Studies on the development of Astaxanthin from Phaffia rhodozyma / 紅酵母還原蝦紅素之開發研究

Phaffia rhodozyma contains a large quantity of carotenoids, and
Astaxanthin amounts to 65~95% of the total carotenoids. The
quantity of Astaxanthin has a great deal to do with the kinds of
medium, the concentration of medium, and the growth stage.
In high density cultivation, we add Astaxanthin to the food, the
cultured fish or shrimps can have the same color as wild ones.
Thus their prices will be raised. Astaxanthin sells at more than
U.S.$ 2,000. per kilogram. This makes us eagerly desire to find
the red yeast with high accumulation of Astaxanthin.
In this experiment, we use pH, temperature, carbon source, and
nitrogen source to find the best growth conditions for red yeast
to accumulate Astaxanthin.
The result show as below; Under pH5 and pH6, red yeast gets
higher accumulation of total carotenoids. Under 24℃or lower
than 24℃, it has a stable accumulation. But under the
temperature higher than 24℃, it will get lower total
carotenoids. When we culture red yeast under 10℃or lower
temperature, we can get three to four times as much total
carotenoids accumulation as we can under about 20℃.
In cooling down temperature treatment, if the temperature cools
down from 26℃or 24℃to 4℃, these two groups have lower total
carotenoids than they do if the temperature remains 26℃or 24℃.
But the temperature cools down from 22℃or 20℃to 4℃, these two
groups have higher total carotenoids than they do if the
temperature remains 22℃or 20℃.
Glucose, fructose, galactose, moltose, sucrose and apple juice
have better total carotenoids accumulation in red yeast for
carbon source test. But cultivation with single carbon source
has lower red yeast number and pigment accumulation than potato
extract mixed with glucose.
Urea has better use as a nitrogen source for red yeast to
accumulate total carotenoids. This is because red yeast is some
kind of Basidiomycelous origin. But on the whole, red
yeast makes low use of nitrogen source.
