Studies on Saprolegniaceous Fungi in Taiwan / 台灣水生菌科之研究

碩士 / 國立臺灣大學 / 植物病蟲害學系 / 85 / More than 400 samples of soilsand water were collected from
various locations of Taiwan. Baiting method was used to isolate
the Saprolegniaceous fungi. Totally 28 species included in 8
genera of Saprolegniaceous fungi were identified . Among them, 3
species , ie. Achlya diffusa, A. inflata and Saprolegnia
australis, are new records for Taiwan. The author screened 40
antibiotics and fungicides to make selective medium for
isolation of Saprolegniaeous fungi. After screening work, the
medium made of , adding 1 ppm Metalaxyl 、250 ppm PCNB、500 ppm
Neomycin sulfate and 1000 ppm Amoxicillin to YpSs+hempseed
extract medium shows the best selective effect of isolation of
Saprolegniaeous fungi . In the survey on survival structures
in the soil, , oospores played the most important role in the
survival of Saprolegniaeous fungi. In addition to its
survivability, oospore was more resistant to the low temperature
(-20℃)and chemicals (KMnO4)than cyst 、 hyphae and gemmae.
In the germination experiment of Achlya prolifera, the oospores
formed in gnobiotic culture gived higher germination ratio than
that formed in axenic culture.Nutrients and high temperature
could promote the germination of oospores. KMnO4 solutions and
light showed no effect on promoting the germination of oospores.
In addition to the resistant to the stress , the single spore
isolates from oospores have more diversity in the growth raets
than those single spore isolates from zoospores.
