CoordenaÃÃo de AperfeÃoamento de Pessoal de NÃvel Superior / Assess the effects of oral administration of GLN on oxidative stress in women with breast cancer undergoing chemotherapy preoperative neo-adjuvante with FAC (5-500 fluouracil mgm2 50 mgm2 500 mgm2 cyclophosphamide doxorubicin of body surface area). Methods: Twenty women (average age: 51.7 years) with ductal carcinoma of the breast, classified as clinical stage IIb, IIIa, IIIb or IIIc, were included in the study, regardless of its State. Packages containing 15 g of glutamine or milk protein were prepared by a pharmacist. Allocation of patients was made following generated by "softeware. Patients receiving packaging type "A" are included in the group G1. Patients treated with the preparation called "B", were included in Group G2. The material was mixed with use of Blender in 150 ml of water, and administered daily to patients during the entire course of neoadjuvant chemotherapy (FAC). Peripheral blood samples were collected at the beginning of the three cycles of chemotherapy, before the infusion of drugs. Samples of tumor and normal breast tissue were harvested at the end of the surgical procedure (Patey surgery). The samples were analyzed to determine the concentration of GSH and RESULTS. Results: TBARSGSH concentrations were not different in the tumor, breast tissue or blood, comparing the groups G1 vs G2. In addition, no significant differences were found in the RESULTS CONCERNING levels and GSH comparing different times within the same group. Conclusion: GLN (grdia 15) orally administered does not provide protection against systemic or local oxidative stress in women with breast cancer, undergone preoperative neoadjuvant chemotherapy (FAC). / Avaliar os efeitos da administraÃÃo oral de GLN sobre o estresse oxidativo em mulheres portadoras de cÃncer mamÃrio submetidas a quimioterapia neo-adjuvante prÃ-operatÃria com o esquema FAC (5-fluouracil 500 mg/m2 + doxorubicina 50 mg/m2 + ciclofosfamida 500 mg/m2 de superfÃcie corporal). MÃtodos: Vinte mulheres (idade mÃdia: 51,7 anos) com carcinoma ductal da mama, classificadas como estÃdio clÃnico IIb, IIIa, IIIb ou IIIc, foram incluÃdas no estudo, independente do seu estado menstrual. Embalagens contendo 15 gr de glutamina ou proteÃna do leite foram preparadas por farmacÃutico. AlocaÃÃo das pacientes foi feita na sequÃncia gerada por âsoftewareâ. Pacientes que receberam embalagens tipo âAâ foram incluÃdas no grupo G1. Pacientes tratadas com a preparaÃÃo denominada âBâ, foram incluÃdas no grupo G2. O material foi misturado com uso de liquidificador em 150 ml de Ãgua potÃvel, e administrado diariamente Ãs pacientes durante todo o curso da quimioterapia neoadjuvante (esquema FAC). Amostras de sangue perifÃrico foram coletadas no inÃcio dos trÃs ciclos de quimioterapia, antes da infusÃo das drogas. Amostras de tumor e tecido mamÃrio normal foram colhidas no final do procedimento cirÃrgico (cirurgia de Patey). As amostras foram analisados para determinaÃÃo das concentraÃÃes de GSH e TBARS. Resultados: ConcentraÃÃes de TBARS/GSH nÃo foram diferentes no tumor, tecido mamÃrio ou sangue, comparando os grupos G1 vs. G2. AlÃm disso, nÃo foram encontradas diferenÃas significativas nos nÃveis de TBARS e GSH comparando momentos diferentes dentro do mesmo grupo. ConclusÃo: GLN (15 gr/dia) administrada por via oral nÃo oferece proteÃÃo contra estresse oxidativo sistÃmico ou local em mulheres com cÃncer de mama, submetidas a quimioterapia neoadjuvante prÃ-operatÃria (FAC).
