The Role of Pax3 in Neuronal Differentiation of the Ophthalmic (OpV) Trigeminal Placode and Neural Tube during Chicken Embryonic Development

Pax3 has been used as a valuable marker in research aimed at understanding tissue interactions involved in trigeminal ophthalmic (opV) placode development. While Pax3 expression coincides with opV neuron specification, the function of Pax3 in these cells has not previously been investigated. Splotch mutant mice (which lack Pax3) have a reduced trigeminal ganglion; however it is not clear whether this reduction is due to neural crest or placode cells. We have used electroporation in the chick model system to block or ectopically express Pax3 at key times in opV placode development. Using several markers of placode cell differentiation, we have determined the experimental effects manipulating Pax3. Blocking placodal Pax3 with gene specific morpholinos resulted in a loss of migratory placode cells, and a downregulation of all opV placode markers in targeted cells. Ectopic expression of Pax3, either within the placode domain or in adjacent cranial ectoderm, resulted in the upregulation of some but not all placode markers. We conclude that opV placodal Pax3 expression is required for normal placode cell development, and hypothesize that its expression must be tightly regulated in order for placode cells to fully differentiate. The precise role of Pax3 and Pax7 in the restriction and differentiation of dorsal interneuron progenitors has been difficult to assess due to the many additional factors involved in specification and patterning of the neural tube. We have used electroporation in the chick model system to ectopically express Pax3 and Pax7 unilaterally in the neural tube. Using several markers for differentiation of ventral and dorsal neuronal progenitors, we have experimentally determined the effects of Pax3 and Pax7 ventrally and dorsally. Ectopic expression of these transcription factors in the ventral neural tube resulted in the loss of motorneurons. Though mis-expression did not qualitatively affect commissural neurons as assayed by neurofilament staining, ectopic expression of Pax3 and Pax7 in the dorsal neural tube stopped dorsal interneuron progenitors from differentiating. We conclude that Pax3 and Pax7 expression is sufficient to restrict ventral neuron identity. We also hypothesize that downregulation of these transcription factors in the dorsal neural tube is required for normal dorsal interneuron differentiation.

Development of a Frost Heave Test Apparatus

Frost heave damage to roadways costs millions of dollars every year. The need for an improved understanding of the fundamental mechanisms associated with frost heave and methods for efficiently improving frost-susceptible materials prompted the Department of Civil and Environmental Engineering at Brigham Young University (BYU) to undertake a project to design, construct, and verify the functionality of a new frost heave testing apparatus. Frost heave research has been carried out for more than 75 years. The equipment used to conduct this testing has advanced in accuracy and utility over the years. To establish a background in past and current frost heave research, a survey of 12 frost heave devices, including their construction and capabilities, was performed in this research. Several design objectives were then delineated, and a nine-specimen frost heave device was designed and constructed to meet the specifications. The apparatus uses one collective heat source and one collective heat sink for all nine specimens. Heave data and temperature data are collected electronically, while the weights of the specimens before and after frost heave testing are measured manually. Preparatory tests were conducted to confirm the functionality of the data acquisition systems, the uniformity of conditions experienced by all specimens, and the replication of natural roadway freezing conditions. Once preparatory testing was complete, a full-scale frost heave test was performed using the apparatus to investigate the efficacy of cement stabilization in reducing the frost susceptibility of a Montana silt and to validate the functionality of the finished device. Results from the testing indicate that adding 2.0 percent cement actually induces frost heave in excess of that exhibited by the untreated soil. However, additions of 3.5 percent and 5.0 percent were found to be effective in preventing frost heave. Although minor, recommendations for further improvements to the frost heave apparatus include provisions to further decrease the thermal gradient across the specimen and installation of an automatic temperature control device for the water source.

Development of a Multiple Microphone Probe Calibrator

This paper presents the theory, design, and validation of a microphone calibrator used to calibrate multiple microphones simultaneously. This work was done in conjunction with the development of an acoustic energy density probe, which was used to validate the calibrator. The probe uses multiple microphones to acquire the data needed to compute the acoustic energy density. The probe microphones are 0.006 m diameter electret microphones which typically do not have an ideal “flat" response over a wide range of frequencies as compared to precision condenser microphones. The probe microphone characteristics prompted the need for simultaneous, multi-microphone magnitude calibration. The idea behind the calibration process was to simultaneously subject each microphone on the probe to the same known acoustic pressure over the frequency range of the probe (0-2 kHz). This is done using equal-length small-diameter tubes connected to a single source at each microphone. The calibrator was modeled using and equivalent circuit model. The model results are presented and compared to measured results. The calibrator was validated to result in the same pressure along individual paths to each microphone simultaneously. Test results show that the calibrator can calibrate each probe microphone within ± 0.5 dB up to 2000 Hz, and within ± 1 dB up to 4900 Hz with a confidence level of 95%. Directivity tests were performed to further characterize the probe. Using a correction factor the probe is shown to be directionally independent.

Wnt5a Signaling Independently of the Planar Cell Polarity Pathway Resulting in Convergent Extension and Neural Tube Closure During Vertebrate Development

Vertebrate development is regulated by cellular communication by mechanisms of cell fate and cell behavior. These crucial mechanisms are regulated by cellular signaling and in the case of cell fate, cellular signaling results in transcription of developmentally important genes. Communication between cells can also result in regulation of cell behavior by acting on cytoskeletal elements rather than nuclear factors. One of the cellular signals that regulate both cell fate and cell behavior is the family of Wnt signaling molecules. Wnt5a is one of 19 Wnt molecules and has been previously demonstrated to play critical roles in many important processes in embryonic development as well tumor suppression. Despite many studies that lend credence to a pathway that regulates cell behavior for Wnt5a rather than cell fate, the identity of the pathway(s) Wnt5a impinges upon remains unclear. Despite the possibility of Wnt5a signaling through multiple pathways, here, focus is given to the non-canonical Wnt signaling pathway, a pathway that regulates cell behavior, also known as the Wnt/Planar Cell Polarity (PCP) pathway. The involvement of Wnt5a in the Wnt/PCP pathway was demonstrated with a genetic approach: crossing Wnt5a heterozygous mice with mice heterozygous for a component of the Wnt/PCP pathway to uncover genetic interactions in vivo. Hence, Wnt5a X Looptail (Lp) (Wnt/PCP) heterozygous crosses have been performed. Double heterozygotes for this intercross did not exhibit a decrease in viable progeny as compared to the decreased numbers of Lp heterozygotes. These observations demonstrated a lack of genetic interaction between Wnt5a and the PCP pathway. Wnt5a mutants possess phenotypes associated with deficits in the Wnt/PCP pathway, namely convergent extension (CE) defects and neural tube closure defects. However, upon further investigation of the increased penetrance of craniorachischisis in Wnt5a-/-;Lp+/-, Wnt5a mutants do not display the characteristic broadening of the neural floor plate commonly associated with Lp-/-. This supports that Wnt5a and PCP signaling are parallel pathways that have converged to regulate different aspects of CE and neural tube closure. Despite the complexity of Wnt5a and its potential involvement in multiple pathways, dissection of this will explain the broad range of phenotypes observed.

Buckling Failure Boundary for Cylindrical Tubes in Pure Bending

Bending of thin-walled tubing to a prescribed bend radius is typically performed by bending it around a mandrel of the desired bend radius, corrected for spring back. By eliminating the mandrel, costly setup time would be reduced, permitting multiple change of radius during a production run, and even intermixing different products on the same line. The principal challenge is to avoid buckling, as the mandrel and shoe are generally shaped to enclose the tube while bending. Without the shaped mandrel, buckling will likely occur sooner, that is, at larger bend radii. A test apparatus has been built for arborless bending. It has been used to determine the limits of bend radius, wall thickness, material properties, etc. on buckling. Key to the process is a set of moveable clamps, which grip the tube and rotate to produce the bend. A complex control system moves the clamps radially to maintain pure bending, without superimposing tension or compression. A series of tests were performed to document the safe region of operation to avoid buckling. Charts have been created to assist the operator, as well as the design engineer, in determining the minimum bend radius. Similar tests will be required for each additional tube size, thickness, material, etc.

In patients with a functional gastrointestinal (GI) tract, enteral feeding is preferred over parenteral feeding as it has fewer complications and a relatively lower cost. Nasogastric and nasoenteric feeding tubes are available options but when long-term enteral feeding is desired, a percutaneous endoscopic gastrostomy (PEG) tube is more convenient. PEG tube can be associated with multiple complications; however, its displacement which causes gastric outlet obstruction (GOO) is a rare one. Here we present a case of an 81-year-old woman with dementia who presented with upper GI bleeding and was found to have GOO causing reflux esophagitis due to PEG tube displacement.

An Investigation Of The Autoignition Of Power Generation Gas Turbine Fuel Blends Using A Design Of Experiments Approach

Natural gas has grown in popularity as a fuel for power generation gas turbines. However, changes in fuel composition are a topic of concern since fuel variability can have a great impact on the reliability and performance of the burner design. In particular, autoignition of the premixed fuel and air prior to entering the main burner is a potential concern when using exotic fuel blends. To obtain much-needed data in this area, autoignition experiments for a wide range of likely fuel blends containing CH4 mixed with combinations of C2H6, C3H8, C4H10, C5H12, and H2 were performed in a high-pressure shock tube. However, testing every possible fuel blend combination and interaction was not feasible within a reasonable time and cost. Therefore, to predict the surface response over the complete mixture domain, a special experimental design was developed to significantly reduce the amount of 'trials' needed from 243 to only 41 using the Box-Behnkin factorial design methodology. Kinetics modeling was used to obtain numerical results for this matrix of fuel blends, setting the conditions at a temperature of 800 K and pressure of 17 atm. A further and successful attempt was made to reduce the 41-test matrix to a 21-test matrix. This was done using special mixture experimental techniques. The kinetics model was used to compare the smaller matrix to the expected results of the larger one. The new 21-test matrix produced a numerical correlation that agreed well with the results from the 41-test matrix, indicating that the smaller matrix would provide the same statistical information as the larger one with acceptable precision. iii After the experimental matrix was developed using the design of experiments approach, the physical experiments were performed in the shock tube. Long test times were created by "tailoring" the shock tube using a novel driver gas mixture, obtaining test times of 10 millisecond or more, which made experiments at low temperatures possible. Large discrepancies were found between the predicted results by numerical models and the actual experimental results. The main conclusion from the experiments is that the methane-based mixtures in this study enter a regime with a negative temperature coefficient when plotted in Arhennius form. This means that these mixtures are far more likely to ignite under conditions frequently encountered in a premixer, potentially creating hazardous situations. The experimental results were correlated as a function of the different species. It was found that the effect of higher-order hydrocarbon addition to methane is not as profound as seen at higher temperatures (>1100 K). However, the ignition delay time could still be reduced by a factor two or more. It is therefore evident that potential autoignition could occur within the premixer, given the conditions as stated in this study.

Driver-gas Tailoring For Test-time Extension Using Unconventional Driver Mixtures

To study combustion chemistry at low temperatures in a shock tube, it is of great importance to increase experimental test times, and this can be done by tailoring the interface between the driver and driven gases. Using unconventional driver-gas tailoring with the assistance of tailoring curves, shock-tube test times were increased from 1 to 15 ms for reflected-shock temperatures below 1000 K. Provided in this thesis is the introduction of tailoring curves, produced from a 1-D perfect gas model for a wide range of driver gases and the production and demonstration of successful driver mixtures containing helium combined with either propane or carbon dioxide. The He/CO2 and He/C3H8 driver mixtures provide a unique way to produce a tailored interface and, hence, longer test times, when facility modification is not an option. The tailoring curves can be used to guide future applications of this technique to other configurations. Nonreacting validation experiments using driver mixtures identified from the tailoring curves were performed over a range of reflected-shock temperatures from approximately 800 to 1400 K, and some examples of ignition-time experiments that could not have otherwise been performed are presented.

A Study Of Syngas Oxidation At High Pressures And Low Temperatures

Ignition and oxidation characteristics of CO/H2, H2/O2 and CO/H2/CH4/CO2/Ar fuel blends in air were studied using both experimental and computer simulation methods. Shock-tube experiments were conducted behind reflected shock waves at intermediate temperatures (825 < T < 1400 K) for a wide range of pressures (1 < P < 45 atm). Results of this study provide the first undiluted fuel-air ignition delay time experiments to cover such a wide range of syngas mixture compositions over the stated temperature range. Emission in the form of chemiluminescence from the hydroxyl radical (OH*) transition near 307 nm and the pressure behind the reflected shock wave were used to monitor reaction progress from which ignition delay times were determined. In addition to the experimental analysis, chemical kinetics calculations were completed to compare several chemical kinetics mechanisms to the new experimental results. Overall, the models were in good agreement with the shock-tube data, especially at higher temperatures and lower pressures, yet there were some differences between the models at higher pressures and the lowest temperatures, in some cases by as much as a factor of five. In order to discern additional information from the chemical kinetics mechanisms regarding their response to a wide range of experimental conditions, ignition delay time and reaction rate sensitivity analyses were completed at higher and lower temperatures and higher and lower pressures. These two sensitivity analyses allow for the identification of the key reactions responsible for ignition. The results of the sensitivity analysis indicate that the ignition-enhancing reaction H + O2 = O + OH and hydrogen oxidation kinetics in general were most important regardless of mixture composition, temperature or pressure. However, lower-temperature, higher-pressure ignition delay time results indicate additional influence from HO2- and CO- containing reactions, particularly the well-known H + O + M = HO2 + M reaction and also the CO + O + M = CO2 + M and CO + HO2 = CO2 + OH reactions. Differences in the rates of the CO-related reactions are shown to be the cause of some of the discrepancies amongst the various models at elevated pressures. However, the deviation between the models and the experimental data at the lowest temperatures could not be entirely explained by discrepancies in the current rates of the reactions contained within the mechanisms. Additional calculations were therefore performed to gain further understanding regarding the opposing ignition behavior for calculated and measured ignition delay time results. Impurities, friction induced ionization, static charge accumulation, boundary layer effects, wall reaction effects, and revised chemical kinetics were all considered to be possible mechanisms for the model and measured data disparity. For the case of wall-reaction effects, additional shock-tube experiments were conducted. For the remaining effects listed above, only detailed calculations were conducted. Results from this preliminary anomaly study are at this time inconclusive, but likely avenues for future study were identified. Additional kinetics calculations showed that the large difference between the experimental data and the chemical kinetics models predictions at low temperatures can be explained by at least one missing reaction relevant to low-temperature and high-pressure experimental conditions involving the formation of H2O2, although further study beyond the scope of this thesis is required to prove this hypothesis both theoretically and experimentally.

Concrete-Filled Steel Tube Columns-Tests Compared with Eurocode 4

This paper summarises the data from 1819 tests on concrete-filled steel tube columns and compares their failure load with the prediction of Eurocode 4. The full data is given on the website http://web.ukonline.co.uk/asccs2 . The comparison with Eurocode 4 is discussed and shows that Eurocode 4 can be used with confidence and generally gives good agreement with test results, the average Test/EC4 ratio for all tests being 1.11. The Eurocode 4 limitations on concrete strength could be safely extended to concrete with a cylinder strength of 75 N/mm2 for circular sections and 60 N/mm2 for rectangular sections.
