Estudo do envolvimento da via NMDA-NO do eixo dorso-ventral do hipocampo sobre o comportamento induzido pelo estresse de nado forçado / Involvement of the NMDA-NO pathway of the dorso/ventral hippocampal axis in the modulation of behavioral responses elicited by the forced swimming test

Acredita-se que diferenças hodológicas e diferente padrão de expressão gênica ao longo do eixo dorso/ventral do hipocampo seriam responsáveis pela distinta função entre a porção dorsal (HD) e ventral do mesmo (HV). HD seria responsável por processos cognitivos, tais como memória e aprendizagem espacial, e o HV pelas respostas neuroendócrinas e emocional-motivacionais ao estresse. No entanto, não há muitos estudos acerca de diferenças entre HD e HV na modulação de comportamentos relacionados à neurobiologia da depressão. Há, contudo, dados indicando que o bloqueio de receptores glutamatérgicos do tipo NMDA ou da síntese de NO no HD induz efeito semelhante ao dos antidepressivos (i.e. do tipo antidepressivo) no teste do nado forçado (TNF). Quanto ao HV, a função da neurotransmissão glutamatérgica/nitrérgica na neurobiologia da depressão permanece não investigado. Dessa forma, o objetivo do presente estudo foi realizar o bloqueio reversível do HD ou do HV, em diferentes momentos, em animais submetidos ao TNF. O próximo passo foi realizar a micro-injeção intra-HD ou intraHV do antagonista NMDA AP-7 ou do inibidor da óxido nítrico sintase neuronal (nNOS), N-PLA, ou do inibidor da guanilato ciclase solúvel (sGC), ODQ. Os resultados mostram que o bloqueio do HD ou do HV com cloreto de cobalto (CoCl2, bloqueador da neurotransmissão sináptica dependente de cálcio) não modificou o comportamento dos animais no TNF. No entanto a administração de AP-7 ou N-PLA ou ODQ no HV antes do teste, reduziu o tempo de imobilidade no TNF. Por outro lado, a injeção dessas drogas no HD foi capaz de reduzir o tempo de imobilidade quando administradas tanto depois do pré-teste quanto antes do teste. Estes resultados sugerem que as vias NMDA-NO do HD e do HV, estariam envolvidas na modulação da resposta comportamental frente ao estresse do nado forçado. Além disso, os dados indicam que a participação dessas estruturas é importante em diferentes momentos após a exposição ao estresse. / It is believed that hodological and genetic pattern of expression differences along dorsal/ventral hippocampal axis would be responsible for distinct functions attributed to its dorsal (DH) and ventral (VH) poles. DH would be responsible for cognitive process, such as spatial memory and learning, whereas the VH would be responsible for neuroendocrine and emotional-motivation responses to stress. However, there is no many studies about possible differences between DH and VH in the modulation of behavioral responses related to the neurobiology of depression. Though, there are data showing that the blockade of glutamatergic NMDA receptors blockers or NO synthesis inhibition within the DH induces similar effect to that of antidepressant drugs (like antidepressant effect) in the forced swimming test (FST). On the VH, The role of the glutamatergic/nitrergic neurotransmission remains to be investigated. Thus, the aim of this work was to perform the reversible blockade of DH or VH, at different times, in animals subjected to FST. Additionally, the next step was to perform a microinjections into the DH or the VH of NMDA antagonist (AP-7), inhibitor of neuronal nitric oxide synthase (nNOS, N-PLA), or the inhibitor of soluble guanylate cyclase (sGC, ODQ). The results show that blocking the VH or the DH (cobalt chloride, calcium dependent neurotransmission inhibitor) did not modify the behavior of animals during the TNF. However administration of AP-7, N-PLA or ODQ intra-HV, before testing, caused antidepressant-like effects. Moreover, injection of such drugs, intra-HD, was able to induce similar results when administered both after the pre-test and before testing. These results suggest that NMDA-NO pathway of both VH and DH is involved in the modulation of emotional responses to the forced swim stress, although there may be an interest differential participation of these structures at different times after exposure to stress.

Συγκριτική μελέτη των φαρμακολογικών ιδιοτήτων του συμπλόκου του υποδοχέα GABAA/Βενζοδιαζεπινών μεταξύ του διαφραγματικού και κροταφικού ιπποκάμπου επίμυος

Η ΒΥΠ διαθέτει αντίτυπο της διατριβής σε έντυπη μορφή στο βιβλιοστάσιο διδακτορικών διατριβών που βρίσκεται στο ισόγειο του κτιρίου της. / Ο ιππόκαμπος στον αρουραίο είναι μια δομή σε C-σχήμα που εκτείνεται από τον διαφραγματικό πυρήνα προσθιοραχιαία έως τον κροταφικό λοβό οπισθιοκοιλιακά. Παρόλο που από παλιά θεωρείτο ως ομοιογενής δομή, έχουν παρατηρηθεί πολλές διαφορές σε όλα τα επίπεδα οργάνωσης μεταξύ του διαφραγματικού και του κροταφικού ιππόκαμπου. Οι βενζοδιαζεπίνες δρουν στις θέσεις δέσμευσης των βενζοδιαζεπινών, οι οποίες είναι αλλοστερικά συνδεδεμένες με το σύμπλοκο του GABAA, όπου ενισχύουν την δράση του GABA στους GABAA υποδοχείς, αυξάνοντας τη συχνότητα ανοίγματος του διαύλου των ιόντων χλωρίου (Cl-) και έχοντας μικρή μόνο επίδραση στο χρόνο ανοίγματος ή στην αγωγιμότητα του διαύλου. Προηγούμενη μελέτη υποδεικνύει ότι η έκφραση των α1-, β2- και γ2-υπομονάδων ήταν μικρότερη, ενώ αντίθετα η έκφραση των α2-, α5- και β1-υπομονάδων ήταν μεγαλύτερη στον κροταφικό ιπποκάμπο σε σύγκριση με τον διαφραγματικό ιππόκαμπο. Σύμφωνα με προηγούμενες μελέτες που αφορούν την συνέκφραση των υπομονάδων στο σύμπλοκο του GABAA υποδοχέα τα αποτελέσματα μας υποδηλώνουν ότι ο α1β2-υποτύπος του GABAA υποδοχέα επικρατεί στον διαφραγματικό ιππόκαμπο (ΔΙ), ενώ ο α2β1-υπότυπος κυριαρχεί στον κροταφικό ιππόκαμπο (ΚΙ). Επιπλέον, φαρμακολογικές μελέτες έχουν δείξει ότι η καταπραϋντική δράση της diazepam πραγματοποιείται μέσω των GABAA υποδοχέων, που περιέχουν την α1-υπομονάδα, ενώ η αγχολυτική δράση της πραγματοποιείται μέσω των υποδοχέων που φέρουν την α2-υπομονάδα. Επομένως, η μελέτη των φαρμακολογικών ιδιοτήτων των υποτύπων του GABAΑ υποδοχέα δίνει την δυνατότητα στο να σχεδιαστούν ειδικά φάρμακα τα οποία θα βελτιώνουν το κλινικό προφίλ ασθενειών, όπως το άγχος, η αϋπνία ή η επιληψία, δρώντας σε εξειδικευμένους υποτύπους του GABAΑ υποδοχέα. Ο στόχος της συγκεκριμένης εργασίας ήταν να μελετηθούν οι φαρμακολογικές ιδιότητες των υποτύπων του συμπλόκου του GABAA υποδοχέα/βενζοδιαζεπινών στον διαφραγματικό σε σύγκριση με τον κροταφικό ιππόκαμπο επίμυος. Έτσι, για τη μελέτη των θέσεων δέσμευσης των βενζοδιαζεπινών μεταξύ του διαφραγματικού και κροταφικού ιπποκάμπου χρησιμοποιήθηκε ο ευρέως φάσματος αγωνιστής των θέσεων δέσμευσης των βενζοδιαζεπινών, [3H]-flunitrazepam, σε μια αυτοραδιογραφική μελέτη. Επιπλέον, χρησιμοποιήθηκαν εξειδικευμένα φάρμακα, που δεσμεύονται σε συγκεριμένους υποτύπους του GABAA υποδοχέα, όπως το zolpidem (ειδικός αγωνιστής των GABAA υποδοχέων, που περιέχουν την α1-υπομονάδα), το etomidate (ειδικός θετικός αλλοστερικός ρυθμιστής των GABAA υποδοχέων, που περιέχουν τη β2-υπομονάδα) και το L-655,708 (ειδικός αντίστροφος αγωνιστής των GABAA υποδοχέων, που περιεχούν την α5-υπομονάδα) σε μια αυτοραδιγραφική κινητική μελέτη. Τα αποτελέσματα μας έδειξαν ότι οι τομές που προέρχονταν από τον ΚΙ συγκρινόμενες με αυτές από τον ΔΙ είχαν: Α) μειωμένη δέσμευση της [3H]-flunitrazepam στις CA1, CA3 και DG περιοχές, Β) μικρότερη χημική συγγένεια της δέσμευση της [3H]-flunitrazepam στη CA1 περιοχή, Γ) καμία διαφορά στον αριθμό των θέσεων δέσμευσης, Δ) μεγαλύτερες τιμές IC50 και EC50 για το zolpidem και το etomidate, αντίστοιχα και Ε) μικρότερες τιμές IC50 για το L-655,708. Συμπερασματικά, τα αποτελέσματά μας υποδεικνύουν διαφορετικές φαραμκολογικές ιδιότητες των υποτύπων του GABAΑ/ΒΖ υποδοχέα μεταξύ του ΔΙ και ΚΙ, ενώ επιβεβαιώνεται και ενισχύεται προηγούμενη μελέτη που υποδηλώνει ότι ο α1β2-υποτύπος του GABAA υποδοχέα επικρατεί στον διαφραγματικό ιππόκαμπο (ΔΙ), ενώ ο α2β1-υπότυπος κυριαρχεί στον κροταφικό ιππόκαμπο (ΚΙ). Η μελέτη αυτή είναι πιθανό να τυγχάνει ιδιαίτερης κλινικής αξίας, στην κατεύθυνση της ακριβέστερης ρύθμισης των αποτελεσματικών δόσεων των διάφορων μορίων που δρουν στις θέσεις δέσμευσης των βενζοδιαζεπινών των α1- και α2-υποτύπων του GABAA υποδοχέα. / The hippocampus in the rat appears grossly as an elongated structure with its long axis bending in a C-shaped manner from the septal nuclei rostrodorsally to the incipient temporal lobe caudoventrally. The long axis of the hippocampal formation is referred as the dorsoventral axis. Although hippocampus has been traditionally thought as a homogeneous structure, several studies have been demonstrated differences at several organization levels (from the behavioural to the cellular) between its dorsal (DH) and ventral (VH) pole. Pharmacological studies have shown that the α1-GABAA receptor subtype is associated with the sedative and anticonvulsant effects of benzodiazepines (BZs), whereas the α2-subtype is associated with the anxiolytic effects of BZs. Recent data have demonstrated a differential distribution of the GABAA receptor subunits between DH and VH, with the α1/β2 GABAA receptor subtype dominating in the DH and the α2/β1 subtype prevailing in the VH. We therefore study possible differences in the pharmacological properties and receptor binding parameters of the GABAA/BZ receptor subtypes between DH and VH, by examining: 1a) the specific binding of [3H]-flunitrazepam (BZ sites agonist), by using quantitative autoradiography, b) the kinetic parameters of [3H]-flunitrazepam specific binding, by using the “wipe off” technique and 2) the competitive displacement of [3H]-flunitrazepam binding by using zolpidem (a specific agonist of α1-subtype) and L-655,708 (a specific inverse agonist of α5-subtype) and the enhancement of [3H]-flunitrazepam binding by using etomidate (a selective positive modulator of β2- subunit), in an autoradiographical saturation kinetic study. Our results showed in VH compared to the DH: A) lower level of [3H]-flunitrazepam binding in CA1, CA3 and DG regions, B) higher KD value for [3H]-flunitrazepam specific binding in CA1 region and no differences in the Bmax value, C) higher IC50 and EC50 values for zolpidem and etomidate, respectively and D) lower IC50 values for L-655,708. In conclusion, the lower affinity of GABAA receptors for [3H]-flunitrazepam binding, the higher IC50 and EC50 values for zolpidem and etomidate, respectively, as well as the lower IC50 values for L-655,708 observed in VH compared to DH, support the evidence that the α1/β2-GABAA receptor subtype dominates in DH and the α2/β1-subtype prevails in VH and suggest differential pharmacological effects of the benzodiazepines in DH compared to VH, which could be of clinical relevence for the more accuate adjustment of the effective dose of α1- and α2-subtype specific BZ binding sites’ ligands.

O Labirinto em T elevado (LTE) é um teste que avalia separadamente dois subtipos de ansiedade: a ansiedade generalizada e o pânico, respectivamente através da avaliação dos comportamentos defensivos de esquivas inibitórias e das fugas. Assim, este estudo teve por objetivo investigar o envolvimento de receptores 5-HT1A no HV sobre a manifestação dos transtornos de ansiedade generalizada (TAG) e pânico (TP) no LTE, tendo como parâmetro a ativação de receptores gabaérgicos (GABAA) nessa área do hipocampo. Foram conduzidos dois experimentos em ratos Wistar: o Experimento 1, visando avaliar o efeito do midazolam (10, 20 ou 40nmol) - um benzodiazepínico que potencializa a ação do GABA em receptores GABAA; e o Experimento 2, para avaliar o efeito de 8-OH-DPAT (0,6, 3 ou 15nmol) – um agonista de receptores 5-HT1A. Animais dos grupos controles nos dois experimentos foram tratados com salina. Dez minutos após as microinjeções foram submetidos ao LTE para avaliação das latências (em segundos) para esquivas e fugas. Imediatamente após, os animais foram colocados no centro da arena para avaliação da atividade motora (número de quadrados percorridos). Os resultados apontaram que a maior dose de midazolam microinjetada no hipocampo ventral causou ansiólise, comprovada pela diminuição na latência das esquivas em relação aos animais controles e àqueles tratados com 10nmol do mesmo fármaco. Resultado semelhante foi constatado nas três doses de 8-OH-DPAT. Não se observou qualquer alteração nas fugas, e nem na atividade motora dos animais tratados com qualquer um dos fármacos testados. Essas evidências comportamentais indicam que a ativação de receptores 5-HT1A no HV diminuiu o comportamento de esquiva dos animais, sem afetar as respostas de fugas, semelhantemente ao que se observou em decorrência da ação do midazolam sobre receptores GABAA. Esses resultados indicam um envolvimento desses receptores na regulação do TAG, mas não do TP. / The little is known about the serotoninergic activity on 5-HT1A receptors in the ventral hippocampus in the regulation of different types of anxiety. The Elevated T Maze (ETM) is a test that evaluates separately two subtypes of anxiety: generalized anxiety and panic, respectively by evaluating the defensive behaviors of inhibitory avoidance and escapes. Therefore this study aimed to investigate the involvement of 5-HT1A receptors in VH on the manifestation of generalized anxiety disorder (GAD) and Panic (PD) in ETM, having as parameter the activation of gabaergic receptors (GABAA) in that area of the hippocampus. Two experiments were conducted in rats: Experiment 1, to evaluate the effect of midazolam (10, 20 or 40nmol) – a benzodiazepine that potentiates the action of the GABA on GABAA receptors; and Experiment 2 to evaluate the effect of 8-OH-DPAT (0.6, 3 or 15nmol) - an agonist of 5-HT1A receptors. Animal control groups in both experiments were treated with saline. Ten minutes after microinjection, the animals were submitted for evaluation to the ETM latencies (in seconds) for avoidances and escapes. Immediately after, the animals were placed in the center of the arena to eavaluation of motor activity (number of covered squares). The results showed that the highest dose of midazolam microinjected in the ventral hippocampus caused anxiolysis, evidenced by the decrease in latency of avoidances compared to controls and those treated animals 10nmol of the same drug. A similar result was observed at all three doses of 8-OH-DPAT. There was no change in the escapes, nor the motor activity of animals treated with any of the drugs tested. These behavioral evidence indicates that activation of 5-HT1A receptors in VH decreases the behavior of avoidance of the animals without affecting the escape response, similar to what was observed due to the action of midazolam on the GABAA receptors. These results indicate an involvement of these receptors in the regulation of GAD, but not of PD.

Implication de l'hippocampe ventral et des noyaux reuniens et rhomboïde du thalamus dans les processus cognitifs sous-tendant la mémoire spatiale chez le Rat / lnvolvement of the ventral hippocampus and reuniens and rhomboid thalamic nuclei in cognitive processes underlying spatial memory in rats

Ce travail de thèse avait pour objectif d’étudier le rôle de l’hippocampe (HPC) ventral et des noyaux reuniens (Re) et rhomboïde (Rh) du thalamus dans les processus cognitifs qui sous-tendent la mémoire spatiale chez le Rat. Par l’utilisation d’approches complémentaires combinant l’imagerie cérébrale, la lésion excitotoxique, l’inactivation fonctionnelle réversible et des évaluations comportementales, nos résultats ont mis en évidence : (1) l’implication spécifique de l’HPC ventral uniquement dans le rappel d’informations spatiales ; (2) un rôle-clé des noyaux Re et Rh dans la persistance d’un souvenir spatial ; (3) l’implication des noyaux Re et Rh dans le labyrinthe du double-H, un nouveau test nécessitant d’une part, l’utilisation d’informations spatiales dépendant de l’intégrité de l’HPC dorsal, et d’autre part, une flexibilité comportementale, impliquant le cortex préfrontal médian. Ainsi, l’ensemble de ces résultats permet de proposer l’existence d’un circuit HPC-préfronto-thalamique impliqué dans divers aspects du traitement des informations spatiales. / The main objective of this thesis was to investigate the role of the ventral hippocampus (HPC) and the reuniens (Re) and rhomboid (Rh) thalamic nuclei in the cognitive processes underlying spatial memory in the Rat. If our results first confirmed, in the Morris water maze, the role of the dorsal HPC in the acquisition and retrieval of a spatial reference memory, we demonstrated the specific involvement of the ventral HPC only in the recall of spatial information. In addition, by using complementary approaches combining brain imaging, excitotoxic lesion and reversible functional inactivation, we were able to show for the first time a key role for the Re and Rh in the persistence of a spatial memory (25 days). Finally, the third set of experiments has highlighted the involvement of the Re and Rh in a mnemonic task performed in a new test, the double-H maze, which requires the use of spatial information depending on the integrity of the dorsal HPC, and a behavioral flexibility, involving the medial prefrontal cortex. Thus, taken together, these results suggest the involvement of a HPC-prefronto-thalamic network in various aspects of spatial information processing.

Contribuição diferencial do hipocampo ventral e do complexo amidalóide na modulação de respostas defensivas inatas e condicionadas de camundongos /

Nas últimas décadas, observa-se um crescente interesse pela utilização de estímulos naturalísticos para o estudo das bases neurais de emoções como o medo e ansiedade. Nesse contexto, o teste de exposição ao rato (RET), um novo modelo etológico de interação presa-predador, utilizando camundongos (presa) e ratos (predador), foi desenvolvido para avaliar a expressão de diferentes comportamentos defensivos na presa. Entretanto, poucos estudos foram conduzidos com esse modelo no intuito de investigar as bases neurais das respostas defensivas de camundongos expostos ao rato. Adicionalmente, evidências da literatura destacam que o hipocampo ventral (HV) e o complexo amidalóide (CA) parecem contribuir diferencialmente na modulação de respostas defensivas frente a estímulos proximais (predador) ou potenciais. Assim, o presente estudo foi conduzido para investigar o papel do HV e do CA nas respostas defensivas de camundongos exibidas diante do predador (rato) e do contexto associado ao predador. Para tal, o presente estudo foi dividido em quatro etapas. Na primeira delas, investigamos se o estresse da exposição ao predador no RET altera a secreção de corticosterona em camundongos e determinamos a magnitude e a duração desta secreção. Na segunda etapa, avaliamos o papel do HV e do CA, através da injeção local do agonista de receptores GABAA muscimol (0,1 μg/0,1 μl), na mediação de respostas comportamentais defensivas de camundongos expostos ao RET (situação proximal) e comparamos com aquelas apresentadas durante a exposição ao labirinto em cruz elevado (LCE - situação potencial). (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: When animals are confronted with environmental threatening situations such as exposure to a predator as well as to height, high illumination and nociceptive stimuli they exhibit defensive behaviors. Over the past decades there has been a growing interest by the neuroscientists in the use of naturalistic stimuli to the study of the neural systems of the emotions such as fear and anxiety. In this context, the Rat Exposure Test (RET) which is a new ethological model of prey-predator interaction using mice (prey) and rats (predator) was developed in order to evaluate the expression of different defensive behaviors in the prey. However few studies using this model have been carried out with the objective of investigating the neural systems of the defensive behaviors in mice exposed to rats. In addition, evidence in literature has shown that the ventral hippocampus (VH) and the amygdaloid complex (AC) contribute differentially in the modulation of defensive behaviors during exposure to either potential or immediate stimuli (predator). Thus, the present study was aimed at investigating the role of VH and AC in the modulation of defensive behaviors of mice when exposed to predators (rats) as well as the predatory context. The experiment comprised four parts: (i) to investigate both whether stress regarding the exposure to the predator alters the corticosterone secretion in mice and to determine the magnitude and the duration of this secretion; (ii) the role of VH and AC was evaluated through local microinjection of the GABAA receptor agonist muscimol (0,1 μg/0,1 μl) in the modulation of defensive behavioral responses of mice exposed to RET (proximal aversive situation). In this sense, the elevated plus maze (EPM) and the rat exposure test are effective to evaluate these states and EPM exposure (aversive stimulus) can result in activation of serotonergic pathways with projections to structures belonging to the defense system, such as , amygdala, septum, hypothalamus, periaqueductal gray (PAG) and hippocampus. The hippocampus has a large amount of serotonin receptors (5- HT) and gamma-aminobutyric acid (GABA). In the present study, we used male mice of Swiss Albino, received surgical implantation of guide cannula and subsequent administration of drugs in the ventral hippocampus. After recovery, the animals were tested in EPM (Experiment 1 and 2) or the test exposure to the rat (Experiment 3 and 4). In Experiment 1, administration of midazolam (3.0 and 30.0 nmol) produced anxiolytic effect characterized by increased percentages of entries and time spent in the open arms of the EPM reduction measures and ethological (risk assessment) as percentages of dives stretched and secured. In Experiment 2, mice received combined injections Saline + Saline, Saline + MDZ, ondansetron (OND) + Saline and MDZ (30.0 nmol) + OND (0.03 nmol, antagonist of 5-HT3). Combined treatment of Sal + MDZ produced anxiolytic effect and this effect was reversed by the combined administration of OND + MDZ. The porcentanges of entries and time spent in open arms were lower (P> 0.05) than those found in group Sal + MDZ. Experiment 3 showed the effects of exposure of mice in the presence of mouse toy (RB = neutral stimulus) or mouse real (VR = aversive stimulus, Long Evans rats), under the administration of midazolam (3.0 14 and 30.0nmol). The animals were exposed to RV shortening the holding box (model effect) compared to animals exposed to RB. Animals treated with MDZ in two doses, there was an increase in transitions between the sides of the apparatus, increased time in the area of operation and increases the latency of escape to the protected area and contact time with the grid. In Experiment 4, we evaluated the effect of combined injection of midazolam and ondansetron protocol (Experiment 2). The MDZ 30.0 nmol produced anxiolytic effects and the blockade of this effect when the mice were combined administration of ondansetron and midazolam in the ventral hippocampus. Data from this study suggest that, first, control over emotional reactions and defense of the ventral hippocampus of mice exposed to EPM test or exposure to the rat are mediated via GABABenzodiazepines. Furthermore, there is a likely cross-modulation between GABAergic interneurons and 5-HT3, for blocking 5-HT3 via ondansetron can decrease the GABA release. / Modelos animais têm sido frequentemente utilizados para a investigação da neurobiologia dos estados emocionais (ansiedade e medo). Neste sentido, o labirinto em cruz elevado (LCE) e teste de exposição ao rato são eficazes para avaliar esses estados e a exposição ao LCE (situação aversiva) pode resultar em ativação das vias serotonérgicas com projeções para estruturas pertencentes ao sistema de defesa, tais como, amídala, septo, hipotálamo, substância cinzenta periaquedutal (SCP) e hipocampo. O hipocampo apresenta grande quantidade de receptores de serotonina (5-HT) e do ácido gama-aminobutírico (GABA). No presente estudo, foram utilizados camundongos machos, da cepa Suíco Albino, que receberam implantação cirúrgica de cânulas guia e posterior administração de drogas no hipocampo ventral. Após recuperação, os animais foram avaliados nos testes LCE (Experimento 1 e 2) ou exposição ao rato (Experimento 3 e 4). No Experimento 1, a administração de midazolam (3,0 e 30 nmol) produziu efeito ansiolítico caracterizado pelo aumento das porcentagens de entradas e tempo gasto nos braços abertos do LCE e redução de medidas etológicas (avaliação de risco) como porcentagens de mergulhos e esticadas protegidas. No Experimento 2, os camundongos receberam injeção combinada de Salina+Salina, Salina+MDZ, ondansetron (OND)+Salina e MDZ (30 nmol) + OND (0,03 nmol, antagonista dos receptores 5-HT3). O tratamento combinado de Sal+MDZ produziu, efeito ansiolítico sendo este revertido pela administração combinada de OND+MDZ. As porcentagens de entradas e tempo gasto nos braços abertos foram menores (P>0,05) do que àqueles encontrados no grupo Sal+MDZ. O Experimento 3 mostrou os efeitos da exposição dos camundongos na presença 12 do rato de brinquedo (RB= estímulo neutro) ou rato de verdade (RV= estímulo aversivo, rato Long Evans), sob a administração de Midazolam (3,0 e 30 nmol). Os animais expostos ao RV apresentaram diminuição do tempo de exploração da caixa (efeito do modelo) quando comparados aos animais expostos ao RB. Para os animais tratados com MDZ nas duas doses, houve aumento nas transições entre os lados do aparato, aumento do tempo na área de exploração e aumentos da latência de fuga para a área protegida e tempo de contato com a grade. No Experimento 4, foi avaliado o efeito da injeção combinada de midazolam e ondansetron (conforme descrito no Experimento 2). O MDZ 30 nmol produziu efeito ansiolítico e a administração combinada de ondansetron e midazolam no hipocampo ventral, reverteu este efeito. Os dados deste trabalho sugerem que, o controle sobre as reações emocionais e de defesa do hipocampo ventral de camundongos expostos ao LCE ou ao teste de exposição ao rato são mediados via receptores GABA-Benzodiazepínicos. Além disso, provavelmente ocorre modulação cruzada entre os interneurônios GABAérgicos e os serotoninérgicos do tipo 5-HT3, pois o bloqueio desses receptores com o ondansetron, pode diminuir a liberação de GABA.

Effets motivationnels des cannabinoïdes dans un modèle animal de la schizophrénie

Depuis quelques décennies, la consommation de cannabis et son usage thérapeutique sont le sujet de nombreux débats. Le cannabis est la drogue illicite la plus consommée au monde et cette consommation se trouve dix fois plus élevée chez les patients atteints de schizophrénie que dans la population générale. L’hypothèse d’une automédication initialement proposée afin d’expliquer la consommation élevée de cannabis chez les patients atteints de schizophrénie est maintenant remise en question. En effet, les rapports indiquant une aggravation des symptômes plutôt qu’une amélioration suite à une consommation à long terme sont de plus en plus nombreux. Sachant que le cannabis peut induire des effets soit plaisants soit aversifs, la question se pose à savoir si une prédominance de la valence motivationnelle positive ou une diminution de la valence négative du cannabis peut expliquer la consommation élevée parmi les individus ayant un diagnostic de schizophrénie? Bien qu’un grand nombre de recherches pré-cliniques aient été menées chez l’animal normal pour évaluer l’effet motivationnel du Δ9-tétrahydrocannabinol (THC) et autres cannabinoïdes synthétiques, aucune n’a abordé cette problématique dans un modèle animal de la schizophrénie. Cette lacune nous a donc amené à étudier la valence motivationnelle du THC et de l’agoniste cannabinoïde WIN55,212-2 (WIN) dans un modèle animal de la schizophrénie: la lésion néonatale de l’hippocampe ventral (NVHL). Dans le premier article, nous présentons les résultats de quatre expériences. Une première avait pour objectif de déterminer si la procédure expérimentale que nous avons utilisée permettait de reproduire des signes distinctifs du modèle animal de la schizophrénie. Par la suite, nous avons évalué i) l’effet d’une dose de WIN sur l’activité locomotrice spontanée et ii) la valence motivationnelle du THC (0.5 mg/kg, i.p) et du WIN (1 mg/kg, i.p) chez les rats adolescents (jour post-natal 28-40, PD28-40) et adultes (PD56) au moyen du paradigme de préférence de place conditionnée (PPC). Tel qu’attendu, la réponse locomotrice à l’amphétamine (0.75 et 1.5 mg/kg) chez les rats NVHL adultes était supérieure à celle des rats contrôles (test distinctif du modèle). Le THC a induit une tendance aversive chez les rats contrôles adultes. Enfin, le WIN a stimulé l’activité locomotrice et induit une aversion significative chez les rats adultes NVHL. Dans un deuxième article, nous avons évalué la valence motivationnelle du THC (0.5 mg/kg), du WIN (1 et 3 mg/kg) et l’effet de l’amphétamine au moyen du paradigme d’autostimulation électrique intracérébrale (ASI). Les résultats montrent que : i) l’effet amplificateur de l’amphétamine sur l’ASI était de plus courte durée chez les rats NVHL; ii) le THC produit une légère atténuation de la récompense chez les rats contrôles tandis que le WIN a produit une atténuation plus prononcée de la récompense chez les rats NVHL, un effet qui a été bloqué par l’antagoniste aux récepteurs CB1, le AM251 (3 mg/kg). Pour la première fois les résultats suggèrent une altération du système endocannabinoïde dans un modèle animal de la schizophrénie. Ils indiquent qu’une exposition aigüe conduit à une prédominance de la valence négative. Bien qu’en apparente contradiction avec les études cliniques, ces résultats soulignent l’importance du contexte socio-environnemental pour expliquer les effets du cannabis chez les patients. De plus ils encouragent les futures études à évaluer cette valence sur un modèle d’exposition chronique. / Over the past few decades, the cannabis consumption and its therapeutic use have been the subject of many debates. Cannabis is the most widely used illicit drug and among patients with a diagnosis of schizophrenia, its consumption is ten times higher than in the general population. The self-medication hypothesis that has been initially proposed to account for the co-morbidity schizophrenia – cannabis is now questioned on the basis of several reports showing that long term cannabis consumption worsen schizophrenia symptoms in patients. Knowing that cannabis can provoke both rewarding and aversive effects in human and in animal, the following question can be raised: can co-morbidity schizophrenia – cannabis be explained by a salient positive or a blunted negative motivational valence of cannabis? Even though many pre-clinical studies have been carried out in normal animals on the motivational effects of Δ9-Tetrahydrocannabinol (THC) or other synthetic cannabinoids, none has measured these effects in an animal model of schizophrenia. On the basis of this, we undertook a series of studies on the motivational valence of THC and the cannabinoid agonist WIN55,212-2 (WIN) in an animal model of schizophrenia : the neonatal ventral hippocampus lesion (NVHL). In the first report, we present the results of four studies. The first was aimed at showing that the experimental procedures that we used reproduced some abnormal features of the animal model. Then we evaluated i) the effect of WIN (1 mg/kg) on spontaneous locomotor activity and ii) the motivational valence of THC (0.5 mg/kg) and WIN (1 mg/kg) in the young (post-natal day 28-40, PD28-40) and adult (PD56) rats with the conditioned place-preference paradigm (CPP). As expected, amphetamine produced a higher locomotor activity in NVHL rats, an effect observed at PD56 and not at PD35 (NVHL usual test). THC tended to induce an aversion in control rats at PD56 while WIN produced a significant aversion at PD56 in NVHL rats only. We also assessed, in a second report, the valence of THC (0.5 mg/kg) and WIN (1 and 3 mg/kg), and amphetamine (0.75 mg/kg) using the brain stimulation reward paradigm. Results show that i) the enhancement effect of amphetamine on reward was shorter in adult NVHL rats; ii) THC induced a weak reward attenuation in control rats while WIN produced a marked dose-dependent attenuation in NVHL rats; this effect of WIN was blocked by AM251 (3 mg/kg), an antagonist at CB1 receptors. For the first time, these results suggest that the endogenous cannabinoid system is altered in this animal model of schizophrenia. They indicate that an acute exposure leads to a predominance of negative valence. Even if this seems contradictory with clinical studies, these results highlight the interconnection between the drug and the socio-environment aspects. In addition, they encourage future studies to evaluate this valence on a chronic exposure paradigm with this animal model of schizophrenia.
