Studies on Interleukin-2-like Molecules in Grass Prawn (Penaeus monodon) Using Boiassay and Immunochemical Techniques / 草蝦類淋巴激素─2免疫化學與生物活性之研究

碩士 / 國立海洋大學 / 水產生物技術研究所 / 86 / The existence of interleukin-2-like molecules was studied in the Oka (lymphoid) organ of grass prawn (Penaeus monodon) and its derived cell line (PMO) by enzyme-linked immunosorbent assay (ELISA) , bioassay and Western blot. PMO cells were found to be capable of releasing IL-2-like molecules at 4 hrs after plating by competitive ELISA assay. PMO cells treated with Con A or LPS for 12 hr gave 50% increase in amount of the IL-2-like molecules in culture supernatant. IL-2-like molecules were also found in the presence Oka organ culture. Furthermore, the IL-2-activity of prawn IL-2-like molecule was demonstrated by HT-2 cell (IL-2 responsive cell line) proliferating assay. Western blot analysis of the PMO cell lysate with goat polyclonal antibodies against murine recombinant IL-2 showed peptides estimated to be 55, 40 kDa and 17kDa in molecular mass, while p55 and p40 were found in Oka homogenate. This is the first evidence for the IL-2-like molecule existing in prawn Oka organ.
