Caracterização morfológica e molecular de Strongyloides ophidiae (nematoda, strongyloididae) parasitas de serpentes

A primeira foi descrita em serpente Mastigodryas bifossatus e a segunda em lagartixa Hemidactylus mabouya. Na descrição de S. ophidiae apenas a fêmea partenogenética foi caracterizada em 1929 e, desde então, nenhum outro relato sobre esta espécie foi publicado. Estudos moleculares foram realizados com o objetivo de contribuir para a caracterização de Strongyloides spp. foram realizados, porém estes não incluíram S. ophidae. No presente estudo foi realizada a caracterização morfológica e molecular de S. ophidiae coletados em serpentes da região de Botucatu, Estado de São Paulo, Brasil. De um total de 125 animais, apenas quatro encontraram-se parasitados por S. ophidiae, porém fêmeas partenogenéticas foram recuperadas de apenas um animal. Coproculturas foram realizadas para obtenção das larvas L1, L2 e L3, bem como machos e fêmeas de vida livre, os quais foram morfologicamente analisados. Os produtos da PCR apresentaram 350 pb e as seqüências de nucleotídeos (número de acesso Genbank EU287935) isoladas deste nematódeo de Oxyrhopus guibei apresentaram 98% de identidade com Strongyloides procyonis (número de acesso AB272234.1 e AB205054.1) e 97% de identidade com Strongyloides cebus, Strongyloides stercoralis, Strongyloides sp. “ex snake” e Strongyloides fuelleborni (Genbank AB277236.1, AF279916.2, AJ417031.1 e AJ407030.1, respectivamente). A análise filogenética das espécies da família Strongyloididae e Rhabdiasidae demonstrou que, embora apresente 97% de similaridade com outras espécies, S. ophidiae encontra-se isolado em um ramo da árvore filogenética, caracterizando uma identidade diferente em relação a outras espécies analisadas. O presente estudo caracterizou morfológica e molecularmente S. ophidiae, parasita de serpentes da região de Botucatu, São Paulo, Brasil. / In Brazil the genus Strongyloides includes two reptile parasite: S. ophidiae and S. cruzi. The former was described in Mastigodryas bifossatus snake and the other in Hemidactylus mabouya gecko. In the description of S. ophidiae just the partenogenetic females was characterized in 1929 and, ever since, no other report about this species was published. Molecular studies for the characterization of Strongyloides spp. were reported, however these did not include S. ophidae. In the present study the morphological and molecular characterization of S. ophidiae obtained from snakes in Botucatu region, São Paulo State, Brazil was accomplished. Among the 125 studied animals only four snakes were found infected by S. ophidiae, however partenogenetic females were recovered in only one animal. Fecal exams had been performed for obtaining L1, L2 and L3 larvae, as well as free-living males and females, which were morphologically analyzed. PCR products amplified from S. ophidiae partenogenetic female samples presented about 350 bp. Nucleotide sequences (Genbank accession numbers EU287935) isolates from these nematodes from Oxyrhopus guibei presented 98% identity with Strongyloides procyonis (Genbank acession number AB272234.1 and AB205054.1) and 97% identity with Strongyloides cebus, Strongyloides stercoralis, Strongyloides sp. ex snake and Strongyloides fuelleborni (Genbank AB277236.1, AF279916.2, AJ417031.1 and AJ407030.1, respectively). The analysis of the phylogenetic relationship among the species of Strongyloididae and Rhabdiasidae demonstrated that, although there are similarity above 97% with the other species, S. ophidiae was allocated in an isolated branch of the phylogenetic tree, characterizing a different identity in relation to other species analyzed. The present study contributed for the morphological and molecular characterization of S. ophidae from the Botucatu region, São Paulo State, Brazil.
