The purpose of this Diploma Thesis is to identify relationships between organizations in a chosen area. It was selected the branches of UniCredit bank, a. s. in the South Bohemian Region. Organizational data have been acquired from publicly accessible resources -- mostly public service registers, the central bank of the Czech Republic's registers and Internet resources of Unicredit Bank as well and through empirical investigation by an interview method with representatives of the branches. In the conclusion of the Thesis are gained knowledge inserted into the Model of inter-organizational relationships and thank to this are characterized the relationships between these organizations.

Vliv syntetického progestinu etonogestrelu na sekundární pohlavní znaky a histologii gonád živorodky Wingeovy (Poecilia wingei)

Synthetic progestins are steroid hormones, which are not only part of the hormonal contraceptives, but they are also used to treat a number of health problems in human. Due to the widespread use of progestin-based drugs, their residues are being found in wastewaters, from where they also enter into the wastewater treatment plants and surface waters. The aim of this thesis was to assess whether the synthetic progestin etonogestrel affects the secondary sexual characteristics and gonad histology of Endler´s guppy (Poecilia wingei). During the experiment, males and females were separately exposed to 3.2 and 320 ng.l-1 etonogestrel for 34 days. A group of fish reared in pure water (K) and group of fish reared in pure water containing the solvent dimethylsulfoxide (KS) served as controls. At the end of the test, all the fish were photographed to capture any color changes. Then the fish were sacrificed and fixed in 10 % buffered formalin solution for morphometric and histological analysis. Fish samples taken for morphometric analysis were photographed using QuickPHOTO MICRO 2.3 software and individual morphometric parameters were measured on the images taken. Fish sample processing for histological analysis included decalcification, dehydration, and pouring of the samples into paraffin blocks. Then paraffin blocks were cut with the thickness of about 4.5 um, resulting slides were stained with hematoxylin and eosin. Histological changes of the gonads were determined on the histological samples prepared in this way. In females exposed to both concentrations of etonogestrel occurrence of color spots was observed. Color spots are characteristic for male and in females indicate masculinization. In addition, females exposed to 320 ng.l-1 etonogestrel showed a change in anal fin morphometry to a gonopodium-like structure (modified anal fin of males serving as helping copulatory organ), a sign also suggesting masculinization. Furthermore, exposure of etonogestrel in both lower and higher concentration of etonogestrel reduced number of mature oocytes in the ovaries, and their occurrence decreased with increasing concentration. In males exposed to both concentrations of etonogestrel a smaller ratio of the length of 4th ray to the length of 6th ray of gonopodium was found. In addition, at concentration of etonogestrel 320 ng.l-1 a greater ratio of the length of 6th ray to body length and extension of length of 6th ray was observed. Although there were no changes, which would indicate hypermasculinization, these changes could have a negative impact on reproduction, or sexual selection by the female. Etonogestrel exposure had no effect on the normal development of testes, because predominantly mature spermatophores were found in males of all treatment groups. Based on these findings, we can say that etonogestrel, like other progestins, represents a real risk to fish if it occures in aquatic environment at concentrations reaching units of ng.l-1.

Colorectal cancer is one of the most common type of malignity. Despite of the existence of numerous studies focused on this carcinoma, there are still many unknown features regarding its diagnosis, treatment or prognosis. In the thesis we focused on the identification of novel prognostics markers that could be useful for the stratification of patients based on the disease outcomes. In the first study we immunohistochemically assessed expression of two proteins associated with cancer stem cells in the samples of primary colorectal cancer and matched liver metastasis. Goal of the study was to evaluate relation among expression of CD44 and CD133 and overall survival and disease free interval in our set of patients. We observed that increased ratio of CD133 positive compared to CD133 negative tumor glands resulted in longer disease free interval, finding which is opposite to the general view on the CD133 role in the cancer development. Our hypothesis is that we analyzed confined group of patients and followed a bit different goal, where we measured ratio between positive and negative glands in the view-field and not the intensity of staining as the previous studies did. The aim of this thesis was to obtain initial data about the biology and variability of morphological characters of Festuca amethystina. The first part contains a literature outline while the second part is practical. The terrain work was carried out in the forest of Hodonínská Doubrava, in the southeast part of Moravia and involved finding and surveying microlocations of F. amethystina. The collected data focused on the size of micropopulations and demographic and morphometric characteristics of the species, i.e. the diameter and number of clumps, leave length, height and number of stems, panicle size, number of spikelets and number of caryopsis. The outcomes of this work contain tabular and graphical summaries of the measured data, maps and photographs.

Cardiac surgery is known to initiate a complex physiological response with the immune system activation (SIRS), neurohormonal response, metabolic changes, coagulopathies etc. SIRS is triggered by tissue injury, myocardial ischemia, reperfusion, use of anaesthesia, cardioplegia, extracorporeal circuit etc. Excessive immune system activation is associated with progression of SIRS, life-threatening multi-organ dysfunction (MOD), and increased morbidity/mortality in the postoperative period. The immune system response is regulated and terminated by both cellular and humoral regulatory and inhibitory mechanisms including changes in expression of in our study monitored molecules: CD200/CD200R, sCD200R and CD95/CD95L. Methods: The study included the measurement the expression of CD95, CD95L, CD200R, and sCD200R molecules in granulocyte and monocyte populations in blood samples of 30 patients who underwent heart surgery using CPB. Samples collected before surgery, after surgery, and in the postoperative period (1st , 3rd , 7th day) were analysed by flow cytometry and sCD200R by ELISA. Results: We discovered a significant increase in the percentage of granulocytes expressing inhibitory molecule CD200R (from 5% to 17.8%) instantly after surgery. Nowadays intensive research is conducted for application of genetic polymorphisms for degraded samples analysis, identification and kinship determination. Another area of research in forensic genetics is biogeographical and phenotypic traits (eye, hair and skin color) determination. First part of presented work dealt with population study on the Czech popu- lation using Investigator DIPplex (QIAGEN, Germany) marker set containing 30 autosomal insertion-deletion polymorphisms. Power of Discrimination (PD), which is the probability of random selection of two persons with different genotypes, was 99.9999999999% for the whole marker set. This part of study concluded that ana- lyzed marker set is suitable as an additional marker panel for identification and kinship determination in the Czech Republic. Second part of the presented study was devoted to population research of Cen- tral Croatia using Mentype Argus X-8 kit (QIAGEN, Germany) containing 8 short tandem repeat polymorhisms located on X choromosomes (X-STR) divided into 4 linkage groups. PD for the whole kit reached 99.9999% and 99.99999999% for males and females, respectively. The aim of the Diploma Thesis is to analyze non-profitable organizations according to the organizational characteristics in a chosen locality and to judge this type of organizations regarding their possible common or different characteristics. Some selected non-profitable organizations in the town of Chrudim are the aggregate research. Each organization is analyzed according to its organizational characteristics, these are the instrumentality, formality, structure and culture. The data have been found mainly in publicly accessible databases like the register of public administration and web pages of these organizations. In conclusion I compare the organizational characteristics of each single organization and assess the potential accordance of these characteristics and I also consider some pattern for non-profitable organizations to work and to be structured.
