Para tanto, foram utilizadas quarenta e nove amostras de sêmen de 20 carneiros da raça Santa Inês. Os estudos foram divididos em três experimentos. O primeiro comparou as duas concentrações de GLI em meio com ou sem a TRE, associada ou não ao EDTA. O segundo experimento avaliou a DMA em duas concentrações (3 e 6%) em meio com ou sem TRE, associada ou não ao EDTA. O terceiro experimento testou os dois diluidores com os melhores resultados in vitro no experimento 1 e os dois do experimento 2. Todos os experimentos tiveram os parâmetros da cinética espermática avaliados subjetivamente e pela análise computadorizada (CASA). A análise da viabilidade espermática foi realizada através da associação das sondas fluorescentes iodeto de proprídio, JC-1 e FITC-PSA. Para a análise estatística das características avaliadas, foi empregado o programa estatístico (SAS), versão 5.0 (1996). A DMA a 6% apresentou toxicidade e a 3% demonstrou baixo efeito crioprotetor sobre o espermatozóide ovino. A adição da TRE promoveu efeito negativo sobre a cinética espermática, entretanto melhorou os níveis de integridade das membranas espermáticas pós-descongelação. O diluidor com GLI a 6% sozinho foi o protocolo mais eficaz para a manutenção da integridade espermática, entretanto a taxa de prenhez das fêmeas não diferiu entre os grupos / The objective of this study was to evaluate the efficacy and the effect of the cryoprotectants glycerol (GLY) and dimethyl acetamide (DMA) in two grades (3 and 6%) in extender with and without trehalose (TRE) extender associated or not with EDTA, on the post-thawing sperm viability. For that, they were used forty nine semen samples of 20 rams of the Santa Inês breed. The studies were divided into three experiments. The first one compared the two GLY grades (3 and 6%) in extender with or without TRE, associated or not with EDTA. The second experiment evaluated the DMA at two concentrations (3 e 6%) in medium with or without TRE, associated or not with EDTA. The third one tested the two extenders with the best in vitro results in experiment 1 and the two from experiment 2. All experiments had the kinetics sperm parameters evaluated subjectively and by computer analysis (CASA). The sperm viability evaluation was performed through the multiple sperm parameter staining with propidium iodide, JC-1 and FITC-PSA. For the statistical analysis of the characteristics evaluated was used the statistical package (SAS), version 5.0 (1996). The DMA at 6% was toxic and at 3% had a poor cryoprotective effect for ram semen. The addition of TRE promoted negative effect on sperm kinetics, entertain improved levels of sperm membranes integrity post-thawing. The extender with GLI at 6% alone was the best protocol for maintaining the sperm integrity, however the pregnancy rates of the ewes did not present difference (P>0.05) among the groups
