碩士 / 國立中山大學 / 海洋資源研究所 / 86 / The X organ -sinus gland complex of eyestalk in crustacean secrete many neuropeptides which regulate many physiological funtion such as reproduction and growth. Among those secreted in this complex, the CHH(crustacean hyperglycemic hormone), MIH(moult inhibiting hormone), GIH(gonad inhibiting hormone) and MOIH(mandibular organ inhibiting hormone) are high homologous in sequences and overlap in biological function, so they were classified into the same group and named as CHH family. the CHH family share two characters : (1) molecules are composamed of 72 - 78 amino acids, (2) high homology in amino acids sequence, with 6 high conservatory cysteine. This report study the CHH precursors of tiger prawn, ~u2;Penaeus~u1;~u2;monodon~u1;. Poly(A) RNA was extracted from the X organ -sinus gland complex, and RT-PCR was performed using reverse transcriptase and conservatory primer which is designed according to the conserve region of CHH from other crustacean. The cDNA products were subcloned to plasmid and then sequenced. Specific primers were determined according to the nucleic acidssequences, then the sequences of either end were obtained using 5' and 3'RACE (rapid amplification of cDNA ends) methods. Two cDNAs encoding tiger prawnCHHs were found and named as ~u2;Pem~u1;-CHH1 and ~u2;Pem~u1;-CHH2, respectively. The cDNA of ~u2;Pem~u1;-CHH1 was cloned into ~u2;E.~u1; ~u2;coli~u1;, the expressed protein was purfied and assayed, the VHH activity was indicated.The anti-~u2;Pem~u1;-CHH1 antibody was induced from rabbit. The antibodycan be used in the verification the producing sute of CHH in X-organ,the examination of the titer of CHH concentration in hemolymph, and the analysis in the CHH analogue in tiger pramn.
