Previous issue date: 2005-04-14 / Bovinos da raça holandesa entre 3-4 meses de idade receberam quatro imunizações subcutâneas do peptídeo sintético 23290 anti-Babesia bovis mais saponina, e foram desafiados com amostra virulenta de B. bovis, sendo avaliados alguns parâmetros clínicos como hematócrito, hemoglobina, parasitemia e temperatura, além do acompanhamento do desenvolvimento da resposta imune humoral, mediante quantificação de IgG total e identificação de isótipos IgG1 e IgG2 antígeno- específicos. Os parâmetros hematológicos e a temperatura foram analisados semanalmente no período de imunização, e diariamente a partir do 7o dia pós-desafio com o parasito; Os exames sorológicos foram realizados semanalmente durante todo o experimento. As análises sangüíneas demonstraram que SBbo 23290 minimizou o impacto da infecção pela B. bovis, além de inibir a carga parasitária. Os estudos sorológicos constataram que o SBbo 23290 estimulou a produção de imunoglobulinas G antígeno-específico, com predominância estatisticamente maior de isótipos IgG1 sobre IgG2 bem como elevação da produção de IgG total. Com relação aos parâmetros clínicos, que foram observados 18 dias pós-desafio, houve um declínio nos valores observados em todos os grupos. Quanto à temperatura, o observado foi que houve elevação significativa da mesma após o desafio com B. bovis, sendo diretamente proporcional à carga parasitária. Sendo assim, pode-se concluir que o peptídeo sintético induz de maneira eficaz uma resposta imunológica antígeno-específica, na qual encontram-se envolvidos mecanismos celulares e humorais. / Tree-four month old bovines of the Holstein Freesan breed received four under–skin immunization of the synthetic peptideo 23290 anti-Babesia bovis plus saponina. They were challenged with an virulent sample of B. bovis and they were evaluated considering clinic parameters such as packet cell volume, hemoglobin, parasitemy (presence of parasite), and temperature, besides, the observation of the development of the humoral immune answer through identification of specific-antigens isotypes IgG1 and IgG2, besides the total IgG . The hematological parameters and the temperature were checked every week during the immunization period, and every day after the seventh day after challenge. The serology were conducted every week during the whole experiment. The blood analyses demonstrated that SBbo 23290 minimized the impact of the infection B. bovis, besides by inhibiting the parasitical charge. The serologic studies evidenced that SBbo 23290 stimulated the production of antigen-specific immunoglobulin G. Showing a predominance of isotypes IgG1 statistically higher than IgG2 a well as an increase of the production of total IgG. Considering the clinical parameters, observed 18 days after challenge, a decrease of values was observed in all groups. On the other hand, the temperature became meaningfully higher after the challenge with B. bovis, at the same rate of the parasitical charge. We can conclude that the syntetic peptideo induct an effective immunological specific-antigen answer, in with cellular and humoral mechanisms were involved. / Dissertação antiga, importada do Alexandria.
