Das Ziel dieser Studie war es, die Pathomechanismen der Enzepahlopathie, welche während einer akuten Nierenschädigung beobachtet werden kann, zu untersuchen. Wir konzipierten ein Zellkulturmodell, welches aus der cerebralen Endothelzelllinie cEND und der Nierentubuluszelllinie HK-2 bestant. Die Nierenzellen wurden einem Sauerstoff-Glukose-Entzug zugeführt, um eine akute Nierenschädigung zu simulieren. Nach weiterer Behandlung mit zwei urmämischen Toxinen (Indoxylsulfate und Indolessigsäure) und gemeinsamer Inkubation beider Zelllinien für insgesamt 48 Stunden wurden die cEND-Zellen geerntet und die Expression von Tight junction-Proteinen und Glukosetransportern untersucht. / The aim of this study was to examine the pathogenesis of the encephalopathy that is observed during acute kidney injury. We created a cell culture model consisting of a cerebral endothelial cell line (cEND) and a kidney cell line (HK-2). The kidney cells were damaged by an oxygen-glucose deprivation (OGD) to simulate an acute kidney injury. After treatment with two uremic toxins (indoxyl sulfate and indolic acetic acid) and joint incubation of both cell lines for 48 hours the cEND cells were harvested and the expression of tight junction proteins and glucose transporters was investigated.
