O Sangue de Dragão (Croton lechleri), por auxiliar na formação de novo colágeno e apresentar propriedades cicatrizante, anti-inflamatória, antimicrobiana, pode ser um meio promissor. Nesse contexto, o objetivo deste trabalho foi avaliar a eficácia da seiva Sangue de Dragão como meio de estocagem para dentes avulsionados por meio da aferição da viabilidade funcional e metabólica celular. Foi testada a diluição a 10% da seiva Sangue de Dragão e como controle, sua diluente solução tampão salina-fosfato (PBS). O leite ultrapasteurizado integral foi utilizado como meio comparativo por ser tradicionalmente utilizado e indicado para esse fim. O controle positivo utilizado foi o DMEM e o controle negativo, a água destilada. Este estudo in vitro comparou a eficácia dos meios para a manutenção da viabilidade de dois tipos celulares: mononucleares de sangue periférico humano e do ligamento periodontal mantidas em cultura. A avaliação da viabilidade foi feita por meio dos testes de exclusão por Azul de Tripan e colorimétrica a base de tetrazolato (MTT), sendo determinada nos tempos 1, 3, 6, 10 e 24h de incubação. Os ensaios foram repetidos 4 vezes, em triplicata para o MTT. A seiva Sangue de Dragão apresentou bons resultados. Para a metodologia com o Azul de Tripan, a seiva Sangue de Dragão foi semelhante ao leite (p>0,0001), ambos com os melhores valores de viabilidade, e para o MTT apresentou resultados superiores a todos os meios, inclusive o leite (p<0,0001). Avulsed tooth storage medium is one of the factors that interferes in the vitality of the periodontal ligament. The Dragon`s Blood sap helps in the formation of new collagen and has been shown healing properties, as well anti-inflammatory, antiviral, antimicrobial and antitumor properties, in which aroused interest in the use as storage medium for avulsed teeth. Thus, the aim of this study is to evaluate the effectiveness of Dragon`s Blood sap as storage medium for avulsed teeth by measuring the functional and metabolic cell viability. It was tested 10% dilution of the sap, and as a control, the diluent phosphate-buffered saline (PBS). Milk was used as comparative because of that is the storage medium traditionally and indicated. The positive control used was DMEM cell culture medium and the water as negative control. This in vitro study compared the effectiveness of the means for maintaining the viability of both cell types: human peripheral blood mononuclear cells and periodontal ligament cells maintained in culture. The cell viability was analyzed by the trypan blue staining exclusion method and the tetrazolium-based colorimetric assay (MTT) and determined at 1, 3, 6, 10 and 24h of incubation. The tests were repeated 4 times, in triplicate for MTT. Dragon`s Blood sap provided positive results. For the trypan blue staining exclusion method, the Dragon`s Blood was similar to milk (p> 0.0001) over time, both with the best values of viability, and for the MTT assay it`s presented better results than all means, including milk (p <0.0001).
