The objective of this thesis is to analyze the impact of the electronic records of sales (ERS) implemented on December 1, 2016, in the Czech Republic. The ERS orders given entrepreneurs to use special cash boxes and to send data about single sales to the Financial Authority immediately after each transaction. Experience shows that the ERS reduced tax evasion and the shadow economy. This thesis fills a gap in literature as no study has yet examined the impact of ERS on macroeconomic variables - gross domestic product (GDP) per capita, unemployment rate and harmonized consumer price index (HCPI). To analyze it, I apply SCM by Abadie and Gardeazabal (2003), Abadie et al. (2010) and Abadie et al. (2015) that proposes the way how to evaluate the impact of political interventions using a quantitative approach. Using data from the Eurostat and OECD databases from Q1 2004-Q3 2019, the method allows me to construct a counterfactual outcome for GDP per capita, unemployment rate and HCPI in the absence of the implementation of the ERS. I also apply the extension of original SCM - generalize synthetic control method by Xu (2017) and augmented synthetic control method by Ben-Michael et al. (2018) as robustness checks. From technical standpoint Im interested in question of bidirectional remediation of new media and painting and how its evolution supports ilussion and imagination. In ontologiacal way im looking for mystical nature of picture. Final exhibition is an metaphor composed in hybryd picture based on technological and theoretical experiences in Painting. The result is little synthetic reality.

The theoretical part of the thesis specifies the reading disorders and the methods of work which lead to the succesful training of the reading abilities. The practical part of the thesis subsumes working with children of the small school "ZŠ Josefa Suka" and the kindergarten "MŠ Křečovice,preparing materials for teaching reading to either children without any learning disabilites or those with disabilites and verifying the materials in practical training.

This thesis deals with the issue of genetic method to reading in the first grade of an elementary school. The theoretical part is focused on the child's entry to school, their school maturity and further education. Other methods to read are presented, analysed and compared with the genetic method as well. Furthermore, the methods and ways how to put the genetic method in practice are pursued in detail along with didactical games and examples of teaching procedures. The practical part looks into the experience the teachers have gained with this method, the school success of the children and the opinion the family members have.

In diploma thesis named "Comparison of the practicing methods of initial reading and writing in relation to the specific learning disorders" we are trying to find out if the implementation of one of three methods of teaching to read and write - analytic synthetic method, genetic method or Sfumato eliminates the development of specific learning disabilities - dyslexia, disgraphia and disorthographia. The first part of work looks into theoretical outcomes of each method of teaching to read and write and defines specific learning disabilities. In the second part of work methods of teaching to read and write depending on specific learning disabilities development are compared and evaluated based on results of questionnaires, interviews and observation.

The main theme of this thesis is available and potential gaseous and liquid alternative biofuels made from biomass and waste. The thesis deals with their detailed description and comparison. The first part covers the basic distribution of biofuels and alternative fuels made from waste. The main part focuses then on the fuels themselves, their properties, production, use and environmental impact. Furthermore, thesis describes legislative issues and fuels are compared from different perspectives. Practical part includes testing of combustion of liquid fuels taken from waste sources. In the next chapters there is executed overview of basic atomization method of liquid fuels and a plan and running of the testing processed. Evaluation of results is based on point of view of suitability for use, the quality of combustion and emission limits.

This diploma thesis is focused to the writing skills in an initiatory education of a literacy during the first grade at a primary school. The thesis is divided into the theoretical and the practical part. The theoretical part presents the genesis of the handwriting during the history, the physical and psychical expectations for the assumption of the writing skills, the methodics of two learning methods which are used in the czech education system most frequently and last but not least a list of the evaluated scales of handwriting. In the practical part we present the evaluation scale which were constructed for the purpose of this thesis. This scale monitors a plot of the written children's products, an error rate, an ability of corections and a graphical part. We kept at disposal about 500 written children's products which were collected from 22 classes from 9 schools from three parts of Czech Republic (Prague, Central and South Bohemian Region and Moravia). Children which were involved to the project, they were educated by the analytic-synthetic method or the genetic metod for the education of the literacy. Diabetes mellitus (DM) is a severe and frequent disease with increasing prevalence. It is not possible to achieve long term cure without late complications. Recent advances in cell fate modifications open a pathway to alternative cell therapies for DM cure. My doctoral thesis "Differentiation of pancreatic stem cells into insulin producing β- cells" is focused on the development of a new source of insulin secreting cells for transplantation. Combinatorial testing of numerous potential transcription factors and epigenetic modifiers resulted in a final protocol for the reprogramming pancreatic of exocrine cells into insulin secreting cells. The key transcriptional factors TF (Pdx1, Ngn3 a MafA) were applied in the form of synthetic mRNA. In four independent experiments we applied transcriptional factors in a specific sequence, thus obtaining 14.3 ± 1.9 % insulin positive cells. When challenged in vitro by the glucose levels of 2.5 and 20 mmol/l glucose, respectively, these cells exhibited glucose-sensitivity of insulin secretion (842 ± 72 and 1 157 ± 58 pg insulin/µg DNA/ml, n=5). They also demonstrated a sensitivity of insulin secretion (863 ± 78 and 1 025 ± 66 pg insulin/µg DNA/ml, n=5) to the concentration of depolarization agent KCl applied at 0 and 30 mmol/l, respectively together with 2.5...

Mediální reprezentace fenoménu deepfakes / Media representation of deepfakeJanjić, Saška January 2022 (has links)
This master thesis explores the media representation of deepfakes. The first part summarizes previous research followed by a comprehensive review of deepfakes, including the technology allowing for their emergence, current uses and methods of regulation and detection. The second part connects the phenomenon with important theoretical concepts such as social construction of reality and the crucial role of media in this process. The empirical part consists of research combining two methods - quantitative content analysis and qualitative critical discourse analysis. The research analysis is focused on media articles dealing with deepfakes in order to find out how the media represent this phenomenon. The results show that current media discourse of deepfakes is strongly negative as the media frame them as a security threat. This negative representation is highly speculative since journalists often invent their own stories of future disastrous consequences of the technology for national security due to lack of current examples. The findings show an apparent hierarchy of the harms posed by deepfakes which is present in media coverage, and reflects gender sereotypes and inequality in the current society.
