Estas fibras possuem uma elevada plasticidade sendo capazes de alterar seu fenótipo em resposta a vários estímulos, como o treinamento físico. Tem sido demonstrado que o treinamento aeróbico frequentemente induz a pequenas alterações na massa muscular, promovendo mudanças no perfil do músculo em direção a um padrão mais eficiente. Vários estudos têm sido realizados para tentar identificar as vias moleculares envolvidas com a adaptação muscular. A maioria destes estudos tem procurado entender os fatores que controlam as alterações no fenótipo muscular, incluindo o fator de crescimento semelhante à insulina-I (IGF-I), a miostatina (MSTN) e a calcineurina (CaN). O objetivo deste estudo foi testar a hipótese de que as alterações que ocorrem em músculos fenotipicamente distintos durante o treinamento aeróbio de longo prazo são regulados por fatores de crescimento miogênicos, como o IGF-I, MSTN e CaN A e B. Ratos Wistar machos (80 dias de idade, 250-300 g) foram divididos em dois grupos: treinado (T; n = 8) e controle (C, n = 8). Os animais do grupo T foram submetidos a um programa de treinamento aeróbio de natação de longo prazo (8 semanas, 5 dias / semana). O volume e intensidade do treino foram progressivos: 10 min, sem sobrecarga (1ª semana), 20 min, com sobrecarga de 1% (2ª semana), 25, 30, 35 e 40 min, 3% de sobrecarga (3ª semana), 45, 50, 55, e 60 min, 5% de sobrecarga (4ª semana) e 60 min, 5% de sobrecarga (5ª a 8 semanas). No final do experimento os animais foram pesados e sacrificados. Muscle fibers have a high plasticity and are dynamic structures liable to modify their phenotype in response to several stimuli, such as physical training. It has been shown that aerobic training frequently leads to minor changes in muscle mass, promoting changes in muscle profile toward a more efficient pattern. Several studies have been performed to try to identify the molecular pathways involved in the muscle adaptation. Most of them converge to understand the factors that control the muscle phenotype changes including the insulin-like growth factor-I (IGF-I), Myostatin (MSTN) and Calcineurin (CaN). The purpose of this study was to test the hypothesis that muscle phenotypically distinct changes that occur during long-term aerobic training are regulated by myogenic growth factors including IGF-I, MSTN and CaN A and B. Male Wistar rats (80 days old, 250-300 g) were divided into two groups: trained (T; n=8) and control (C; n=8). T group underwent a swimming aerobic training program for 8 weeks (5 days/week). Training volume and intensity were progressive: 10 min without overloading (1st week), 20 min, 1% overload (2nd week), 25, 30, 35, and 40 min, 3% overload (3rd week), 45, 50, 55, and 60 min, 5% overload (4th week and 60 min, 5% overload (5th to 8th weeks). At the end of the experiment animals were weighed and sacrificed. Soleus (SOL) and Plantaris (PL) muscles were removed and processed for histochemical, morphometrical, biochemical and molecular analyses. No significant difference was observed in the initial body weight among the groups (p >0.05).
