Previous issue date: 2007 / A tecnologia microarray revelou ser metodologia eficiente na avaliacao da expressao diferencial de genes e, portanto, tornou-se ferramenta usual em investigacao molecular do cancer. Neste estudo, utilizou-se microarrays de cDNA contendo 2000 genes diferentes para analisar a expressao genica em 10 casos de carcinomas endometrioides humanos pos-¬menopausa pareados contra seus tecidos normais correspondentes adjacentes para identificar genes diferencialmente expressos. Varios genes foram encontrados diferencialmente expressos neste estudo. Destacou¬-se o MAP3K8, gene que nunca havia sido descrito superexpresso neste tipo de neoplasia maligna. Para validar a expressao diferencial deste gene, assim como a membrana array, executou-se analise RT-PCR semiquantitativa. O MAP3K8 mostrou-se superexpresso em 30 por cento das amostras de carcinoma de endometrio. Este e o primeiro relato mostrando o oncogene MAP3K8 relacionado ao carcinoma de endometrio humano, sugerindo ser outra molecula envolvida no cancer endometrial humano / Gene microarray technology is highly effective in screening for differential gene expression and has hence become a popular tool in the molecular investigation of cancer. In the present study, cDNA microarrays containing 2000 different genes were used to analyze gene expression profiles in 10 human postmenopausal endometrioid-paired carcinomas specimens versus corresponding adjacent normal tissue to identify differentially expressed genes. In our study several genes were found differentially expressed. One of them was the MAP3K8, a gene that was never described to be overexpressed in this kind of malignancy. To validate the differential expression of this gene as well as the membrane array, we performed semiquantitative reverse transcription-PCR analysis. The MAP3K8 was found overexpressed in 30% of endometrial carcinomas samples. To the best of our knowledge this is the first report showing the MAP3K8 oncogene linked to human endometrial carcinoma suggesting that it may be another molecule involved in human endometrial cancer. / BV UNIFESP: Teses e dissertações
