The studies of the Penaeus monodon baculovirus ( MBV ) DNA / 草蝦桿狀病毒DNA聚合酵素及草蝦組織細胞培養的研究

碩士 / 東吳大學 / 微生物學系 / 82 / Penaeus monodon-type baculovirus( MBV ) was isolated and
purified from the postlarvae of MBV infected Penaeus monodon
Fabricius.MBV DNA was extracted by CTAB/NaCl method and used as
a template in a polymerase chain reaction( PCR ).The primers
were chosen from conserved regions of the DNA polymerase gene
of Autographa californica nuclear polyhedrosis virus( AcMNPV )
and Lymantria dispar nuclear polyhedrosis virus( LdNPV ).Two
DNA fragments( 800bp and 700bp ) were amplified after PCR.
Using the conserved probe to identify these two fragments both
are part MBV DNA polymerase gene. The second part of this
report describes attempts to establ- ish a culture system for
cells derived from the lymphoid organ ( Oka organ ) of the
grass prawn Penaeus monodon.The best gro- wth of Oka organ
cells occurred in a culture system containing Leibovitz's L-15
medium supplemented with 5g/l NaCl, 1g/l glucose and 10% FBS at
28℃.The effect of different growth factors on Oka organ cell
culture was observed.Insulin( 10 ug /ml ) and Insulin-like
growth factor I ( 25 ng/ml ) make cells have round morphology
and growing clusters. And then yeast contamination and die.They
were subcultured for 5 and 4 gener- ations. Treat the Oka organ
cells by adding 20 ng/ml basic fibroblast growth factor make
cells transformed. They grow as anchorage independent behavior
and can growing in reducing serum concentration ( 5% FBS ) and
does not need growth factor any more. This cell ( F-20 ) grow
for 28 generations now and still growing. It's a new cells that
never found before.
