Previous issue date: 2006 / Objetivo: Há desenvolvimento e crescimento de nova mucosa intestinal sobre diferentes superfícies de apoio. Os biomateriais, principalmente as matrizes acelulares, constituem estruturas ricas em proteínas de sustentação, como o colágeno, e permitem o desenvolvimento de neomucosa do intestino delgado. O objetivo do presente estudo foi analisar a morfologia de neomucosa do intestino delgado formada sobre retalho de matriz tecidual acelular. Métodos: Vinte e oito ratos machos Wistar foram utilizados no estudo. Três animais forneceram o intestino delgado para a obtenção da matriz acelular.
Aleatoriamente, os outros 25 animais foram distribuídos em três grupos: eutanásia com duas semanas – cinco ratos; eutanásia com quatro semanas – dez ratos; e eutanásia com oito semanas pós-operatórias – dez ratos. A matriz acelular foi obtida do tratamento detergente-enzimático de segmentos de intestino delgado, com deoxicolato de sódio 4% e deoxirribonuclease. Os retalhos de matriz acelular foram interpostos na parede antimesentérica do íleo terminal. Foram analisados os dados morfométricos da mucosa adjacente normal do íleo e da nova mucosa em desenvolvimento. Resultados: Com oito semanas de período pós-operatório, todo o retalho estava preenchido por nova mucosa. A neomucosa apresentou valores morfométricos
estatisticamente maiores que a mucosa adjacente. Conclusão: A neomucosa intestinal formada sobre matriz acelular obtida pelo
tratamento detergente-enzimático do intestino delgado de ratos é semelhante à
mucosa normal adjacente. ___________________________________________________________________________________ ABSTRACT / Background / Purpose: New intestinal mucosa can grow and be developed at different scaffolds. The acellular matrix is made off rich sustain protein and it allows the development of small bowel neomucosa. The purpose of this study was to analyze the morphology of the small bowel neomucosa at the acellular matrix tissue graft. Methods: Twenty-eight Wistar male rats were enrolled in the study. The small bowel of three animals was used to obtain the acellular matrix. Twenty five other animals were randomly assigned in three groups according to the time of post-operative euthanasia. In five rats the euthanasia happened at two weeks, in ten rats at four weeks and in ten rats at eight weeks. The acellular matrix was
extracted from a detergent-enzymatic process at small bowel segments with sodium deoxycholate at 4% and deoxyribonuclease. The acellular matrix grafts were interposed at the terminal ileum antimesenteric wall. The morphometric characteristics of adjacent normal ileum mucosa and the developmental
neomucosa were analyzed. Results: After eight weeks post operatory, all grafts were filled with new mucosa. The neomucosa showed morphometric values statistically bigger than
the adjacent mucosa. Conclusion: The intestinal neomucosa developed under acellular matrix, which is obtained by a detergent-enzymatic process of the small bowel in rats, is quite normal as the adjacent intestinal neomucosa.
