Pigment Analysis on Peridinium bipes in Techi Reservoir / 德基水庫多甲藻色素分析研究

碩士 / 國立中興大學 / 植物學系 / 86 / A dinoflagellate, Peridinium bipes Stain fa. occulatum usually
forms bloom in Techi Reservoir in summer. It is interesting to
know the photosynthetic pigmentof this species. After
calculation the ratio of nitrogen to carbon of P.bipes cellswas
about 10 fold lower than that of reported dinoflagellates. The
ratio of chlorophyll a to carbon was 10 fold higher than other
dinoflagellates. These date might indicate that the
photosynthetic products of P.bipes was transferred to the need
of cell growthTwelve pigments of P.bipes were separated from
thin-layer chromatography. Each pigment was identified by
visiblelight spectrometry and mass spectrometry(inclucding EI MS
and FAB MS).It was found that P.bipes contained 8 carotenoids,
including diadinoxanthin,mutatoxanthin,diatoxanthin,B-carotene,
prasinoxanthin,chrysanthemaxanthin,neochrome and peridinin,in a
addition to two major chlorophylls,chlorophull a and chlorophyll
c2.Quantitative analysis of the carotenoids profile in P.bipes
was also studies by a reversed-phase HPLC.Four major carotenoids
were found to be peridinin(30.0-44.0%),
mutatoxanthin(21.7-30.8%),B-carotene(6.8-15.3%) and
diatoxanthin(5.8-17.9%). However accordingly the content of
total amounts of these four carotenoids were found to exceed 90%
in carotenoids of P.bipes.Peridinin and diadinoxanthin in P.
bipes were found to be relatively small in comparison with those
of other reported dinoflagellates.
