The final thesis deal with possible use of audiovisual movie in Tatra mountains, specifically Starý Smokovec village. That is a model area of whole mountains. The key words as a landscape, structure of landscape and characteristic features of landscape are being summarized in text of the thesis and perception of the landscape and scenery as well. Evaluation of characteristic landscape features and its connection with development of environment are being the basis for the thesis. Limits and problems in the model area where defined according to study of prior problems and terrain research. Results of analysis are defined in text of the thesis as well as in attached map. Studium fytoplazmy ESFY a jejich projevů u odrůd meruněk a broskvoníKopřivová, Vladimíra January 2014 (has links)
The aim of this study was to obtain detailed knowledge of the symptomatic expression of various isolates European Stone Fruit Yellows phytoplasma (ESFY) on selected rootstocks, susceptibility testing of selected rootstocks to phytoplasma ESFY and exploration of the extent and impact of phytoplasma ESFY to the biological properties of pollen, seeds, some phenological and pomological characters of varieties of apricot fruit. After three years of observation of pomological characterictics of fruits from ESFY infected trees and comparing to pomological characteristics of fruits from healthy trees of 16 apricots varieties the most susceptible varieties to ESFY were infected trees Arzami Aromatnyj, Hatif Colomer, Veselka and Vestar. These varieties were significantly reduced overall size and weight of fruit due to present ESFY phytoplasma. During the research no shrunken or deformed fruits were found, fruits from infected trees were just rounder than fruit from healthy trees. Effect of ESFY phytoplasma on pollen germination, early flowering, ripening time, soluble dry matter of the fruit and seed viability was not so significant. Symptomatic manifestation of the annual shoots grown through the inocula, originating from nine selected infected trees of different varieties of apricots and one peach tree and symptomatic manifestation of plants from five different rootstocks, which were inoculated, was during the three years of observing very diverse. It was found, that it is dependent on botanical species, variety, rootstock, growing conditions and the interactions of the pathogen and the host. The most susceptible annual shoots to present ESFY phytoplasma were the grown from inocula sampled from infected tree variety Jantze peach and Murfatlar apricot. Chlorotic leafroll, yellows and growth depressions appeared most on annual shoots. The least susceptible response to ESFY phytoplasma was demonstrated on shoots grown through from inoculum originating from apricot tree variety Poyer. In this variety there were the most numerous shoots without symptoms compared with other varieties. In most varieties (Jantze, Saldcot, Poljus Južnyj a Hargrand_2) with a higher frequency of positive shoots the chlorotic leafroll was the most represent symptom, so it will be a typical symptom for ESFY pyhtoplasma. Symptomatic manifestation of rootstock plants in combination with inocula from ten selected varieties reached lower frequency of evaluated characteristics compared to annual shoots. Based on the resulting reaction of shoots grown through inocula of tested varieties and reaction of rootstock plants to presence of ESFY phytoplasma, the most susceptible rootstock was GF 305, susceptible rootstock was GF/8/1, less susceptible was M/LE/1 and the least susceptible rootstocks were Torinel and St.Julien 655/2. Sensitivity reaction of rootstocks file to present ESFY phytoplasma was observed for three years at fifteen selected rootstocks, whose plants were inoculated by two inocula from different varieties. Rootstocks plants without any symptoms had the highest frequency (84.37 %), and the most widespread symptom was the leaf yellows (average 10.5 %). Rootstocks were divided into four groups according to their susceptibility to ESFY phytoplasma. The most susceptible response was manifested on rootstocks GF 305 and GF 677, rootstocks MRS 2/5, VVA1, Strážovický myrobalán, Lesiberian, AP1 and GF 31 were susceptible, rootstocks M/LE/1 and Myrobalán 29C were less susceptible and the least susceptible rootstocks to ESFY phytoplasma were Torinel, St. Julien 655/2, GF/8/1 and MY/KL/A.

This thesis examines the growth and harvesting characteristics of the non-traditional fruit species of dogwood. The work in the explanatory section deals with the origin of this fruit shrub, its distribution, the demands it makes on the environment, its growth properties and propagation technology. Then it describes the chemical composition of the fruit and possibilities for using it in the food industry or in horticulture. The selection of six dogwood species (´Elegant´, ´Fruchtal´, ´Jolico´, ´Lukjanov´, ´Vydubec´ and ´Vyshegorod´) was evaluated at an experimental area of the breeding institute in Zabcice. For each variety, we examined and compared the extent of the phenological phases, the size of the bush volume, the size of the fruit harvest and both external and internal features of the fruit. This last observation involved evaluation of the length, width and weight of the fruit, the proportions and weight of the stone, plus the levels of vitamin C, dry matter and refractometric dry matter. The results obtained from the measurements were processed and statistically evaluated. Through a combination of our results and reference to expert studies, we are able to recommend the most suitable varieties for cultivation under our climatic conditions.

163 Abstract The paper is concerned with Karel Čapek's dramas, Loupežník, R.U.R., Věc Makropulos, Bílá nemoc and Matka. It analyzes systematically the structure of dramas, especially from the semiotic and stylistic point of view. Both disciplines are considered in quite a broad sense, hence at first we examined their basic characteristics, and then clarified how Čapek's dramas actually relate with them. First of all, we focused on the meaning and the function of sign and enquired specific features of a dramatic text. Furthermore, we dealt with dramatic elements, in particular with time, space, dramatic characters, and with each element, we examined its characteristics and functions in connection both with drama and theater. We presented main and significant theories and methods of scholars. Regarding the analysis of K. Čapek's dramas from the semiotic point of view, we clarified characteristics and syntactic structure of dramatic characters in connection with the theory of actants. Such syntactic structure makes it possible to clarify and understand the construction of a narrative and the function of characters. Development cartographic means of expression for representing castles on old maps of the Bohemia Abstract This thesis examines the development of cartographic means of expression that are used for the display of castles, palaces, strongholds and ruins on the old maps of Bohemia in the period from the early 16th century to early 18th century. The work evaluates the positional inaccuracies of location of interest sites through cartometric analysis. The first part is theoretical, in which the issues outlined cartographic semiotics and cartographic expression. Next two parts deal with the oldest cartographic references of the Bohemia, or the description of the analyzed maps. Further, the thesis illustrated the method by which the analysis are extracted. Production of ammonia by the colonies of mutants and aging of wrinkled colonies of Saccharomyces cerevisiae The aim of this diploma thesis is to observe the development, respectively the aging of cells in yeast colonies Saccharomyces cerevisiae. Yeast cells S. cerevisiea form multicellular organized structures on a solid substrate, i.e. colonies, which the intercellular interactions occur in. These interactions influence forming, morphology and aging of yeast colonies. This diploma thesis is focused partly on the changes in ammonia production by giant colonies of deletion mutants and partly on the aging of colonies with the wrinkled morphology. I characterized mutant strains of S. cerevisiae with the deletion in RTG1, RTG2, RTG3, FIS1, CIT2 genes. Their products play an important role in the colony development. The transcription of these genes changes during the transition from the acidic to alkali phase during developmental process of the colonies. I have found out that the ammonium production rate was in accordance with the results of the alkalization in giant colonies surroundings and mentioned mutants derived from the BY strain has been producing ammonia since the 15th day. The purpose of this diploma thesis is to identify organizations in a designated area. The city of Dačice has been selected. Organizational data have been acquired from publicly accessible resources -- mostly public service registers. Based on the data obtained, organizations are divided in terms of their legal form, principal activity and contribution to civic amenities in the selected area. One particular organization based in selected area is then described according to its organizational characteristics. In the conclusion of the thesis, comparison of diversity of organizations between the city of Dačice and several other cities with similar population is performed.

The broader purpose of the work is to find and interpret polymorphism in the genes of natural immunity of cattle to be used to improve disease resistance. The NGS method on the PacBio platform was applied for the resequencing of the gene for the key receptor of innate immunity TLR2 and two genes coding for the components of the downstream transcriptional factor NF-κB. In the population of 149 bulls of the Czech Simmental breed, 22 polymorphisms were found in the gene NFKB1 (5 new), while in the NFKB2 gene 13 SNP were found (10 new). 21 SNP were found in the TLR2 gene (3 new). Of the 56 found polymorphisms, 6 SNPs were nonsynonymous. One SNP leads to a change R474G in the NFKB1 product and five to changes E63D, R152Q, I211V, R563H and H665Q in the protein TLR2. Knowledge of the haplotypes facilitated the development of individual genotyping reactions. In TLR2, a high number of haplotypes was detected, both from the PacBio reads and the statistical reconstruction. In addition, two clusters of haplotypes were ditinguished inTLR2, possibly due to diversifying selection or introgression. The knowledge of genetic diversity in the population allows for the planned association studies with health data.
