Resumo: Na espécie Casearia sylvestris Sw. (Flacourtiaceae) são consideradas duas variedades botânicas: C. sylvestris var. língua, com característica arbustiva e ocorrência em savanas arbustivas e C. sylvestris var. sylvestris, de caráter arbóreo em vegetação florestal ou arbustiva. A literatura afirma ser bastante problemático definir os limites destas variedades através de parâmetros morfológicos sendo portanto, importante identificar diferenças genéticas e fitoquímicas entre estas. Este trabalho teve como objetivo identificar em seis populações naturais de Casearia sylvestris de ocorrência no Cerrado e Mata Atlântica do estado de São Paulo diferenças genéticas e químicas entre as variedades lingua e sylvestris, realizando estudo da variabilidade genética da espécie, por meio de marcador molecular do tipo RAPD. Foram coletados dados de localização geográfica (altitude, latitude e longitude). As extrações de DNA foram feitas a partir de 100 mg de folhas utilizando o método de extração CTAB. Nove primers foram utilizados para aplicação da técnica. A partir de matriz binária de similaridade gerou-se o dendrograma pelo índice de Jacard, utilizando o método de agrupamento UPGMA. A correlação das distâncias geográficas com distâncias genéticas foi obtida pelo teste de Mantel, enquanto que a análise de variância molecular (AMOVA) e o índice de Shannon foram utilizados para verificar as diferenças entre e dentro das populações. Folhas da espécie foram submetidas à extração em metanol, e a quantificação de rutina foi realizada utilizando um cromatográfo líquido de alta eficiência. Praticamente toda variabilidade da espécie é representada pela variabilidade intrapopulacional e a análise de agrupamento mostrou maior similaridade entre as populações de um mesmo bioma, revelando uma diferença genética entre as duas variedades botânicas... (Resumo completo, clicar acesso eletrônico abaixo). / Abstract: Two botanical varieties have been considered in Casearia sylvestris species: C. sylvestris, lingua variety with a shrubby characteristic, occurring in bushy savannah and C. sylvestris, sylvestris variety with an arboreous type in forest or shrubby vegetation. The limits of these varieties based on morphological parameters are vey tenuos, making molecular and phytochemical markers very important to identify. This work aimed to identify genetic and chemical diffrences between lingua and sylvestris varieties in six natural populations of Casearia sylvestris from Cerrado and the Atlantic Forest in São Paulo state, using (RAPD) molecular marker and chemical markers. Samples have been collected in order to analyze geographic location data (altitude, latitude and longitude). DNA extraction was performed using 100 mg of leaves through the CTAB procedure. Nine primers were used for the technique application. The dendrogram was obtained from the matrix of pairwise distances through the Jaccard's similarity index using the UPGMA grouping method. A Mantel test was used to analyze correlations between genetic and geographical distances, while the molecular variance analysis and the Shannon index were used to determine the intra- and interpopulation divergences. For the phytochemical study leaves of the species were submitted to extraction in methanol and rutin quantification was carried out using a high-performance liquid chromatographer. Practically all species variability are represented by the intrapopulation variability and the grouping analysis showed higher similarity among populations of the same biome, revealing a genetic diversity between the two botanical varieties. The majority of soils where was collected"guaçatonga" is present is of high acidity and predominantly of medium texture. Nevertheless, most soil indicator values showed diversity among populations of savannah and forest...
