A lesão por isquemia-reperfusão (I/R) é o mecanismo fisiopatológico central no desenvolvimento da insuficiência hepática pós-operatória. Diversas estratégias para minimizar suas consequências estão sendo desenvolvidas, mas ainda sem resultados satisfatórios. Recentemente o pré-condicionamento isquêmico remoto (PCIR), método em que ciclos breves de I/R aplicados em um órgão ou membro é capaz de atenuar os resultados da I/R em um órgão distante, vem sendo utilizado, em modelos experimentais, com resultados promissores. No entanto seu mecanismo de ação ainda não foi esclarecido. Um dos mecanismos propostos é a modulação na expressão das citocinas sintetizadas durante a resposta inflamatória que acompanha o processo de I/R. Foram utilizados 36 ratos (Rattus norvegicus), machos, com peso entre 250 e 280 g, divididos em três grupos: Grupo Sham, cirurgia simulada; Grupo IR, isquemia de 70% do fígado por 45 minutos e reperfusão; e Grupo PCIR, pré-condicionamento isquêmico remoto do fígado através de seis ciclos de isquemia-reperfusão da pata do animal, com quatro minutos de isquemia e quatro minutos de reperfusão em cada ciclo, seguido de isquemia hepática semelhante ao do Grupo IR. Terminado os procedimentos cirúrgicos, metade dos animais foi morta decorridos 60 minutos de reperfusão, e a outra metade após 180 minutos. Foi coletado tecido hepático do lobo submetido à isquemia, para estudo histopatológico, utilizando o índice de injúria hepática modificado; e sangue, para dosagem plasmática de TNF-α, IL-6, IL-10 e ALT. A análise histopatológica mostrou que a necrose celular foi significativamente reduzida no Grupo PCIR quando comparado com Grupo IR (p <0,0001). As transaminases mostraram o mesmo padrão com redução significativa dos seus valores no Grupo PCIR quando comparados com o Grupo I-R (p <0,0001). A dosagem das interleucinas mostrou redução significativa na expressão da IL-6 no Grupo PCIR quando comparado com o Grupo IR (p<0,001). Houve aumento da expressão de IL-10 nos grupo PCIR, porém não atingiu significância estatística. Não foi identificada diferença na dosagem de TNF-α nos grupos estudados. O PCI-R foi eficaz na redução na necrose celular resultante da lesão por I-R nos grupos estudados. A redução na síntese de IL-6 segue o padrão observado em outros estudos. / Ischemia/Reperfusion (I/R) injury is an important pathophysiological mechanism in the postoperative liver failure. Different strategies to minimize the I/R liver injury have been developed during the last decades but the results had been disappointing. Recently, the remote ischemic preconditioning (RIPC), a method that involves a brief ischemic episode on an organ or tissue that subsequently affords protection to a remote organ or tissue, have been use in various experimental models with promising results. The precise pathway activated by the RIPC isnt clear, but cytokine release modulation has been proposed as a candidate mechanism. Thirty-six male rats (Rattus norvegicus) were divided in 3 groups: Sham; I/R injury, a 45 minutes lobar (70%) liver ischemia and reperfusion; and RIPC, 6 cycles of 4 minutes of ischemia and 4 minutes of reperfusion of the right hindlimb followed by a 45 minutes lobar (70%) liver ischemia and reperfusion. Liver tissue in the affected lobe and blood samples were collected after 60 minutes and 180 minutes of reperfusion for histopathological study of liver I/R, plasma cytokines (TNF-α, IL-6 and IL-10) and liver aminotransferases measurement (ALT). The histopathological study demonstrated a significant lesser degree of liver necrosis in the RIPC group (p <0,001). The aminotransferases levels followed the same pattern, with significant lower levels in the RIPC group (p <0,001). The cytokines assessment showed a reduction in the expression of IL-6 in the RIPC when compared with the I/R group (p <0,01). Interleukin-10 levels were higher in the RIPC group, but the difference wasnt significant. The TNF-α measurement didnt show any difference in the groups. The RIPC model presented consistently reduced the I/R injury to the liver and the IL-6 expression was similar to the reported in other studies.

Resultado do tratamento da doença de Crohn com anti-fator de necrose tumoral alfa / Outcomes in the treatment of Crohn´s disease with anti tumor necrososis factor-alphaAnna Paula Rocha Malheiros 19 August 2008 (has links)
A doença de Crohn é uma inflamação crônica do trato gastrointestinal. O tratamento convencional é muitas vezes desapontador. Apesar da variedade de drogas disponíveis para o tratamento da doença inflamatória intestinal, tais como: salicilatos e seus derivados, corticosteróides, antibióticos e imunossupressores, nenhuma destas mostrou ser totalmente eficaz ou definitiva para o tratamento da doença e seus surtos de exacerbação. Pesquisas têm sido desenvolvidas com o objetivo de apresentar drogas mais efetivas. Dentre estas, destacam-se as drogas biológicas. O infliximabe é um anticorpo monoclonal quimérico anti-fator de necrose tumoral alfa e está indicado na doença de Crohn refratária e fistulizante. O objetivo deste estudo visa avaliar prospectivamente os resultados e efeitos colaterais precoces e tardios do uso do anti-TNF alfa no tratamento de 60 doentes com doença de Crohn, no período de julho de 1999 a dezembro de 2005. Os doentes foram tratados com anti-TNF alfa (infliximabe), na dose de 5mg/kg de peso, aplicado por via endovenosa em intervalos de dois meses. A avaliação foi realizada por protocolo clínico que classificava os quesitos: estado geral, sintomas intestinais e doença perianal em melhor, inalterado e pior, e pelo índice de atividade da doença de Cronh. Os doentes tratados com anti-TNF alfa apresentaram mediana de duração da doença de sete anos, variando de um a 28 anos entre a data do início dos sintomas e a data de início da pesquisa. 34 doentes (56,7%) já haviam sido submetidos a uma ou mais operações abdominais e 38 (63,3%) a operações orificiais. O software utilizado para a realização dos cálculos foi o SPSS® 9.0 for Windows, sendo estatisticamente significantes os testes com p<0,05. Foram aplicadas 225 doses de anti-TNF alfa, em média, 3,7 doses por paciente num período de aproximadamente cinco anos, variando de uma a 14 doses. No tratamento inicial 76% dos pacientes responderam a droga. As principais indicações para o emprego do anti-TNF alfa foram a presença de doença perianal em 36 casos (60%) e a intratabilidade clínica em 24 casos (40%). Observou-se que após a primeira dose da medicação, os doentes com mais de dez anos de doença e submetidos à operação abdominal tiveram resultado satisfatório semelhantemente aqueles doentes com menos de cinco anos de doença e não operados com p<0,05. O índice de atividade da doença de Crohn foi em média de 189,7 antes do início do tratamento e na primeira aplicação diminuiu em média para 135,4, e progressivamente ao longo das aplicações (115, 102, 109 e 88,4 até a quinta dose), sendo o resultado estatisticamente significativo. Houve efeito colateral em 40 aplicações (17,8%), sendo os efeitos principais: eritema cutâneo, dispnéia e dor abdominal. O tratamento com anti-fator de necrose tumoral alfa, obedecidas as indicações precisas, associou-se a baixo índice de efeitos colaterais graves tendo apresentado bons resultados na resolução da doença de Crohn perianal, na melhora da sintomatologia intestinal e no estado geral dos pacientes / Crohn´s disease is a chronic inflammatory disorder of the gastrointestinal tract. Conventional treatment is many times disappointing. Besides the great number of available medications to treat inflammatory bowel diseases, such as salicilates and derivatives, corticosteroids, antibiotics and immunosuppressive agents, none of them proved to be totally efficient or the ultimate treatment for inflammatory diseases and their exacerbation. Researches have been carried out to find more effective therapeutic drugs. Among these therapeutics, biologic treatments have been in evidence. Infliximab is a chimeric IgG1 monoclonal antibody against tumor necrosis factor-alpha, and is indicated for refractory luminal and fistulizing Crohns disease. The aim of this study is to prospectively evaluate the outcome, early and late adverse events, in 60 patients diagnosed with Crohn´s disease and treated with infliximab between July 1999 and December 2005. All patients were treated with anti-TNF-alpha (infliximab), 5mg/kg/dose, intravenously, each two months. Patients were clinically evaluated using a protocol that classified the evolution of the health status, intestinal symptoms and perianal disease, as better, worse or unchanged, during the treatment. Crohn´s disease activity index was also evaluated. Patients treated with anti-TNF-alpha presented a median disease duration of seven (range 1-28) years, between the beginning of the disease symptoms and the beginning of the research protocol. Thirty-four patients (56.7%) have been submitted to one or more abdominal surgeries before, and 38 (63.3%) to anal-rectum surgeries. All statistics tests were performed with computer software Statistical Package for the Social Sciences (SPSS® 9.0) for WindowsTM, and p values of less than 0.05 were considered statistically significant. Totals of 225 anti-TNF-alpha doses were administered. The mean doses administered per patient, in a period of approximately five years, were 3.7 (range 1-14) doses. After the initial treatment, 76% of the patients achieved a response. The most frequent indications for anti-TNF-alpha was perianal disease, occurring in 36 patients (60%), and clinical failure to the conventional treatment, happening in 24 patients (40%). After the first dose of anti-TNF-alpha, patients with more than 10 years of treatment and previously submitted to abdominal surgery presented a satisfactory outcome, similar to those with less than 5 years of disease and not submitted to surgery, p<0.05. Crohn´s disease activity index showed a mean index of 189.7 before treatment, that decreased to 135.4 after the first dose, and progressively decreased with the subsequent doses (means: 115, 102, 109 and 88.4, until the fifth dose, p<0.05). Adverse events were reported in 40 administrations (17,8%) from the total. The most prevalent adverse events were: rash, dyspnoea and abdominal pain. The treatment with anti-TNF-alpha, following precise indications, was associated with a low incidence of severe adverse events and presented good outcomes in the resolution of perianal Crohn´s disease, improving intestinal symptomatology and patients´ health status

Associação entre graus de mucosite e quantificação da interleucina 6 (IL- 6) e fator de necrose tumoral alfa (TNF-?) na saliva de pacientes submetidos a transplante de células-tronco hematopoiéticas (TCTH) / Association between degree of oral mucositis and quantification of interleukin 6 (IL-6) and tumor necrosis factor-alpha (TNF-?) in patients undergoing haematopoietic stem cell transplantation (HSCT)Paula Verona Ragusa da Silva 25 July 2016 (has links)
A mucosite oral (MO) constitui uma condição dolorosa que se desenvolve entre 47% e 100% dos pacientes submetidos a transplante de células-tronco hematopoiéticas (TCTH), impactando enormemente em sua qualidade de vida. Investigar fatores preditivos para MO por meio de exames não invasivos faz-se necessário, visando melhorar a qualidade de vida dos pacientes. O objetivo do estudo foi investigar a relação dos regimes de condicionamento e dos níveis salivares de Interleucina 6 (IL- 6) e Fator de Necrose Tumoral alfa (TNF-?) com a MO, bem como investigar o impacto destes na qualidade de vida. Foram selecionados 82 pacientes submetidos a TCTH, que foram avaliados em 4 momentos diferentes: no início do condicionamento para o TCTH (M1), no dia da infusão das células (M2), após 12/20 dias do início do condicionamento para transplante autólogo e alogênico, respectivamente (M3), e após 30 dias ou na alta hospitalar para ambos (M4). Nestes momentos, foi avaliado clinicamente o grau de MO segundo critérios da Organização Mundial da Saúde (OMS), coletada saliva total e aplicados 3 questionários de avaliação de qualidade de vida em relação à MO e à saúde bucal: PROMS, OHIP-14 e OMQoL. As informações clínicas e laboratoriais foram correlacionadas através do STATA 13.0 com 5% de nível de significância. Verificou-se que a maior incidência e intensidade de MO, os piores índices de qualidade de vida e os maiores níveis de IL- 6 e TNF-? foram registrados no M3, porém não houve correlação entre as citocinas e graus de MO. Houve associação entre altos níveis salivares de IL-6 e maiores pontuações no PROMS. O regime de condicionamento mieloablativo (ML) foi relacionado à MO intensa (graus 3 e 4) e à maiores pontuações nos 3 questionários de qualidade de vida, e os escores dos questionários foram maiores conforme maior foi intensidade da MO (p<0,05). / Oral mucositis (OM) is a painful condition that develops between 47% and 100% of patients undergoing hematopoietic stem cell transplantation (HSCT), impacting greatly on their quality of life. Investigation of predictive factors for OM through noninvasive exams is necessary, in order to improve the quality of life of patients. The aim of the study was to investigate the relationship of conditioning regimens and salivary levels of Interleukin 6 (IL-6) and Tumor Necrosis Factor alpha (TNF-?) with OM, and to investigate their impact on quality of life. We selected 82 patients undergoing HSCT, which were assessed at four different moments: at the start of conditioning for HSCT (M1), on the cell infusion day (M2), after 12/20 days of the start of conditioning for autologous and allogeneic transplantation, respectively (M3), and after 30 days or at hospital discharge for both (M4). In these moments, it was clinically evaluated the degree of OM according to criteria of the World Health Organization (WHO), collected whole saliva and applied 3 questionnaires of assessment of quality of life related to OM and oral health: PROMS, OHIP-14 and OMQoL. Clinical and laboratorial data were correlated using STATA 13.0 in a 5% significance level. It was found that the highest incidence and intensity of OM, the worst indices of quality of life and higher IL-6 and TNF-? levels were found in M3, but there was no correlation between cytokines and levels of OM. There was an association between high levels of salivary IL-6 and higher scores in PROMS. The myeloablative conditioning regimen (ML) was related to intense OM (grades 3 and 4) and to highest scores in the 3 questionnaires of quality of life, and the scores of questionnaires were higher as higher was the intensity of OM (p<0,05).

Previous issue date: 2008 / Resumo: A Chlorella vulgaris (CV) é uma alga microscópica bastante rica em nutrientes. Além de alto valor nutritivo, a Chlorella tem demonstrado possuir importantes atividades terapêuticas. Vários trabalhos do nosso laboratório e outros demonstram os efeitos protetores desta alga contra infecções virais e bacterianas, tumores, entre outros. No presente trabalho avaliamos os efeitos do tratamento profilático/terapêutico com a dose de 50mg/Kg/dia de CV sobre a resposta imunohematopoética de animais normais e portadores de tumor ascítico de Ehrlich (TAE). O estudo dos mecanismos envolvidos na proteção produzida pela alga frente às alterações induzidas pelo TAE no sistema imuno-hematopoético, demonstrou aumentos significativos na
atividade de células NK, na produção de citocinas com padrão Th1 (IL-2 e INF-g) e na liberação de TNF-a por células esplênicas, paralelamente a uma diminuição na produção de IL-10 (padrão Th2). Além disso, o tratamento com CV protegeu o hospedeiro da mielossupressão provocada pelo desenvolvimento tumoral, seja agindo diretamente na formação e manutenção do estroma medular, estimulando a atividade das células estromais, ou ainda estimulando a produção de citocinas reguladoras da hematopoese como a IL-6 e a IL-1a. Constatamos ainda que o tratamento com CV aumenta a atividade estimuladora de colônias (CSA) no soro de animais portadores de tumor e a capacidade proliferativa de células esplênicas. Outros resultados que merecem destaque são aqueles relacionados à ativação macrofágica. Nossos resultados demonstram que, em animais normais, o tratamento com CV promove aumento significativo nos níveis de TNF-a nos sobrenadantes das culturas de macrófagos peritoneais e esplênicos sem alterar a produção de IFN-g e IL-10. Além disso, o restabelecimento da produção normal de TNF-a, IFN-g e IL-10 por macrófagos peritoneais de animais portadores de TAE e reversão parcial dos efeitos produzidos pela presença do tumor na atividade dos macrófagos esplênicos, também foram observados. Outro efeito do CV sobre macrófagos peritoneais de animais portadores de TAE foi o estímulo da produção de H2O2 e NO2 -. Analisando nossos resultados podemos dizer que a CV além de possuir alto valor nutritivo, capacidade antioxidante e atividade anti-viral e antibacteriana, apresenta importante atividade imunomoduladora frente ao desenvolvimento tumoral, já que na presença do tumor o hospedeiro que vem recebendo Chlorella é capaz de reagir mais prontamente e com maior vigor na indução de mecanismos essenciais de defesa imunológica. Neste sentido, sugerimos que a alga Chlorella vulgaris pode ser um candidato a agente preventivo e complementar no tratamento do câncer. / Abstract: Chlorella vulgaris (CV) is a microscopic alga rich in nutrients. Besides the high nutritive value, this alga has shown important therapeutic properties. Many studies from our laboratory and others have shown the protective effects of CV against viral and bacterial infections, tumors, among others. In this work we evaluated the effects of the prophylactic/therapeutic treatment with a dose of 50 mg/Kg/day in normal and Ehrlich ascites tumor (EAT)-bearing mice. The study of the mechanisms involved in the protection produced by the CV against the EAT, induced alterations in the immune-hematopoietic system; demonstrating significant improvements in the NK cells activity, Th1 (IL-2 and INF-g) and TNF-a cytokines production by mononuclear spleen cells, concomitantly with the decrease of IL-10 (Th2) production. Moreover, treatment with CV protected the host of myelosuppression caused by tumor development, it is acting directly in the formation and maintenance of bone marrow stroma, by stimulating the activity of stromal cells, or stimulating the production of important hematopoiese regulatory cytokines like IL-6 and IL-1a. We are also certain that the treatment with CV increases the colony-stimulating activity (CSA) in the
serum of tumor bearing mice and the proliferative capacity of spleen cells. Other results that deserve attention are those related to macrophage activation. Our results show that in normal mice, treatment
with CV, promotes significant increase in the TNF-a levels in upernatants of the cultures of peritoneal and spleen macrophages without altering the production of IFN-g and IL-10. Moreover, restoration of the normal production of TNF-a, IFN-g and IL-10 by peritoneal macrophages from animal carriers of EAT and partial reversal of the effects produced by the presence of tumor in the splenic macrophages activity were also observed. Another effect of CV on peritoneal macrophages of tumor bearing mice was the stimulation in the production of NO2 - and H2O2. Looking at our results we can say that the alga Chorella vulgaris in addition to its high nutritional value, antioxidant capacity and antiviral and antibacterial activities, it also presents important immunomodulatory activity against tumor development, since that in the presence of the tumor, the host that has received Chlorella is able to react more promptly and with greater force in the induction of essential mechanisms of immune defense. Accordingly, we suggest that the alga Chlorella vulgaris can be a candidate for the preventive and complementary treatment of cancer by strengthening the immune system. / Doutorado / Farmacologia

Abstract
Apoptosis plays a crucial part in human ovarian function from fetal development to the end of reproductive potential. Failures in the regulation of ovarian apoptosis are associated with many pathological conditions such as premature ovarian insufficiency, infertility and cancer. The purpose of the present study was to analyze the factors regulating cell survival in human fetal and adult
ovaries.
The fetus is exposed to maternal- and placental-derived estrogens and insufficient estrogen action has destructive effects on rodent ovarian development. We detected estrogen receptors and estrogen-converting enzymes in human fetal ovaries after primordial follicle formation, indicating that estrogens participate in human fetal ovarian development, especially after folliculogenesis.
The WNT4 gene is crucial for female sexual differentiation, follicle formation and oocyte survival. We detected WNT4 in follicular cells of fetal and adult human ovaries. In addition, Wnt4- knockout mice demonstrated a dramatic loss of oocytes before birth. However, no changes were detected in protein expression patterns of common apoptosis-related proteins. The results support the possible role of WNT4 in human ovarian function and strengthen previous knowledge on the antiapoptotic role of Wnt4.
Apoptosis signaling is mediated by extracellular- and mitochondria-associated- pathways, ending in caspase cascade activation and fragmentation of cellular structures. In the present study we analyzed the expression of several apoptosis-related factors and detected TRAIL, TNF, Bcl-XL, Bok and caspase-3 in human ovaries. In addition, TRAIL was found to be a potent and rapid inducer of human granulosa tumor cell (KGN) apoptosis. Lentiviral downregulation of Bok or Bcl-XL protein expression in KGN cells also resulted in significant changes in cell vulnerability to apoptosis. The results show for the first time the spatiotemporal expression patterns of TRAIL, TNF, Bcl-XL, Bok and caspase-3 in human ovaries and suggest an important functional role of TRAIL, Bok and Bcl-XL in regulation of human ovarian apoptosis.
The present study offers novel information on the expression and function of cell survival factors in human ovaries. These new findings open possibilities for future clinical research in attempts to understand and treat ovarian diseases caused by imbalanced regulatory pathways of apoptosis.

The Effect of Macrophage-secreted Factors on Preadipocyte SurvivalMolgat, André January 2013 (has links)
Adipose tissue (AT) expansion and remodeling that maintains healthy function relies on stromal preadipocytes capable of differentiating into new adipocytes (adipogenesis). During chronic positive energy balance, a relative deficit in adipogenesis, from either a decrease in preadipocyte number or their capacity to differentiate, leads to excessive adipocyte hypertrophy and AT dysfunction. AT contains macrophages whose number and activation state is dynamically regulated with changes in AT mass. This study aims to investigate the effect of macrophage-secreted factors on preadipocyte survival.
To assess the effect of macrophage-secreted factors on preadipocytes, murine 3T3-L1 preadipocytes or human primary preadipocytes were incubated with macrophage-conditioned medium (MacCM), prepared from either murine (J774A.1, RAW264.7, bone marrow-derived) or human (THP-1, monocyte-derived) macrophage models, respectively. MacCM inhibited preadipocyte apoptosis and activated pro-survival signaling in both preadipocyte models. Inhibition of PDGFR, Akt, or ERK1/2 reduced the pro-survival effect of MacCM in 3T3-L1 preadipocytes. Inhibition of reactive oxygen species (ROS) generation, or enhancement of ROS clearance, reduced MacCM-dependent 3T3-L1 preadipocyte survival. Whereas anti-inflammatory activated macrophages retained the ability to prevent preadipocyte apoptosis, pro-inflammatory activated macrophages did not. TNF-α immunoneutralization restored the survival activity of pro-inflammatory MacCM on 3T3-L1 preadipocytes.
These studies reveal a novel pro-survival effect of MacCM on preadipocytes, and identify signaling molecules (PDGF, Akt, ERK1/2, and ROS) that underlie this action. Macrophage activation was found to regulate the pro-survival activity of MacCM. These in vitro cell culture studies are consistent with a model in which the extent of preadipocyte apoptosis in vivo may determine preadipocyte number and the ability of AT to expand while maintaining healthy function during chronic positive energy balance.

O marcador genético mais associado à psoríase em todas as populações é o HLA-CW06. Polimorfismos na região promotora do TNF-a, especialmente a troca de uma guanina por uma adenina nas posições -238 e -308 estão relacionados à alta produção de TNF-a e risco aumentado para psoríase nas populações caucasóides e não em asiáticos. Com o objetivo de determinar se polimorfismos destes genes podem ser fatores de risco para susceptibilidade ou gravidade da doença em pacientes brasileiros, foi realizado um estudo caso-controle com 69 pacientes com psoríase de início até 40 anos, com acompanhamento por dez anos para caracterização de sua evolução clínica em doença leve (grupo I) e grave (grupo II), e 70 indivíduos sadios. Foi feita a identificação dos alelos HLA classe I e II e SNPs da região promotora do TNF-a -238 e -308. Coletaram-se 10 mL de sangue periférico dos indivíduos e se realizou a extração do DNA através do método salting out. O DNA foi amplificado pela reação em cadeia da polimerase (PCR), com primers sequência-específica. As freqüências alélicas e gênicas foram estimadas por contagem direta e a comparação entre as frequências dos grupos foi efetuada por Teste de Fisher (programa GraphPad InStat 3.05). Dois métodos computacionais foram usados para determinar os haplótipos dos indivíduo: (1) o algoritmo ELB implementado pelo software Arlequin 3.1 e (2) um método de base coalescente implementado pelo software PHASE v2, e as freqüências de cada haplótipo foram comparadas por Teste de Fisher. No grupo II, observou-se maior associação com fatores desencadeantes como estresse, início na adolescência e predominância do sexo masculino. Pode-se sugerir que os alelos HLA-B*37, -Cw*06, -Cw*12 e -DRB1*07 foram associados ao curso mais grave da doença, enquanto - B*57 à doença mais leve. O aielo DRBT04 teve tendência a associação negativa. Ao se comparar o grupo I com o grupo II, o alelo HLA-B*37 pode ser interpretado como fator de mau prognóstico. Não houve diferença estatística entre polimorfismos da região promotora do TNF-a entre pacientes e controles. Este estudo apontou uma alta frequência do genótipo TNF-a -238 G/G {OFt 3,21; Cl:1,06-9,71; p=0,04), assim como do alelo -238 G, no grupo com doença mais grave e, ao contrário, o genótipo -238 G/A com frequência maior no grupo de boa evolução. O haplótipo -238A-308G mostrou frequência reduzida conferindo um efeito protetor. Estes dados não correspondem ao reportado para as populações caucasianas, considerando que a população brasileira é miscigenada. Polimorfismos dos SNPs do TNF-a não parecem ser um fator de susceptibilidade genética mais importante do que o já conhecido HLA-Cw*06 em pacientes brasileiros, mas podem ter relação com as manifestações e evolução da doença. / Abstract: Psoriasis is an erythematous, scaly inflammatory dermatosis with a complex immunologic basis. The strongest genetic marker for psoriasis is HLA-Cw*06. Polymorphisms in the TNF-a promoter region, especially replacement of guanine with adenine in positions -238 and -308 are related to higher TNF-a production and higher risk for psoriasis in Caucasoid populations, not found in Asians. We did a case-control study of 69 patients with psoriasis type I and 70 controls, characterized clinical progression along 10-years of follow-up in mild or severe disease and determined HLA class I, II and TNF-a SNPs -238 and -308 polymorphisms to demonstrate whether these polymorphisms may be genetic risk for susceptibility to psoriasis or severity of the disease in Brazilians. Peripheral blood (10 ml) was collected. Genomic DNA from both psoriasis patients and controls was isolated using a salting out procedure. Polymorphisms were identified by PCR/SSP. Alleles and genotypes frequencies were compared by Fisher's test (GraphPad InStat 3.05 software). Two computational methods were used to determine the haplotypes of each subject, without taking into account any prior information: (1) the ELB algorithm implemented by the ARLEQUIN 3.1 software and (2) a coalescence based method as implemented by the PHASE v2 software. The haplotype frequencies were compared between group pairs by Fisher's test. Severe disease was found more frequently in male patients, associated with environmental factors and onset at adolescence. It may be suggested that alleles HLA- B*37, -Cw*06, -Cw*12 and -DRB1*07 were associated with severe disease course, while -B"57 with mild disease. No statistical difference was found between the patients and controls regarding polymorphisms frequencies in TNF SNPs. This study pointed to a higher TNF-238 G/G genotype frequency (OR 3,21; Cl:1,06-8,71; p=0,04) in the group with severe disease and -238A-308G haplotype was found in reduced frequencies revealing a protective effect. These data do not correspond to those reported for the Caucasian population, considering that Brazilian population is admixed, and this is the first consideration about TNF-a SNPs in psoriasis in this population. Previous issue date: 2015 / Resumo: As plaquetas são importantes células na inflamação, entretanto, os trabalhos que estudam as citocinas na reatividade plaquetária são raros. O objetivo do presente trabalho foi estudar os efeitos do fator de necrose tumoral-alfa (TNF-?) em plaquetas. Ensaios de agregação foram realizados incubando-se plaquetas com crescentes concentrações de TNF-? (1 - 3000 pg/ml) por diferentes intervalos de tempo (5 - 60 min), na ausência ou presença do antagonista não seletivo dos receptores TNFR1 e TNFR2, o R7050. Também foi estudado o efeito do TNF-? na viabilidade plaquetária utilizando-se o MTT. O efeito do TNF-? na mobilização de Ca2+ em plaquetas foi investigado através de ensaios de fluorescência utilizando-se o fluo-3-AM; os ensaios de western blotting foram realizados para o estudo da ativação da enzima c-Src e do receptor de fibrinogênio. Finalmente, foram determinados os níveis intraplaquetários de AMPc e GMPc por ELISA. O TNF-? inibiu a agregação plaquetária induzida por ADP ou trombina de forma dependente da concentração da citocina e do tempo de incubação. O efeito inibitório máximo do TNF-? na agregação induzida por ADP (5 ?M) foi obtido com a concentração de 300 pg/ml por um tempo de incubação de 30 min (90 ± 7% de inibição), o qual foi significativamente prevenido pela pré-incubação das plaquetas com o R7050. A viabilidade plaquetária não foi modificada pela incubação por 60 min com o TNF-? (30 e 3000 pg/ml). A incubação de plaquetas com TNF-? (300 pg/ml, 30 min) reduziu em 53% o aumento da concentração de Ca2+ total causado pela adição de trombina (200 mU/ml). A queda da concentração de Ca2+ citosólica plaquetária causada pelo TNF-? foi em decorrência da diminuição em 1,8 e 3,4 vezes da mobilização interna do íon e do influxo do mesmo, respectivamente. O TNF-? reduziu em 60% os níveis de AMPc em plaquetas ativadas com ADP. Por outro lado, o TNF-? aumentou significativamente os níveis de GMPc em plaquetas ativadas por ADP (aumento de 51%). A pré-incubação de plaquetas com o inibidor da guanilil ciclase ODQ não reduziu o efeito inibitório do TNF-? na agregação induzida por ADP. Os ensaios de western blotting mostraram que o TNF-? reduziu significativamente a fosforilação do resíduo de Tyr416 da c-Src em plaquetas ativadas. Da mesma forma, o TNF-? reduziu em 37% a fosforilação do resíduo de Tyr773 da subunidade ?3 da integrina ?IIb?3 (receptor de fibrinogênio) em plaquetas ativadas por ADP. Portanto concluímos que o TNF-? inibe a agregação plaquetária via receptores TNFR1 e/ou TNFR2, sem reduzir a viabilidade das plaquetas. O efeito inibitório do TNF-? na agregação é acompanhado pela redução de Ca2+ citosólico e inibição de c-Src e do receptor de fibrinogênio em plaquetas, sendo estes independentes de AMPc ou GMPc / Abstract: Platelets have been described as important cells in inflammation; however, the effects of cytokines on platelet reactivity are rarely studied. The objective of the present work was to investigate the effects of the tumor necrosis factor-alpha (TNF-?) in platelets. Aggregation assays were carried out incubating platelets with increasing TNF-? concentrations (1 - 3000 pg/ml) for different intervals of times (5 - 60 min), in the absence or in presence of the non-selective antagonist of TNFR1 and TNFR2, R7050. Effect of TNF-? on platelet viability was determined using MTT. The effect of TNF-? on the Ca2+ mobilization in platelets was investigated through fluorescence assays using fluo-3AM and Western blotting assays were carried out to determine the activation of c-Src and the fibrinogen receptor. Finally, the cAMP and cGMP levels in platelets were determined by ELISA. TNF-? dose- and time-dependently inhibited ADP or thrombin-induced platelet aggregation. The inhibitory effect of TNF-? on ADP(5 ?M)-induced platelet aggregation was maximum in a concentration of 300 pg/ml incubated with platelets for 30 min (90 ± 7% of inhibition), which was significantly prevented by the incubation of platelets with R7050. Platelet viability was not modified by TNF-? (30 and 3000 pg/ml) incubated for 5 to 60 min. Incubation of platelets with TNF-? (300 pg/ml, 30 min) reduced the increased total Ca2+ concentration induced by thrombin (200 mU/ml) by 53%. Decreasing Ca2+ internal mobilization (1,8 fold) and decreasing in external Ca2+ influx (3,4 fold) led to a reduction of total cytosolic Ca2+ in TNF-? activated platelets. TNF-? reduced the cAMP levels in ADP-activated platelets by 60%. On the other hand, TNF-? significantly increased cGMP levels in ADP-activated platelets (51% increase). Pre-incubation of platelets with the guanylyl cyclase inhibitor ODQ did not modify the inhibitory effect of TNF-? on ADP-induced platelet aggregation. Western blotting analysis showed that TNF-? significantly reduced phosphorylation on Tyr416 of c-Src in activated platelets. Similarly, TNF-? reduced by 37% the Tyr773 phosphorylation of ?3 subunit of ?IIb?3 integrin (fibrinogen receptor) in ADP-activated platelets. Therefore, our results show that TNF-? inhibits platelet aggregation via TNFR1 and/or TNFR2 receptors, without affecting platelet viability. Inducering av interferon-gamma och tumörnekrosfaktor-alfa i helsaliv : En icke-invasiv metod för att diagnostisera celiakiKokrehel, Dorina January 2022 (has links)
Celiaki (glutenintolerans) är en kronisk, autoimmun sjukdom med diffusa symtom. Vid förtäring av glutenhaltig mat uppstår en allergisk reaktion hos glutenintoleranta individer. Gluten kan inte fullständigt brytas ned av kroppens enzymer, vilket betyder att icke nedbrutna peptidfragment (såsom glutamin) absorberas i tarmslemhinnan. Enzymet transglutaminas katalyserar omvandlingen av glutamin till glutamat. Glutenkänsliga T-celler aktiveras av glutamat att utsöndra proinflammatoriska cytokiner såsom interferon-gamma (IFN-γ) och tumörnekrosfaktor-alfa (TNF-⍺). Syftet med studien var att undersöka om gluten- och gliadinstimulering av celler i helsaliv in vitro kan inducera produktion av IFN-γ och TNF-⍺. Pilotstudier med 2 försökspersoner utfördes, där celler i salivprover stimulerades med enzymatiskt nedbrutet gliadin (<40 mg gliadin), samt PHA (5 µg/mL), PMA (50 ng/mL) och LPS (1 µg/mL) 20 timmar vid 37 ˚C. Cytokinproduktionen i salivproverna kvantifierades med ELISA och uppreglering av IFN-γ och TNF-⍺ undersöktes med RT-qPCR. Efter metodutveckling upprepades stimulering och ELISA med salivprov från 12 försökspersoner (6 individer med och utan celiaki). Immunreaktionen som uppstår hos glutenintoleranta individer in vivo kunde inte återskapas i saliv in vitro med den framtagna metoden. Hos övervägande delen av salivproverna var cytokinproduktionen under detektionsgränsen, 4 pg/mL för IFN-γ och 15,6 pg/mL för TNF-⍺. Det finns risk för att outforskade detaljer eller agens saknades från reaktionskedjan och därmed kunde den förväntade immunreaktionen inte återskapas. En annan felkälla kan vara för låg koncentration av immunceller i saliven. / Celiac disease is a chronic, autoimmune disease that has diffuse symptoms. Upon consuming gluten containing food, an allergic reaction occurs in gluten-sensitive individuals. Gluten cannot be fully digested by human enzymes, which leads to non-digested peptide fragments (such as glutamine) to be absorbed in the gastrointestinal wall. The transglutaminase enzyme catalyzes the conversion of glutamine to glutamate. Glutamate activates gluten-specific T-lymphocytes to produce proinflammatory cytokines e.g., interferon-gamma (IFN-γ) and tumor necrosis factor alpha (TNF-⍺). The aim of this study was to investigate whether stimulation of cells in whole saliva in vitro with gluten and gliadin can induce production of IFN-γ and TNF-⍺. Pilot studies were conducted, where cells in saliva from 2 subjects was stimulated with enzymatically digested gliadin (<40 mg gliadin) together with PHA (5 µg/mL), PMA (50 ng/mL) and LPS (1 µg/mL) for 20 hours at 37 ˚C. The production of cytokines was quantified by ELISA, and the upregulation of IFN-γ and TNF-⍺ was analyzed by RT-qPCR. After method development, the stimulations and ELISA quantifications of the proinflammatory cytokines were repeated in saliva samples from 12 subjects (6 individuals with and without celiac disease). The immune reaction that occurs in people with celiac disease could not be recreated in saliva in vitro with the developed method. In most of the samples the production of cytokines was under the detection range, 4 pg/mL for IFN-γ and 15,6 pg/mL for TNF-⍺. There is risk of unstudied details or agents missing from the reaction chain, and therefore the expected immune reaction could not be recreated. Another source of error could be low concentration of immune cells in saliva.

Elucidating the Molecular Mechanism of CYLD-Mediated Necrosis: A DissertationMoquin, David M. 13 May 2013 (has links)
TNFα-induced programmed necrosis is a caspase-independent cell death program that is contingent upon the formation of a multiprotein complex termed the necrosome. The association of two of the components of the necrosome, receptor interacting protein 1 (RIP1) and RIP3, is a critical and signature molecular event during necrosis. Within this complex, both RIP1 and RIP3 are phosphorylated which are consequential for transmission of the pro-necrotic signal. Namely, it has been demonstrated that RIP3 phosphorylation is required for binding to downstream substrates. Nevertheless, the regulatory mechanisms governing necrosome activation remain unclear. Since necrosis is implicated in a variety of different diseases, understanding the biochemical signaling pathway can potentially yield future drug targets. I was interested in identifying other regulators of necrosis in hope of gaining a better understanding of the necrosis signaling pathway and regulators of the necrosome. To address this, I screened a cancer gene siRNA library in a cell line sensitive to necrosis. From this, I independently identified CYLD as a positive regulator of necrosis. Previous studies suggest that deubiquitination of RIP1 in the TNF receptor (TNFR)-1 signaling complex is a prerequisite for transition of RIP1 into the cytosol and assembly of the RIP1-RIP3 necrosome. The deubiquitinase cylindromatosis (CYLD) is presumed to promote programmed necrosis by facilitating RIP1 deubiquitination in this membrane receptor complex. Surprisingly, I found that TNFα could induce RIP1-dependent necrosis in CYLD-/- cells. I show that CYLD does not regulate RIP1 ubiquitination at the receptor complex. Strikingly, assembly of the RIP1-RIP3 necrosome was delayed, but not abolished in the absence of CYLD. In addition to the TNFR-1 complex, I found that RIP1 within the necrosome was also ubiquitinated. In the absence of CYLD, RIP1 ubiquitination in the NP-40 insoluble necrosome was greatly increased. Increased RIP1 ubiquitination correlated with impaired RIP1 and RIP3 phosphorylation, a signature of kinase activation. My results show that CYLD regulates RIP1 ubiquitination in the NP-40 insoluble necrosome, but not in the TNFR-1 signaling complex. Contrary to the current model, CYLD is not essential for necrosome assembly. Rather, it facilitates RIP1 and RIP3 activation within the necrosome and the corollary is enhancement of necrosome functionality and subsequent necrosis. My results therefore indicate that CYLD exerts its pro-necrotic function in the NP-40 insoluble necrosome, and illuminates the mechanism of necrosome activation.
