Studies on the Trypsin in the Hepatopancreas of Metapenzeopsis barbata / 鬚赤對蝦胰蛋白�@之研究

碩士 / 國立海洋大學 / 水產養殖學系 / 85 / Hepatopancreas is the main digestive nutrition absorbent and
storage organ of Metapenzeopsis barbata. In order to elucidate
the relationship between the digestive enzyme-trypsin in
hepatopaancreas and nutritional requirements of Metapenzeopssis
barbata. Histological method,and histochemical method and
methods of enzymology were used in this experiment to elucidate
trypsin in hepatopancreas and nutritional requirements of
Metapenzeopsis barbata. From the result of histology,
hepatopancreas ofMetapenzeopsis barbata is composed of method,
there is higher concentration trypsin near the digestive tract
in the hepatopancreas. From the result of enzymology method,
the optimum pH and temperature of trypsin of Metapenzeopsis
barbata for hydrolysis of TAME was found to be pH8.0 and 50℃.
The activity of this enzyme is more stable in neutral and slight
alkaline pH than that in acidic pH,and this enzyme have been
found to be unstable aabove 50℃. The molecular weight of this
enzyme was determined by SDS-polyacrylamide gel electrophoresis
to be 26,600. The activity of this enzyme was inhibited by Hg,
Cu,TLCK and PMSF. However,the activity of this enzyme was not
inhibited by EDTA and TLCK. The kinetic study of this enzyme
was carried out on the hydrolytic reaction of the synthetic
substrate TAME at pH 8.1 and 25℃. Michaelis constant Km of
this enzyme was 2.109 millimolar.
