Previous issue date: 1988 / Resumo: A concentração eletrolítica crítica (CEC) da cromatina corresponde à concentração salina na qual se constata total abolição do fenômeno de metacromasia, quando se usam azul de toluidina como corante catlônico e íons Mg2+ como microcátions. Neste trabalho a CEC foi "determinada em hetero e eucromatina de hepatócitos de camundongo e de células epiteliais de túbulos de Malp1gh1 de Triatoma infestans submetidos a jejum. O objetivo foi determinar variações de CEC que pudessem ser correlacionadas a alterações estruturais e/ou de composição desses tipos cromatínicos, induzidas pelo "stress" do jejum. Constataram-se alterações de CEC na hetero- e na eucromatina de hepatócitos de camundongos e na heterocromatina de células de Triatoma infestans, nas condições de jejum. O padrão de resposta ao jejum, no entanto, variou conforme o materiaI, sugerindo diferenças estruturais entre os tipos cromatínicos considerados. O sensível decréscimo no valor da CEC na hetero cromatina de Triatoma infestans após o jejum certamente se relaciona à sua nítida descompactação nesta situação fisiológica. Já nos hepatócitos; com o jejum, há de início uma compactação de hetero- e eucromatina, evidenciada através do aumento dos valores de CEC e mesmo visualmente, e só então, após tempo mais elevado, decréscimo no valor de CEC e também em compactação cromatínica. A realimentação propiciou a que os valores de CEC se aproximassem aos do controle, sugerindo retomada da compactação cromatínica normal e possivelmente de suas funções / Abstract: The critical electrolyte concentration (CEC) of chromatins corresponds to the salt concentration at which the phenomenon of metachromasy is completely abolished when toluidine blue is used as the cationic dye and Mg2+ ions as microcations. In this work the CEC values were determined in the hetero- and euchromatin of mouse hepatocytes, and in the epithelial cells of the Malpighian tubules of Triantoma infestans all after being submitted to atartion. The objective was to determine variations in CEC which could be correlated to structural alterations and/or to the composition of these types of chromatins induced the by stress of starvation. Under starvation conditions, alterations of the CEC were shown the hetero- and euchromatin of the hepatocytes and in the heterochromatin of the Triatoma infestans cells. The pattern response to starvition, however, varied according to the material, suggesting structural differences between the types of chromatin considered. The noticeable decrease in the CEC value of the heterochromatin Triatoma infestans after starvation is clearly defined unraveling wich occurs under this physiological situation. In the case of starved hepatocytes, there is initially a condensation of hetero- and euchromatin as measured both by an increase in the CEC values and visually, which is followed after a certain time by a decrease in the CEC value and also unpackage of the chromatin. With the resumption of feeding the CEC values approximate those of the control, suggesting a return to normal packing state of the chromatin and possibly a normalization of its functions / Mestrado / Biologia Celular / Mestre em Ciências Biológicas
