Molecular genetic analyses focused on selected aspects of technological quality were carried out on 25 wheat genotypes with non-standard coloured caryopses. Occurrence of null alleles was observed for the GBSSI gene responsible for amylose synthesis, the so-called waxy gene. Null allele Wx-B1 was found in 20 % of the genotypes, other null alleles were not observed. Allelic state of Pina and Pinb genes affecting caryopsis hardness was analysed too. Allele identification of three genotypes was not successful. In the Pina case, only the wild-type allele Pina-D1a was present, in the Pinb wider variability was observed, 36 % of the genotypes carried wild-type Pinb-D1a allele, 40 % carried Pinb-D1b allele and 12 % Pinb-D1d allele. Instrumental hardness was measured and it correlated with the detected allelic state. Caryopses texture was determined based on the laser beam transmission through the caryopsis. Occurrence of mealy caryopses ranged from 0 % to 100 %. Caryopsis texture and hard-ness do not correlate neither on the instrumental nor on the genetic basis. Null allele Wx-B1 occurrence did not influence the caryopsis hardness nor their texture. Partial sequences of F3H a DFR structural genes of the anthocyanin biosynthetic pathway were obtained at five genotypes with blue, purple and white caryopses. Single nucleotide polymorphisms among individual gene copies were observed.

Focus of the diploma thesis is to investigate the influence of the selected factors (type of wood, thickness, densification) on the wood hardness. For the experimental measurements the Brinell hardness test method was used. The hardness measurements were performed on the specimens made from two different types of wood. Obtained results were statistically analyzed into the graphs. The main goal of the thesis was to determine the change in the static hardness in dependence on the previously mentioned factors. From the experimental data it can be seen that the wood hardness increases with the higher degree of densification . This phehomenon applies to both investigated wood materials which are European beech (Fagus sylvatica) - hardwood specimen and European aspen (Populus tremula) - softwood specimen.

With the rising amount of AD technology utilizers, the issue how to storage, such liquid material, which digestate is. In recent time, there are some other ways to utilize it, except fertilizing with liquor, liquid part of digestate. Digestate can be dried and pressed into briquette or pelleted form. In briquette form is important to understand relations between properties of individual components because one property can influence another one, which constituting texture of the material and its reprocessing into the final form of the product. Thus this study was conducted, to find and/or quantify the relation between the texture of biomass briquettes and their durability and strength of shape during manipulation, storage and use. Basic hypothesis of this work is based on open presumption that briquette texture is dependent on many parameters. Some of those parameters were tested. Namely, composition of matter, physical properties of digestate briquettes, durability and hardness, size analysis and other. Digestate has approved as good material for compression with high content of nutrients. The rate of abrasion varied 7-12% at the beginning decreasing to 3-5%. Shore testing showed results predicating relation between particle size and hardness. Sieve and image analysis then showed range of particles ranging size 0.01 mm from 99.5% and 10 mm from 99.7%, most of them with needle-like shapes. This study suggested the method that allows analysing particle size distribution in sample and describes other briquette properties, which can be useful for next research and commercial purposes.

This diploma thesis deals with methods of heat treatment of tool steel. The thesis briefly describes iron alloys and their properties, possible heat treatment is described. In the experimental part, there is hardening of K720 steel, (ČSN 41 9312) into various hardening environments is performed and mechanical properties such as hardness, impact work and steel strength are detected. The results evaluate the best heat treatment for blade production, both functional and ecological. According to the best method of heat treatment, the production of the product will be realized.

Vliv chemicko-tepelného zpracování na povrchové vlastnosti ocelí

This thesis deals with heat treatment and chemical heat treatment of steel. The first part is devoted to the theory of heat treatment and chemical heat treatment and detailed description of the most used methods. Next, the work describes the tests used in the practical measurement. In the experimental part, there is hardness comparison of unalloyed and low-alloyed steels after chemical heat treatment in six ways. The resulting structures are evaluated metallographically. In the last part, the work evaluates the influence of chemical heat treatment on corrosion resistance after salt spray test. Conclusion is devoted to the discussion about application of chemical heat treatment in technical practice.
