Understanding Reification in the Composition of New Concert MusicPantelidis, Christopher January 2021 (has links)
This thesis explores the relationships that exist between reification, the conceptualisation of music, and the composition of new concert music. In general terms, reification can be described as the mental process of conceiving abstract concepts as tangible and concrete things. The problem of reification in the conceptualisation of musical works is one that exists between a rock and a hard place: we rely on something like reification in order to gain any sense of meaning from our experiences of the abstract aspects of musical works, but treating the reified understanding of these aspects as if they are what makes musical works meaningful ignores the emergent and transitory aspects of our interactions with musical works as being inherently meaningful in and of themselves. Through a variety of ethnographic, phenomenological and narrative methods, this thesis aims to challenge the long-held notions of meaning construction within the field of analytical aesthetics. It also aims provide a conceptual framework with which composers can use to practically study, collect data and analyse the conceptualisation of meaning in their compositions, as well as apply this understanding to audiences’ conceptualisation of meaning in musical works. The results of this thesis project culminate in the creation and presentation of three artistic outcomes: a new and original work for chamber ensemble, and two audio papers that explore the synthesis of interview responses and recordings of musical works within the framework of an audio-based discourse. Semi-structured interviews in the form of stimulated recall sessions were conducted as a means of obtaining data from participants about their interpretation of meaning in musical works. The use of metaphors as a conceptual framework with which to code and analyse data collected from these sessions allowed for the linguistic conceptualisation of musical meaning that avoided the theory construction and analytical aesthetic tendencies of modern music philosophy. This conceptual framework was also applied in the coding and analysis of a compositional perspective of the new work in order to compare and contrast the similarities and differences between composers and audiences in their interpretations of musical meaning. While this thesis does not purport to offer a “theory” for the conceptualisation of meaning in musical works (such an exercise is both impossible and pointless), the findings presented in the analysis of the new composition and audio papers allows for a conceptual framework that takes into account the everyday use of language in communicating the experiences of composers and audience members in their conceptualisation of meaning in musical works.

HAND I HAND : En studie av abstrakt och konkret kommunikation i textilslöjd / HAND IN HAND : A study of abstract and concrete communication in textile industryKlingström Signorelli, Luisa January 2024 (has links)

Ultrafast Raman Loss Spectroscopic Investigations of Excited State Structural Dynamics of Bis(phenylethynyl)benzene and trans-StilbeneMallick, Babita January 2017 (has links) (PDF)
The subject of this thesis is the design and development of a unified set up for femtosecond transient absorption and ultrafast Raman loss spectroscopy and demonstrate its potential in capturing the ultrafast photophysical and photochemical processes with excellent time and frequency resolution. Ultrafast spectroscopy has been serving as a powerful tool for understanding the structural dynamical properties of molecules in the condensed and gas phase. The advent of ultrashort pulses with their high peak power enables the laser spectroscopic community to study molecular reaction dynamics and photophysics that happen at extremely short timescales, ranging from picosecond to femtosecond. These processes can be measured with extremely high time resolution, which helps to resolve the under-lying molecular process. But in order to understand the global mechanism of the underlying molecular processes, we have to resolve the nuclear dynamics with the proper frequency resolution. However, achieving both, time and frequency resolutions simultaneously is not possible according to the Heisenberg uncertainty principle. Later, this limitation was overcome by femtosecond stimulated Raman spectroscopy (FSRS), a third order non-linear Raman spectroscopy. In this thesis we introduced the ultrafast Raman loss spectroscopic (URLS) technique which is analogous to FSRS, offering the modern ultrafast community to resolve molecular processes with better signal-to-noise ratio along with proper time and frequency resolution. We demonstrate the experimental procedure including the single shot detection scheme to measure whitelight background, ground state Ra-man, transient absorption and transient Raman in shot-to-shot detection fashion. URLS has been applied to understand the excited state planarization dynamics of 1,4-bis(phenylethynyl)benzene (BPEB) in different solvents. In addition, excitation wavelength dependent conformational reorganization dynamics of different sub-sets of thermally activated ground state population of BPEB are also discussed. Using the same techniques along with femtosecond transient absorption, we demonstrate the ultrafast vibrational energy transfer and the role of coherent oscillations of low frequency vibrations on the solution phase photo-isomerization of trans-stilbene from an optically excited state. The effects of solvents on the coherent nuclear motion are also discussed in the context of reaction rates.

Promoting social presence in a social networking environment in a Kuwaiti higher education contextAlshuaib, Anwar January 2014 (has links)
Recently, the numbers of Higher Education institutions that are using Web 2.0 technologies and social networking sites are increasing dramatically. These sites offer unique and diverse learning opportunities. There is evidence that a sense of community can be created online and that this community is connected with perceived learning. Garrison, Anderson and Archer (2000) introduced and developed the Community of Inquiry framework as a dynamic process model and a comprehensive framework to guide the research and practice of online learning communities, and to describe and measure elements supporting the development of these communities. This framework consists of three elements - social, teaching and cognitive presence - as well as categories and indicators to define each presence and guide the coding of transcripts. The categories of social presence are affective responses, open communication and group cohesion. The categories of teaching presence are instructional design and organisation, facilitating discourse and direct instruction. Previous studies suggest that a positive social climate on an online learning community is important as it can improve learning experience and cognitive presence. This study aims to explore and understand the nature of Community of Inquiry presences, in particular teaching presence and social presence. The aim of the study is to understand the influence of a different teaching presence on students’ development of social presence. This study provides a more comprehensive picture of developing students’ social presence over changing teaching presence in a social network environment in a Kuwaiti higher education context. In order to achieve the purpose of this study, the following research questions are explored: 1- How does a students’ sense of social presence change with a different teaching presence in the social network environment within a Kuwaiti higher education context? a. Does a students’ sense of social presence (affective responses, open communication and group cohesion) change as a result of a different teaching presence (facilitating discourse - direct instruction) in the social network environment in a Kuwaiti higher education context? b. Why do participants maintain or change their social presence level during the course? 2- How does the use of teaching presence promote the development of students’ social presence in a Kuwaiti higher education context within the social network environment? The study was conducted in the second semester of 2010/2011 at The Education Technology Department - The College of Basic Education - The Public Authority for Applied Education and Training (PAAET), within the State of Kuwait. The study was carried out on the Educational Communication module and involved 46 male participants. This study uses the equivalent of mixed methods design to answer research questions. The sequential explanatory strategy is embedded within an applied quasi-experimental approach. Quantitative data is collected and analysed, which is then followed by the collection and analysis of qualitative data. The researcher used a messages analysis and a content analysis approach to reveal the level of social presence in an online community and then develop stimulated recall interview questions. A combination of individual interviews and focus group interviews were used. Garrison et al.’s (2000) social presence coding schemes were developed to make them more suitable in the study context. Quantitative and qualitative data show that there is no significant difference between the effects of facilitating discourse and direct instruction in terms of students’ social presence level. Stimulated recall interviews reveal that most participants cannot distinguish between facilitating discourse and direct instruction. The participants believe that facilitating discourse and direct instruction are similar. The participants’ responses lead the researcher to search for other motives that could promote the development of students’ social presence in the higher education context in the social network environment. The study concludes that there are two factors that could promote the development of students’ social presence. First, instructional design and organisation, such as web design satisfaction, network effect, instructor responsiveness, the nature of the task and awarding degrees. Second, learner-specific matters, such as previous experience, peer influence, friendship, attitude, self-esteem and self-confidence and something I refer to as the Wave Effect.

Hepatitis C infection is a worldwide health problem since the risk to develop a persistent infection is relatively elevated (40 to 60%) and nearly half of the infected patients do not respond to the classical anti-HCV therapy based on a combination of PEG-IFNα and ribavirin. Viral persistence is based on powerful evasion strategies of the host’s innate immune system. In our study, we characterized antiviral response in primary human normal and chronically HCV-infected hepatocytes, a cutting-edge in our field due to the difficulty to isolate this particular cell type. In order to better define the antiviral response in freshly isolated human primary hepatocytes, we stimulated these cells with extracellular and intracellular dsRNA to trigger TLR3/TRIF and RIG-I-MDA5/CARDIF-mediated antiviral signaling pathways. By using qRT-PCR and microarray analysis, we report that both detection pathways are functional in normal human hepatocytes, their activation leading to the expression of both common (IFIT1, OASL, ISG15 and CXCL10) and specific genes (IL28A, IL28B and IL29), these last ones being a signature of the intracellular dsRNA-mediated pathway. HCV NS3/4A plays a key role in the viral polyprotein processing and upon viral RNA detection by interfering with the host’s antiviral signalling cascades. We report that major antiviral genes induction following activation of RIG-I mediated pathway are severely impaired in ectopically NS3/4A expressing normal hepatocytes due to CARDIF cleavage, but can be restored by specific NS3/4A inhibitor BILN2061. Our microarray analysis also revealed a role for NS3/4A following TRL3-mediated pathway activation on regulation of apoptosis and programmed cell death, which could be linked to strategies for the virus to persist in its host. Despite HCV strategies to circumvent the host’s immune defense system, we observed significant upregulation of ISGs and chemokines in liver biopsies and corresponding isolated hepatocytes from chronically HCV-infected patients. However, no type I and III interferon, neither key-antiviral genes (e.g., CCL5) were detected, underlying an ongoing –but inefficient- antiviral response unable to eradicate the virus. Moreover, we obtained significant inverse correlations between ISGs mRNAs and viral RNA in addition to CARDIF decrease, clearly unravelling efficient viral interfering strategies in a context of chronic HCV infection. This sustained -albeit incomplete- hepatic innate immune response is certainly associated to the failure of the classical IFN-based therapy in half of the infected patients and to the chronic inflammation causing liver damages and eventually leading to hepatocarcinoma which is often observed at late stage of the disease.

Monoacylglycerol as a metabolic coupling factor in glucose-stimulated insulin secretionZhao, Shangang 12 1900 (has links)
Insulin secretion by the pancreatic b-cell in response to post-prandial increase in blood glucose levels is an essential physiological process that is governed by cellular, nutritional and pathological factors. Other fuels including amino acids like leucine and glutamine and also fatty acids contribute to further augment insulin secretion. Failure to secrete adequate amount of insulin according to the changing demands of the body by b-cell is a key determinant of diabetes. The role played by the elevated Ca2+ influx and K+-ATP channels in insulin secretion is well known. Even though the precise mechanism of the lipid amplification of insulin secretion and the involved molecular signals are not clear, Glycerolipid/Free fatty acid (GL/FFA) cycle and its lipolytic segment have been recognized as essential components in the lipid amplification pathway of insulin secretion. Diacylglycerol produced by lipolysis was proposed as an important lipid amplification signal. However, hydrolysis of triglycerides and also of diacylglycerols is shown to be essential for glucose stimulated insulin secretion (GSIS), indicating a possible role for monoacylglycerol (MAG) in this process.
In the present study we demonstrate that the obliterated GSIS due to lipolysis inhibition in b-cells can be restored by providing exogenous MAG. In the b-cells MAG levels increase significantly in the presence of high glucose concentration and specific inhibition of the major MAG hydrolase, abhydrolase-6 (ABHD6), in b-cells and islets with WWL70 leads to accumulation of MAG with concomitant increase in insulin secretion. Lipidomics analysis indicated that the major MAG species that is elevated by high glucose as well as WWL70 addition is 1-stearoylglycerol (1-SG). Exogenously added 1-SG and also 1-palmitoylglycerol (1-PG) strongly enhanced GSIS and this augmentation is not dependent on the generation of FFA by these MAGs. This indicates that MAG is a potential candidate for being the lipid signal for GSIS amplification. Further evidence for this was provided by the observation that overexpression of the MAG hydrolase ABHD6 in INS832/13 cells, resulted in decreased insulin secretion, probably owing to the lowered MAG level inside the b-cells.
Pharmacological studies using AMG9810, a specific antagonist of transient receptor potential vanilloid-1 (TRPV1) receptor that binds MAG, revealed that a blockade of TRPV1 strongly attenuated the MAG-augmented insulin secretion. Since MAG is a potential activator of TRPV1, it is likely that MAG binds on the inner surface of the cell membrane to TRPV1, which in turn triggers rapid influx of Ca2+ thereby promoting insulin granule exocytosis. Thus, AMG9810 was found to lower Ca2+ influx into dispersed rat islet cells that was induced by high glucose and also WWL70.
These results collectively suggest that MAG is the potential mediator of the lipid amplification of glucose-stimulated insulin secretion. Our results also indicate that pharmacological intervening at the ABHD6 hydrolysis step enhances insulin secretion; this enzyme protein can be a promising thrapeutic target for the development of anti-diabetic drugs that promote insulin secretion.

Distribuce mitochondriálních odpřahujících proteinů ve vybraných tkáních myši a potkana / Distribution of mitochondrial uncoupling proteins in selected tissues from mice and ratAlán, Lukáš January 2010 (has links)
Mitochondrial uncoupling proteins (UCPs) belong to the superfamily of mitochondrial anion-carriers. The longest known is UCP1, predominantly expressed in brown adipose tissue, where it takes part in nonshivering thermogenesis. In the late 1990s were discovered other sequence homologs of UCP1 with tissue specific distribution. The Function of these "new" uncoupling proteins is still uncertain. It is assumed that each of the isoforms has a specific function depending on the type of tissue. This thesis showed differences in tissue transcription pattern between rat and mice using RT-PCR absolute quantification. Significant differences in pattern were found in lungs, brain and muscle. In each case UCP expression was higher in mice tissues. Mice lungs express mainly UCP2. The difference in mice brain is caused by ucp4 and ucp5 genes transcription and finally in muscle is highest content of UCP3 mRNA. We investigated whether any of ucp transcript can complement ucp2 transcripton in spleen or lungs of ucp2 -/- mice. We did not find any difference which can explain, that in isolated lung mitochondria of fasted ucp2-/- mice were uncoupled in state 4. In the last project, we found relationship between ucp2 transcription in insulinoma INS-1E cells and oxygen levels of the cultivation atmosphere.

Optically stimulated luminescence (OSL) ages were obtained in eolian deposits from the east coast of Maranhão, Brazil. OSL dating allowed the identification of at least four generations of eolian dunes, including the active dunefields. The two oldest generations (G1/G0: OSL ages 258.9±24.6 to 88.0±8.6 ky B.P.) are not distinguishable by field criteria, only geochronological data. Generation 2 corresponds to eolian features stabilized by vegetation that occurs up to 135 km from the current coast line. OSL-SAR ages of stabilization of these features are between 19 and 14 ky B.P., a narrow time interval during Pleistocene. This event is related to the climatic event HS-1. During this period the intensity of trade winds decreased and precipitation increased in the area due to a greater influence of the Intertropical Covergence Zone, which was shifting south. These climatic variations reduced the capacity of eolian transportation and the led system to to stabilization. Provenance analyzes by heavy minerals identified the continental shelf located east of the Parnaiba river mouth as sedimentary source for the east coast of Maranhão, which is fed by longitudinal litoral drift to west. Parnaiba river apparently has a minor contribution as sedimentary source for recent and past eolian generations. Sedimentary dynamic of the recent eolian system, as indicated by heavy minerals analyzes, grain size analyzes and morphoplanimetric parameters, is characterized by strong reworking by rivers, braided intermittent drainages, that cross the dunefield. All analyzed Brazilian sediments presented relatively high OSL sensitivity and good behavior regarding their luminescence characteristics relevant for radiation dose estimation. The OSL sensitivity of quartz was identified as the main characteristic interfering the OSL dating quality. We propose to standardize the definition of quartz OSL sensitivity using the Quartz-to-\'Al IND.2\'\'O IND.3\':C sensitivity ratio (\'delta\'\'POT.s\'Q). The standardization is a prerequisite to compare data obtained under different measurement conditions and to investigate and develop geological applications of quartz OSL sensitivity. We observed that the major factor controlling the OSL sensitivity is the sedimentary history of the sediment. Longer sedimentary transport provided by huge drainage systems and long-distance littoral drift increases the OSL sensitivity. The use of OSL sensitivity as provenance proxy was successfully applied at Ilha Comprida-SP to analyze climate variations.

The Volumetric Modulated Arc Therapy (VMAT) is an advance technique of Intensity Modulated Radiation Therapy (IMRT). This progress is due to the continuous gantry rotation with the radiation beam modulation providing lower time of the patient treatment. This research aimed the verification of the isodose curves in a simulation of a vertebra treatment with spinal cord protection using the thermoluminescent (TL) and optically stimulated luminescence (OSL) dosimetry techniques and theLiF:Mg,Ti (TLD-100), CaSO4:Dy and Al2O3:C dosimeters and LiF:Mg,Ti microdosimeters (TLD-100). The dosimeters were characterized using PMMA plates of 30x30x30 cm3 and different thickness. All irradiations were done using Truebeam STx linear accelerator of Hospital Israelita Albert Einstein, with 6 MV photons beam. After the dosimeter characterization, they were irradiated according the specific planning simulation and using a PMMA phantom developed to VMAT measurements. This irradiation aimed to verify the isodose curves of the treatment simulation using the two dosimetry techniques. All types of dosimeters showed satisfactory results to determine the dose distribution but analysing the complexity of the isodose curves and the proximity of them, the LiF:Mg,Ti microdosimeter showed the most appropriate for use due to its small dimensions. Regarding the best technique, as both technique showed satisfactory results, the TL technique presents less complex to be used because the most of the radiotherapy departments already have a TL laboratory. The OSL technique requires more care and greater investment in the hospital.
