Effects of developmental stages, medium, light and containers on concentrations of CO2 and ethylene in Anthurium andraenum, Zantedeschia hybrida and some ornamental plants in vitro / 發育階段、培養基、光線及容器對火鶴花、彩色海芋與數種觀賞植物組織培養苗瓶內CO2和乙烯濃度之影響

碩士 / 國立臺灣大學 / 園藝學系研究所 / 86 / SummaryEffects of explants and environmental factors on concentration of carb
on dioxide and ethylene and on growth of explants in vitro were studied in Ant
hurium andraeanum, Zantedeschia hybrida, Limonium sinuatum ( L. ) Mill., Phala
enopsis spp., Cattleya spp., Oncidium spp., Anoectochilus formosanus Hayata, P
hilodendron×mandaianum ''Red Emerald'', Philodendron scandens subsp. Scandens,
Syngonium podophyllum ''White Butterfly'', Caladium., Dischidia pectinoides, Spa
thiphyllum ''Sensation'' and Anubias barderi at various developmental stages. Th
e CO2 concentration of proliferation stage in Anth. andraeanum and Z. hybrida
grown in GBC containers was higher than that of rooting stage grown in PVC con
tainers. At proliferation stage, CO2 concentration was measured to be 2.5~5 %
and ethylene concentration up to 1.25 ppm in Limonium sinuatum ( L.) Mill. The
ethylene concentration in vitro remained constant in Anth. andraeanum and Z.
hybrida. The general trend in ethylene concentration in vitro during vultural
stages was to decrease in Phal., Onc., Cattleya., and Anoectochilus formosanus
. This was also measured to decrease at lateral developemental stages for foli
age plants. The CO2 concentration was higher in later growth stages than ealie
r stage in orchid plants. At later developmental stages, it was low below air
concentration (about 350 ppm) in foliage plants. The concentration of CO2 was
increased as medium volumn increased and as agar added after sterilization. An
addition of activated charcoal result in the increase of CO2 concentration an
d in absorbing ethylene. Cultural medium containing 30 g/L sucrose produced we
ll grown the plantlets of Anth. andraeanum ''Tropical''. The CO2 concentration a
t the end of light period was lower than those at the end of dark period, and
the differences in CO2 concentrations between lighting treatments were increas
ed as sucrose concentration increased. The optimum concentration of sucrose fo
r the growth of the plantlets was 20 g/L in Z. hybrida ''Black Magic'' and ''Best
Gold''. The CO2 concentration at end of light period was higher than the end o
f dark period, and the differences of CO2 concentration between lighting treat
mnet were largest in 20 g/L sucrose treatment. Sealing with foam serum, whose
t50 value was 1.30 h, resulted in good growth in Anth. andraeanum ''Tropical''.
There were no difference in growth between various container treatments in Ant
h. andraeanum. Abnormal growth was observed in Z. hybrida ''Black Magic'' grown
in medium with ACC, which resulted in CO2 concentration increased in vitro. As
AgNO3 concentration increased, the height of plantlets of Z. hybrida ''Black M
agic'' was decreased.
