteristics of fatigue failure, compared with the massive tendons. It has also been performed by finite element analysis to evaluate the stress distribution within the resin, being validated by the use of strain on the wire rope. It was designed and constructed a pipeline with the proposed configuration, in addition to connections. This pipeline was tested in tensile and fatigue compared to a reference pipeline in terms of weight, axial and flexural stiffness and axial tension. The results suggest a large increase in fatigue life on end-fitting and the environment with seawater. Furthermore, the flexible pipe developed successfully endure all modes of loading imposed on the reference pipe, surpassing the number of cycles thereof. The failure analysis indicated that the flexible pipe developed showed ductile fracture mode, while the trials with boxes indicated signs of fatigue failure in the regions with the surface of the wires. The axial stiffness is similar to the reference pipeline and the flexural stiffness calculated is higher than the reference product, due to the greater diameter of the layers of armor. Despite the greater weight of the product developed, the specific resistance is similar.

CoordenaÃÃo de AperfeiÃoamento de NÃvel Superior / Conselho Nacional de Desenvolvimento CientÃfico e TecnolÃgico / No presente trabalho, foi realizado um estudo preliminar sobre a expressÃo de um gene de quitinase de classe I de feijÃo-de-corda (Vigna unguiculata L.Walp), a clonagem e expressÃo desse gene em cÃlulas de Escherichia coli BL21(λ)DE3 e a determinaÃÃo via modelagem por homologia da estrutura tridimensional dessa proteÃna. A expressÃo do gene da quitinase de classe I de feijÃo-de-corda foi obtida a partir de RT-PCR com oligonucleotÃdeos iniciadores especÃficos. Nessas reaÃÃes, foram usadas amostras de RNA total de sementes e vagens em diferentes estÃgios de crescimento (2, 4, 6, 8 , 10, 12, 14, 16 e 18 dias), pertencentes a dois genÃtipos contrastantes quanto a infecÃÃo pelo Callosobruchus maculatus, IT81D-1053 (resistente) e TE97-419-07F (suscetÃvel). A expressÃo foi avaliada tambÃm em folhas, raÃzes, epicÃtilo e hipocÃtilo de dois genÃtipos contrastantes quanto a infecÃÃo pelo nematÃide das galhas Meloidogyne incongnita, CE-31 (resistente) e TE97-411-1F (suscetÃvel). A clonagem do gene VuChiI foi realizada a partir do produto amplificado da RT-PCR de sementes de IT81D-1053, e do produto amplificado a partir do DNA genÃmico de MONTEIRO. Foram obtidos 11 clones confirmados, dos quais 9 foram seqÃenciados. A subclonagem do clone R7 foi realizada em pET15b e a expressÃo da proteÃna recombinante foi induzida na presenÃa de IPTG 1mM. A proteÃna recombinante, com aproximadamente 30 kDa, foi visualizada atravÃs de um SDS-PAGE. A proteÃna purificada atravÃs de uma cromatografia de afinidade em coluna de Sepharose com NÃquel imobilizado nÃo apresentou atividade quitinÃsica significativa. Os modelos gerados para os clones obtidos e para a quitinase nativa, indicam que as mutaÃÃes ocorridas nÃo alteram os sÃtios ativos das molÃculas. Dessa forma, a quitinase de classe I do feijÃo-de-corda parece apresentar expressÃo constitutiva em todas as partes da planta. A quitinase recombinante obtida foi pouco ativa. As mutaÃÃes pontuais nos clones obtidos sugerem a ocorrÃncia de isoformas dessa proteÃna, o que ainda deve ser elucidado no futuro / In this work we have made a preliminary study on the expression of a class I chitinase gene
from cowpea (Vigna unguiculata L.Walp), cloning and expression of this gene in Escherichia coli BL21(λ)DE3 cells and, through homology modeling, we determined the three dimensional structure of this protein. The expression of the class I quitinase gene from cowpea was performed by RTÂPCR from the total RNA, with specific primers, from seeds and pods in distinct stages of development (2, 4, 6, 8 , 10, 12, 14, 16 and 18 days), belonging to two contrasting genotypes regarding to infection by Callosobruchus maculatus, IT81DÂ1053 (resistant) e TE97Â419Â07F (susceptible). The gene expression in leaves, roots, epicotyls and hipocotyls from two contrasting genotypes considering the infection by the nematoid Meloidogyne incongnita, CEÂ31(resistant) e TE97Â411Â1F (susceptible). The VuChiI gene cloning was accomplished from the amplified product on RTÂPCR with IT81DÂ1053 seeds, and the amplified product from the genomic DNA of MONTEIRO.
Eleven clones were obtained, from which nine were sequenced. The cloning of R7 clone was
accomplished in pET15b vector and the expression of the recombinant protein was induced in
the presence of IPTG 1mM. The protein, with 30 kDa, was visualized through a SDSÂPAGE.
The protein purified through an affinity chromatography in Sepharose column with
immobilized Nickel did not had a significative hydrolytic activity. The models generated for
the clones and for the native chitinase, have indicated that mutations that occurred did not changed the molecule's active sites. Thus, the class I chitinase gene from feijÃo de corda seems to present constitutive expression in all parts of the plant. The recombinant chitinase obtained was inactive. The specific mutations in the resulting clones suggests the occurrence
of isoforms of this protein, what should be elucidated in the future.

FLOW FIELD IN A HIGH HEAD FRANCIS TURBINE DRAFT TUBE DURING TRANSIENT OPERATIONSGoyal, Rahul January 2017 (has links)
Hydroelectricity plays an important role to balance the stability of grid network. In order to improve the stability of presently high loaded grids, hydropower plants are being operated over a wide range of operations and experiencing frequent start-stop, load rejection, and load acceptance. The turbines need to sustain sudden change in their operating condition to balance the grid frequency. Francis turbines have been widely used because of their wider operating range and higher stability in operation during rapid load variation. This has resulted in severe damage to the turbines as they are not normally designed to operate under such transient conditions. Several low and high frequency pressure fluctuations prevail during transients operating conditions. Generally, wall pressure measurements are performed which may not provide sufficient information to investigate the flow instabilities related to these fluctuations. Thus, the main objective of the present work is to simplify and perform optical measurements in a turbine during transient operating conditions to investigate the flow field. The measurements have been performed at the Water Power Laboratory using a high head model Francis turbine. The turbine is a 1:5.1 scale down model of a prototype operating at the Tokke Power Plant, Norway. The model runner diameter, net head, and discharge at the best efficiency point (BEP) were 0.349 m, 12 m, and 0.2 m3 s-1, respectively. A total ten pressure sensors were mounted at different locations namely, turbine inlet, vaneless space, and draft tube. The data were acquired at a sampling rate of 5 kHz. The instruments and sensors have been calibrated according to guidelines available in IEC standards. The determined total uncertainty in the measurement of hydraulic efficiency was ±0.15% at BEP condition. The velocity measurements in the draft tube cone were performed using a 2D PIV system and the images were sampled at a rate of 40 Hz. Steady state measurements were carried out considering the realistic design and off-design operating conditions of the prototype turbine. Therefore, the angular speed of the runner was maintained constant for all steady state conditions during the measurements. The maximum hydraulic efficiency (92.4%) was observed at nED = 0.18, QED = 0.15, and a = 9.8º, which is named BEP. It is observed that the turbine experiences significant pressure fluctuations at the vaneless space, runner, and the draft tube. The fluctuations due to rotor-stator interaction (RSI) were observed to be most dominating at high load condition, however, fluctuations due to the rotating vortex rope (RVR) at part load (PL) condition. Two different modes (synchronous and asynchronous) modes of vortex rope are observed at PL condition of the turbine. An asymmetry in the flow leaving the runner was detected at both design and off-design conditions, with a stronger effect during off-design operating condition. Numerical simulations of the model turbine were carried out at PL operating condition. The simulations were performed using two turbulence models, standard k-ε and SST k-ω, with high-resolution advection scheme. The numerical pressure values obtained with both standard k-ε model and SST k-ω showed a small difference with the experimental values. The amplitudes of numerical pressure values were higher (~2.8%) in the vaneless space and lower (~5.0%) in the draft tube than the experimental values. The frequencies of the RSI and RVR were well captured in the turbine but the amplitudes were overestimated. During load rejection from BEP to PL, the plunging mode of the vortex rope was observed to appear first in the system than that of the rotating mode. Whereas during the load acceptance from PL to BEP, both the modes were observed to disappear simultaneously from the system. In the velocity data, the axial velocity only contributed to the development of the plunging mode and radial velocity to the rotating mode. The region of low velocity, stagnation point, flow separation, recirculation, oscillating flow and high axial velocity gradients were well captured in the system during the transients. The induced high-velocity gradients during the load acceptance from BEP to HL was observed to develop a vortex core in the draft tube. During startup and shutdown, the guide vanes angular position was moved from one to another steady state condition to achieve the minimum load condition of the turbine. At this condition, the generator of the turbine was magnetized at the synchronous speed during startup and shutdown, respectively. The frequency of wave propagation was observed to vary with the runner angular speed during startup and complete shutdown of the turbine. Comparatively high-pressure fluctuations in the draft tube were observed during the guide vane movement from the high discharge conditions. Some unsteady phenomena such as the formation of dead velocity zone, backward flow, and flow oscillations were observed during startup and shutdown of the turbine. The current work has been also used to continue a series of workshops, i.e., Francis-99. The first workshop was held on December 2014 with the cooperation of LTU and NTNU. The measurements performed in this work were used for the second workshop which was held on December 2016. The investigations presented in this thesis will be further explored in the third workshop scheduled for December 2018.

ROPES : an expert system for condition analysis of winder ropesWilliamson, Lance K January 1990 (has links)
Includes bibliographical references. / This project was commissioned in order to provide engineers with the necessary knowledge of steel wire winder ropes so that they may make accurate decisions as to when a rope is near the end of its useful life. For this purpose, a knowledge base was compiled from the experience of experts in the field in order to create an expert system to aid the engineer in his task. The EXSYS expert system shell was used to construct a rule-based program which would be run on a personal computer. The program derived in this thesis is named ROPES, and provides information as to the forms of damage that may be present in a rope and the effect of any defects on rope strength and rope life. Advice is given as to the procedures that should be followed when damage is detected as well as the conditions which would necessitate rope discard and the urgency with which the replacement should take place. The expert system program will provide engineers with the necessary expertise and experience to assess, more accurately than at present, the condition of a winder rope. This should lead to longer rope life and improved safety with the associated cost savings. Rope assessment will also be more uniform with changes to policy being able to be implemented quickly and on an ongoing basis as technology and experience improves. The program ROPES, although compiled from expert knowledge, still requires the further input of personal opinions and inferences to some extent. For this reason, the program cannot be assumed infallible and must be used as an aid only.

The first part of this thesis gives an overview at the project and presents an analysis of the environment and the conditions in which the project will be realized. The work is a project of a component that allows lift transform rollers skewing. Three possible ways of construction are introduced. Their advantages and disadvanatages are mentioned. The best option was selected and checked by FEM. Prestressed bolted connection was also worked out. In the conclusion of the paper there is a source code of the computer programme for maximum rollers skew calculation.

The aim of the master's thesis is to provide the analysis of the composition of towing used by flying clubs in the Czech Republic. Another part of the analysis is the comparison of home towing types with types used abroad. Another aim is to analyse legislative and operating demands on towing types used in the Czech Republic and process procedures of safety and effective towing. Based on these findings the author formulates economic and operating requirements which an ideal tow plane should meet and compare these requirements with attributes available on the market.
