Study of carp ovarian cystatin and cysteine proteinase / 鯉魚卵巢硫胱胺酸蛋白酶及其抑制因子之研究

博士 / 國立臺灣大學 / 動物學研究所 / 83 / The ovarian fluid of carp has several components not reacted with the antiserum against carp serum. Among them, a protein of 12 kDa (p12) was purified to homogeneity. p12 can inhibit the enzymatic activity of papain. The expression of p12 is confined in ovary. The p12 thus purified is a nicked protein. The primary structure of p12 was determined by Edman degradation and molecular cloning. A cDNA of 531 bp encoding p12 was cloned.
It has an open reading frame of 387 bp. The precursor of p12 Degenerated oligonucleotide primers based on the conserved
domain of cysteine proteinase were used for polymerase chain reaction. A 490 base pair PCR product was obtained from carp
ovary. With the above PCR product as a probe, a 1228 bp
full length cDNA was cloned. The protein encoded by the
above cDNA is homologous to cysteine proteinase. Western blot
analysis by using the antiserum against recombinent carp
cysteine proteinase showed that head kidney, kidney, spleen
and muscle have a higher content of cysteine protease than brain, liver, pituitary and testis. The level of cysteine protease in ovary raises at the interval of 4 to 7
hours after injection of the pituitary homogenate.
