O Ambergris é um dos perfumes de origem animal mais valorizados no mercado. Esta substância é um produto metabólico encontrado no trato intestinal do cachalote azul (Physeter macrocephalus L.). O substituto sintético comercialmente mais importante para o ambergris é o óxido de nor-labdano (-)-Ambrox® (2). Este trabalho relata a síntese do composto 145, um intermediário avançado na síntese do (-)-ambrox, a partir da cicloalcanona 147 (preparada a partir do furfural e ácido malônico). A anelação de Robinson estereosseletiva de 147, seguida de alquilação redutiva das octalonas com metal em amônia líquida e posterior redução da carbonila via Wolff-Kishner, leva ao composto 145, que possui um núcleo 4,4,10-trimetil-trans-decalínico semelhante ao da molécula alvo. O intermediário avançado 145 foi sintetizado em 4 etapas com 23% de rendimento, a partir da cicloalcanona 147. / Ambergris is one of the most valuable perfumes of animal origino This substance is a metabolic product that accumulates in the gut of the spermwhale (Physeter macrocephalus L.). The most important equivalent of this scarce natural source is the norlabdane oxide Ambrox®. In this work, the synthesis of compound 145, an important synthetic intermediate for the syntheses of the olfactive (-)-ambrox, from cyclohexanone 147 is reported. Stereoselective Robinson annelation of 147, folowed by reductive alkylation with metal in liquid ammonia, and Wolff-Kishner reduction, gave compound 145, wich has a 4,4,10- trimethyl-trans-decalin nucleus. Compound 145 has been synthesised in 4 steps (23 O/o yield) from cyclohexanone 147.
