tchamo_cclm_dr_jabo.pdf: 1695408 bytes, checksum: 35b843331f0d7d9684e135a19a514e9a (MD5) / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) / Tendo em conta a importância do sangue do cordão umbilical (SCU) como fonte de células-tronco objetivou-se este estudo, visando quantificar células CD117+, CD45+ e subpopulações linfocitárias; e determinar os valores eritroleucométricos bem como a contagem de reticulócitos no sangue do cordão umbilical (SCU) e sangue periférico de suínos imediatamente ao nascimento (SPI) e quarenta e oito horas depois (SP48), utilizando-se de 48 amostras de SCU e periférico de neonatos. Comparando-se os grupos, a um nível de significância de 5% na quantificação de células CD117+ a marcação foi muito baixa presumivelmente porque o CD117 marca células em estágio de diferenciação. Pelo que se recomenda que mais estudos sejam feitos, combinando-o com o marcador da verdadeira célula- tronco. As contagens de células CD5+ , CD4+ e CD8+ por citometria de fluxo, no SCU e SPI foram inferiores àquelas reportadas para o sangue periférico de adultos, indicando um componente imunológico imaturo. A proporção CD4:CD8, tanto para o SCU quanto para SPI, assim como SP48, demonstrou dominância das células TCD4+. Relativamente ao eritrograma, houve diferença significativa entre as variáveis nos grupos estudados, excetuando-se o volume corpuscular médio (VCM), hemoglobina corpuscular média (HCM) e concentração da hemoglobina corpuscular média (CHCM) que não variaram entre os grupos, SCU e SPI; a contagem de reticulócitos no grupo SP48 foi superior ao dos grupos SCU e SPI. No leucograma, todas as variáveis mostraram diferença significativa, excetuando-se a contagem de monócitos, que não variou entre os grupos, SCU e SPI. Já nos grupos SPI e SP48 foi observada diferença significativa na contagem de neutrófilos segmentados e monócitos, enquanto o total de leucócitos, neutrófilos bastonetes e linfócitos não diferiram / Given the importance of Umbilical cord blood (UCB) as a source of stem cell, this study was aimed to quantify CD117+, CD45+ and lymphocyte subsets and determine the eritroleucometric values and the recticulocyte count in umbilical cord blood (UCB) and peripheral blood of piglets immediately at birth (PBI) and forty-eight hours late (PB48), using 48 samples of umbilical cord and peripheral blood of neonates. Comparing the groups at a significance level of 5 %, in the quantification of CD117+ cells, marking was very low presumably because CD117 mark cells in differentiation stage. It is recommended that more studies be done combining it with the marker of true stem cell marker. The lymphocyte subsets counts CD5+, CD4+ even CD8+ of UCB and PBI performed by flow citometry, were lower than those reported for peripheral blood of adult pigs, indicating an immature immunologic component. The ratio CD4:CD8 obtained in this trial, both UCB, PBI and PB48 showed dominance of CD4+ T cells lymphocytes. Regarding the erythrogram there was a significant difference between variables of groups with exception for mean corpuscular volume (MCV) mean corpuscular hemoglobin (MHC) and mean corpuscular hemoglobin concentration(MCHC) that did not vary between UCB and PBI groups; the reticulocyte count in PB48 group was higher than that of UCB and PBI groups. In the WBC, all variables showed significant differences, except for the number of monocytes, which vary between UCB and PBI groups. In PBI and PB48 groups there was no significant difference in the mature neutrophils and monocyte count, while the total leukocyte, band neutrophils and lymphocytes did not differ
