碩士 / 國立臺灣大學 / 藥理學研究所 / 83 / The effects of 2-phenyl-4,4-dipyridylmethyl-1,2,3,4-tetrahydro-isoquinoline-1,3-dione hydrochloride;HBT46 and DPMPI isoquinoline derivative, on the spontaneous mechanical responses of circular smooth muscle of the guinea pig stomach were studied.HBT46(5muM) and DPMPI(1muM) dose dependly increased the amplitude of spontaneous mechanical contractions without altering their frequency. The effect was not altered in the presence of cimetidine,atropine. HBT46 also increased the amplitude of spontaneous mechanical contractions in low sodium,potassium as well as calcium. however, the effect was decreased in the presence of diphenhydramine,HBT46 did not alter the acetylcholine elicited responses of the circular muscle of the stomach,vas deferences and norepinephrine elicited response of the pulmonary artery.the results suggest that HBT46 and DPMPI increase the amplitude of the spontaneous mechanical activity of the circular muscle of the stomach. the effect may be associated with the histamine receptor in the stomach but not the adrenergic, cholinergic receptor of the stomach.HBT46 and DPMPI on the central nerve system were studied using in vitro rat hippocampal brain slices. DPMPI(10-63muM) reversibly increases the amplitude of population spikes by a concentration dependt manner.DPMPI (63muM) increased the amplitude of population spikes and induction long term potentiation of the hippocampal slices,while it did not affect the maintenance LTP of the slices. The increasing effect of DPMPI on the PS was potentiated in presence of 4-AP.
DPMPI(63muM) decreased the ratio of twin pulses facilitate of the hippocampal slices. the decreasing effect was not altered in the presence of nifedipine (30muM) or streptomycin(10muM).It was concluded that DPMPI increased the PS of brain slice.The effet may be due mainly to its inhibitory effect on the potassium current of the nerve.
