neurotrophins of nervous system, cytokines of hematopoietic system, hormones of endocrine system, and morphogens of embryos,play important roles for regulation of embryonic development,maintenance of internal homeostasis, and responses to various external stimuli. These secretory molecules contained within secretory granules or synaptic vesicles are released to the
extracellular space via constitutive and regulated secretion pathways.
The aim of this study was to design a screening strategy for
isolation of genes encoding secretory proteins. This strategy combines the following techniques: in vitro tissue perfusion, preparative gel electrophoresis, production of polyclonal antisera and immunoscreening of an expression cDNA library. The system we chose was carp head kidney because this organ consists of neuron, endocrine cells, and hematopoietic cells and we expected that protein factors were released from this organ.
A full-length cDNA clone encoding the precursor of carp apolipoprotein A-I was isolated by this screening strategy. Carp apolipoprotein A-I consists of an 18-amino acid prepeptide, a 4-amino acid propeptide and a mature protein of 234 amino acids.
Overall structure of the mature protein is dominated by tandem repeated, 11-amino acid blocks which are predicted to exhibit an amphipathic alpha-helical secondary structure.Carp apolipoprotein A-I protein is expressed in a variety of tissues including brain, head kidney, kidney, liver, spleen, ovary, testis, and muscle.
The distribution pattern is different from that of mammalian apolipoprotein A-I. Moreover, consensus sequence responsible for the interaction with mammalina apolipoprotein E receptor are observed in carp apolipoprotein A-I. The data suggest that carp apolipoprotein A-I may function as both apolipoprotein A-I and E.
