We analyzed samples of fish species Characidium cf. fasciatum, C. cf. zebra, C. cf. lagosantense, Characidium sp. and three strains of C. cf. gomesi, representatives of the subfamily Characidiinae and a sample of Crenuchus spilurus, representative Crenuchiinae, using conventional cytogenetic techniques for staining with Giemsa, location of NORs by impregnation with silver nitrate, C-banding and molecular cytogenetic techniques for fluorescence in situ hybridization with probes of 18S and 5S rDNA, probes produced from the sex chromosome W and a probe generated from the genome of C. cf. zebra, with emphasis on probes produced from the sex chromosome W. All species of Characidium showed a diploid number of 50 chromosomes, with the predominance of chromosomes of metacentric and submetacentric types. The occurrence of supernumerary chromosomes was observed in one sample of C. cf. gomesi and also the presence of a system of sex chromosome ZZ-ZW was observed in Characidium cf. gomesi, Characidium cf. fasciatum, Characidium sp. The species Characidium cf. zebra, Characidium cf. lagosantense and Crenuchus spilurus showed no differentiated sex chromosomes. The technique of chromosome painting using probes produced by microdissected chromosomes amplified by DOP-PCR was performed to test the homeology of Z and W sex chromosomes found in seven species of Characidium. Probes were produced from the W chromosome of Characidium cf. gomesi (DCg) and Characidium cf. fasciatum (DCf). The probes showed hybridization signals on chromosomes Z and W in Characidium cf. gomesi, Characidium cf. fasciatum, Characidium sp., showing homeology between sex chromosomes in the species and a possible common origin of this sexual differentiation system in these species. Moreover, in Characidium cf. zebra, Characidium cf. lagosantense and Crenuchus spilurus the hybridization with ...(Complete abstract click electronic access below)
