tiveron_mb_me_botfm.pdf: 627008 bytes, checksum: 84b536da37b087bcb80573d412269a64 (MD5) / Fundação para o Desenvolvimento Médico e Hospitalar (Famesp) / Com o objetivo de investigar as repercussões clínicas e as alterações biomecânicas e histológicas, no tecido ósseo, causadas pela menopausa induzida por ooforectomia, foram estudadas 64 ratas, adultas e virgens, Rattus novergicus albinus (linhagem Wistar). Os animais foram divididos em dois grupos em relação ao tratamento: ooforectomia bilateral ou cirurgia placebo. O procedimento cirúrgico foi realizado aos quatro meses de idade. Cada grupo foi subdividido em quatro subgrupos, com 8 animais, segundo o tempo de pós-operatório no momento de sacrifício: três, seis, nove e 12 meses. Para o estudo clínico foi considerado o peso dos animais no momento do sacrifício. O estudo biomecânico foi realizado na tíbia direita por meio de ensaio de flexão em três pontos com velocidade de aplicação de carga de 30 mm/min, obtendo-se a carga máxima, na ruptura, e o coeficiente de rigidez, calculado a partir do diagrama carga-deformação. Para o estudo histológico, utilizou-se a tíbia esquerda (porção proximal) e análise de histomorfometria com o cálculo da área de trabéculas ósseas. A análise estatística do resultado do peso corpóreo e das propriedades mecânicas foi realizada pela técnica de análise de variância complementada com o teste de comparações múltiplas de Tukey; o estudo da área trabecular foi feito pela análise de variância não paramétrica. A análise dos resultados obtidos permitiu concluir que a menopausa induzida pela ooforectomia bilateral causou aumento do peso corpóreo; redução da resistência do osso diafisário, ao seis meses de privação hormonal, mas este efeito foi igualado ao longo do tempo pelo envelhecimento do controle; a rigidez óssea foi menor no grupo ooforectomizado em todo o experimento; também foi constatada redução da massa óssea da região metafisária. / In order to investigate the clinical consequences and the histological and biomechanical changes in the bony tissue, causes by menopause induced by oophorectomy, 64 adult and virgin rats, Rattus novergicus albinus (Wistar lineage) were studied. The animals were divided into two groups regarding the treatment: bilateral oophorectomy or placebo surgery. The surgical procedure was carried out when they were four months old. Each group was subdivided into four other groups with eight animals according to the post operation time at the moment they were sacrificed: tree, six, nine and 12 months. The animals’ weight was taken at the moment they were sacrificed. The biomechanical study was carried out on the right tibia by bending test on three spots at the rate level of 30 mm/min, reaching maximum load at the rupture, and the stiffness coefficient assessed from the load-deformation curve. The left tibia was used for the histological study (proximal portion) for the analysis of histomorfometry (assessment of trabecular area). The analysis of the body weight result and the mechanical properties was carried out by two way ANOVA test; the study of the trabecular area was carried out by non parametric ANOVA test. The analysis of the results obtained allowed us to understand that the induced menopause by bilateral oophorectomy increased the body weight; reduced the diaphysis bone strength at the sixth month of lack of hormone but this effect was equaled after a while by the aging control; the bone stiffness was lower in the oophorectomized group throughout the experiment; it was also noticed bone mass reduction in the metaphysis region.
